Characterization of proteins interacting with proteolytic system of ticks
Matoušková, Zuzana ; Mareš, Michael (advisor) ; Hudeček, Jiří (referee) ; Perner, Jan (referee)
(EN) Ticks are ectoparasites found worldwide that feed on the blood of their hosts and transmit several important pathogens to humans and domestic animals. Tick saliva contains bioactive molecules that are injected into the host tissue to aid in successful blood feeding. Among these molecules, proteinaceous protease inhibitors are being extensively studied for their potential biomedical applications. This work focuses on novel protease inhibitors from the saliva of the ticks Ixodes ricinus, vector of Lyme disease and tick-borne encephalitis, and Ornithodorus moubata, vector of relapsing fever and African swine fever. Research has focused on the biochemical and structural characterization of three members of the protease inhibitor families of cystatins, serpins, and tyropins, and has attempted to elucidate their biological function in tick-host interactions. Cystatin OmC2 from O. moubata was identified as a broad-spectrum inhibitor of host cysteine cathepsins with both endopeptidase and exopeptidase activity. Crystal structure determination allowed description of the relationship between the structure of OmC2 and its inhibitory specificity. The ability of OmC2 to modulate the host immune response was demonstrated in bioassays and a suppressive effect on O. moubata in vaccination experiments. The...
Prefixes and prefixoids of Latin and Greek origin as a prerequisite for understanding foreign words in four-year grammar school students
Králová, Eliška ; Janovec, Ladislav (advisor) ; Šindelářová, Jaromíra (referee) ; Mitter, Patrik (referee)
Classical languages - Latin and Greek - have been influencing the vocabulary of the Czech language throughout its independent development. Both languages contribute to the process of creating new words to a large extent even today. This does not apply only to terminologies of various scientific fields but also to the naming of phenomena and facts of our everyday life. In connection with globalization and the general effort for language internationalization, classical languages, thanks to their flexibility and ability to link with domestic and foreign word-forming components, are very intensively involved in neology. The aim of this dissertation is to demonstrate how high the level of understanding of word-forming components from Latin and Greek is shown by students in the 1st and 4th year of the four-year general grammar school, and to what extent possible unfamiliarity prevents them from a correct understanding of similarly formed expressions. The tool for determining the scope of knowledge among students was a didactic test administered at five four-year grammar schools in the Czech Republic, and every time, for comparison, in all parallels of the 1st and 4th year. The results of the research may help to improve the didactic processing of the curriculum in Czech language classes, and to enhance...

This study provides an empirical overview of teaching aspirations among 15-year-old school pupils in European countries based on data from the international PISA survey.
Analysis of the 2024 state budget revenue in light of the 2019 pre-crisis year
Janský, Petr ; Kolář, Daniel
This analysis compares planned state budget revenues in 2024 with the state budget outcomes of 2019. It charts a period in which the Czech economy and public budgets have been heavily impacted by the consequences of several major crises (coronavirus, the invasion of Ukraine and the war, an energy crisis, and an inflationary wave) as well as by significant parametric changes in the Czech tax system. 2019 can be considered the last pre-crisis year and 2024 the first post-crisis year, so they correspond to 'normal times' more closely than the years in between. This analysis builds on a previous analysis of the state budget expenditure development between 2019 and 2024 and includes a brief synthesis of findings on revenues and expenditures.

Media discourse on Green Deal from energy perspective
Kolářová, Marta
Green Deal, the European Commission's initiative to protect the climate and reduce emissions greenhouse gases, has sparked controversy in the Czech public debate. An analysis of media discourse on the Green Deal from an energy perspective shows that the Green Deal is perceived predominantly negatively. In the media, it has been labelled with condemnatory statements and negative characteristics, especially by leading Czech politicians. A more favourable view of it is held by Czech companies, municipalities and experts. The public debate is being transformed in particular by the impact of the war in Ukraine in favour of energy security and accelerating the green transformation.

The second part concerned the zero-drift characterization of a common-path differential interferometer. It was a measurement of interference phase fluctuations during the reflection of interferometer arm beams on a common mirror (zero-drift) lasting at least 100 hours with a resolution better than 1 nm for a pair of differential interferometers with simultaneous measurement of atmospheric parameters.
Pathways to a just landscape transformation
Nohlová, Barbora ; Sagapová, Nikola ; Harmáčková, Veronika Zuzana
The Just Scapes project was officially launched in 2020 and is a consortium of three countries (UK, France and Czech Republic) researching three types of landscape: the Scottish Highlands, the French rural landscape of the south-eastern Pyrenees and the Czech cultural landscape of southern and eastern Moravia. The project focuses primarily on the mechanisms and processes of equitable transformation and answers the following questions, crucial for the future: How are the impacts of climate change distributed? How are impacts and effects distributed?\nof landscape management and climate change policies? The research was based on the approach of the so-called ,,just transformation laboratories,, (T-labs). This approach is specifically tailored to foster collaborative solutions to complex societal problems. The research also included extensive field visits and engagement with relevant key actors. Numerous interviews and workshops were also conducted. This text is the result of a long-term research collaboration with key actors managing the landscape in five wine and agricultural regions of South and East Moravia. The research focused on mechanisms of equitable transformation. If the current state of the landscape leads to its vulnerability to the impacts of climate change, it is necessary to ask what changes are needed to increase its resilience. And this is exactly what the project has succeeded in defining.

Spectral Indices as a Tool for Hop Growth Evaluation
Seidlová, Jana ; Procházka, Pavel ; Kumhálová, Jitka
The use of unmanned aerial vehicles (UAV) to monitor crop growth is nowadays a common non-invasive way how to obtain information on the current state of crops. Spectral indices derived from multispectral images obtained in the right growth stage can then serve as a good data source for agro-technical interventions and yield estimation. Hop belongs among the crops where it is possible to scan the individual growth parameters very exactly. In the year 2021, significant precipitation amounts were recorded during the growing season, when it turned out that UAVs are a very powerful tool for determining the quality of production or quantification of vegetation damage compared to the previous year (2020). It was found that the common spectral indices were possible to use for calculation leaf area, structure, vigor and chlorophyll content of hop gardens.

Hydrogeological and hydrological data contained in records of contaminated sites in the SEKM database
Tylčer, Jiří ; Suchánek, Zdeněk
As of May 2022, approximately 10,200 contaminated and potentially contaminated sites are registered in the database of the Contaminated Sites Registration System (SEKM) updated by the National Inventory of Contaminated Sites project (NIKM, 2018-2021). Each structured record of such site contains a number of hydrogeological and hydrological characteristics, which serve primarily to assess the possibility of contamination migration. The most important synthetic parameter of the hydrogeological and hydrological conditions at the site is the "Possibility of migration of contamination". The basis for the assessment is the assignment of the site to one of the five groups of types of hydrogeological structures of contaminated sites. For the set of localities of the highest priority category A, we demonstrate statistical data on the occurrence of the monitored "hydro" parameters, broken down by five types of migration options, for each subcategory A1, A2 and A3. The summary map of the territory of the Czech Republic shows the location of 451 subject contaminated sites of priority category A.

Effectiveness of the use of clues of contaminated sites obtained by remote sensing for the National Inventory of Contaminated Sites
Suchánek, Zdeněk ; Řeřicha, Jaroslav
The National Inventory of Contaminated Sites (NIKM) project was aimed to fill the database of the Contaminated Sites Register (SEKM) with approximately 9053 updated and validated records with evaluated priority, taking into account the risk from the point of view of life and health of the population and the environment. We expect the number of records to be approximately 10100. Thousands of contaminated and potentially contaminated sites (KM / PKM) entered in the SEKM register are also waste management sites. The article focuses on the possibility of using information from the inventory for waste management. The initial project task implemented in the years 2018–2019 concerned the mapping of KM / PKM clues by remote sensing (RS) methods. The mapping provided a wealth of insights into the territorial distribution of clues. Using remote sensing methods, a significant part (approx. 18%) was obtained from the total number of approx. 10100 evaluated sites registered in SEKM. 48.5% of all records of evaluated localities arising from clues come from the group of types of clues determined using remote sensing methods and related to municipal waste (types of clues c, p, l, r). The use of remote sensing methods, such as the interpretation of orthophotomaps, can be a significant benefit for the performance of public administration in the field of spatial planning and environmental protection. Today, SEKM is a complete public database of the Ministry of the Environment that can be used by the professional and general public. Many of information contained in it can be used for the issue of waste disposal on the surface.

Old municipal waste landfills inventoried in the Contaminated Sites Registration System
Suchánek, Zdeněk ; Řeřicha, Jaroslav
The database of the Contaminated Sites Registration System, filled in at the end of 2021 as part of the National Inventory of Contaminated Sites for the number of 10,134 records of assessed contaminated and potentially contaminated sites, contains, amongst other data, 4,619 sites registered as a “Solid Municipal Waste Landfill” type. This part of the database, together with additional information sources, was analysed in terms of parameters such as landfilling duration, landfill closure date, landfill size (area), location (territorial representativeness), state of knowledge about the landfill, and its risk and time urgency for possible corrective measures. We interpreted the obtained historical and current data on old municipal waste landfills and used them for the preparation of criteria for multi-criteria selection of the main types of old landfills and the selection of specific sites for sampling and technical works, to be done within the subsequent research project. The selection of landfills for sampling will be representative in the case of old municipal waste landfills, which are located in the South Bohemian, South Moravian, Central Bohemian and Vysočina regions. They should have an area of more than 100 m2, be of categories P and A, not having any set corrective measures, with an absence of the financing of remedial measures, be landfilled between 5 to 55 years and their landfilling activities having been terminated in the 1990s.

Methods of landfill gas treatment in landfills in integrated prevention system
Keprtová, Karolína ; Kolář, Jan
The article focuses on the issue of landfill gas man-agement in landfills, which are within the scope of Act No. 76/2002 Coll., on integrated prevention. Al-though landfilling as a way of disposing of municipal waste is in decline, the subsequent care of landfills will continue for another decade after their closure, and landfill gas must be captured even then. The article summarizes infor-mation on the method of landfill gas management ob-tained through the information system of integrated pre-vention as well as the rules for landfill gas management according to the valid Czech technical standards. Accord-ing to data obtained from the public database of the inte-grated prevention system, 136 relevant landfills were identified. The application of a cogeneration has the larg-est share in the way of landfill gas management. If a co-generation unit is installed at the landfill, it is in most cases operated by a company other than the landfill oper-ator. It was also found that the region with the lowest population density has the largest number of landfills with low capacity. Due to the fact that the production of landfill gas decreases over time, it is possible that there will be changes in the way landfill gas is handled at indi-vidual landfills.

BAT Use Treating and Recycling Construction and Demolition Waste
Keprtová, Karolína ; Kolář, Jan
Prevention of waste production is a priority according to a Czech legislation. In cases such as demolition, when the generation of waste cannot be prevented, is desirable to use the material. The material recovery of demolition waste requires technical and technological equipment at the appropriate level, which is very expensive. To obtain the necessary equipment it is possible to use, for example, the grant calls that support new technologies, applying the principles of circular economy and environmental friendliness. For the assessment of environmental friendliness it is possible to compare a particular technique with the best available technique (BAT, Best Available Techniques). Technology, which comply with BAT aspects, ensure compliance with both legislative and performance requirements for a number of years to come while guaranteeing sufficient capacity to recover waste to create valuable and recyclable products.

A New Hollow Fiber-Based Liquid-Phase Microextraction Method for the Determination of Antihypertensive Drug Lercanidipine in Biological Samples
Labzova, O. ; Hrdlička, Vojtěch ; Navrátil, Tomáš ; Šelešovská, R.
A new hollow fiber-based liquid-phase microextraction (HF-LPME) method for the determination of antihypertensive drug lercandipine (LCN) in biological samples was developed. HF-LPME was\ncombined with optimized square wave voltammetry (SWV) on a cathodically pre-treated screenprinted boron-doped diamond electrode (SP-BDDE). Optimum HF-LPME conditions were:\nsupported liquid membrane (SLM) dodecane, 0.02 mol L-1 Britton-Robinson buffer (BRB, pH = 3) acceptor phase, BRB (pH = 7) donor phase, and time of extraction 30 min. Limits of quantification (LOQ) and detection (LOD) were 3.3 and 1.1 nmol L-1, respectively. The applicability of the developed method was verified on human urine, blood serum, and blood plasma with 20 and 100 nmol L-1 LCN addition.
A New Approach to Determining the Drug Guaifenesin Using Voltammetric and Flow Injection Analysis
Kelíšková, P. ; Matvieiev, O. ; Sokolová, Romana ; Janíková, L. ; Behúl, M. ; Šelešovská, R.
This study presents a new method for detecting guaifenesin (GFE) in pharmaceutical samples. We employed screen-printed sensors with a boron-doped diamond electrode (BDDE) to develop and validate both voltammetric and flow amperometric methods. These approaches were effectively used to analyze model solutions, pharmaceutical preparations, and serum samples spiked with GFE, producing accurate results comparable to those obtained with HPLC/DAD analysis. The developed methods provide straightforward, sensitive, and selective means for determining GFE in various sample matrices, underscoring the potential of SP/BDDE in electrochemical analysis.

Instruments for precipitation measurement from history to the present
Lipina, P. ; Procházka, J. ; Tesař, Miroslav
The precipitation amount is one of the most important meteorological elements. It is of fundamental importance for the creation of climatological characteristics, it is the basis of the water balance and the primarily monitored element in the forecasting and warning service. Precipitation measurement has been burdened with many uncertainties for a long time and is therefore consistently given due attention.\nThe first measurement of precipitation in Europe dates back to the 17th century. Regular but not complete measurements of precipitation at Prague's Klementine observatory began in 1752 (continuous series since 1804), in Brno in 1803. In 1817, M.A. David wrote the first guide for observing the weather in Bohemia, published in German, intended for volunteer observers. The activities of institutions and various associations, such as the Prague Klementinum, the Natural Research Society in Brno, the Central Institute for Meteorology and Earth Magnetism and others, had a fundamental influence on the establishment of meteorological stations and precipitation measurements. The Hydrographic Commission for the Kingdom of the Czech Republic and its hydrometric section were founded in 1875 under the leadership of F. J. Studnička E. Purkyně was responsible for the significant expansion of precipitation measurement at that time as part of the construction of a network of forest precipitation measuring stations. \nFor more than 200 years, rain gauges and precipitation measurements have been developed. The Metra 886 manual rain gauge, which has been used for a long time in the Czech lands, was developed according to the Austrian model of prof. Kostlivý. Since 1995, the automation of rainfall measurement was started, first with tipping bucket raingauges, which are gradually being replaced by weighing raingauges. Alternative methods of measuring precipitation are being tested in\nremote and forested areas. This post tries to bring the history, standards and development of precipitation measurement in our country at least as a guide.
Temperature regime of forest soils
Dohnal, M. ; Votrubová, J. ; Mazúchová, R. ; Blunár, P. ; Tesař, Miroslav
Contribution deals with the monitoring of the temperature regime of forest soils and the soil heat flux at small mountainous basins Liz in the Šumava Mts. and Uhlířská in the Jizera Mts. Soil temperatures are measured with the help of soil moisture meters (Pt100 at Liz basin and CS107 at Uhlířská basin), the soil heat flux is measured with the use of the sensor Hukseflux HFP01. Measured results were statistically evaluated, the analysis of the consistence of the measurement was done and cumulative soil heat fluxes were calculated for the individual hydrological years.
Palingenesis - the restoration of everything? (Matthew 19,28)
Dus, Jan A
In Matthew’s version of Jesus’ promise (Matthew 19:28–30), in contrast to Mark and Luke, there is no explicit mention of the present time, the restitution of all that the followers of Jesus left behind is thus pushed forward to the future, in which they will take over the reigns with the Son of Man. The promise features the rare term “palingenesis”, whose precise temporal and factual determination cannot be deduced from the sentence itself. While in Mt 19:28 palingenesis is, according to the prevailing interpretation, a synonym for the future restoration of the universe, in Tit 3:5 (the only other occurrence in the NT) the same word refers to the rebirth of some people that took place in the past (at baptism). Even non-biblical literature does not provide a clear precedent - palingenesis tends to be set both in the past and in the future, it concerns both the world and individuals or groups (Israel). – The syntax and punctuation of the Greek sentence show that both interpretations are equally possible: palingenesis can be related either (A) to the following verbal form “sitting (the Son of Man on the throne)”, i.e. to the future, or (B) to the preceding verbal form “following (disciples behind Jesus)”, i.e. into the past, in addition, there is a third, open option (C). In the 16th-century Greek, Latin, and Czech versions, all possibilities are represented: Erasmus punctuates the Vulgate differently (B), Beza first respects the Vulgate (A), but in his Annotations he considers the second solution (B), in the fourth and fifth editions he finally leaves the matter open (C), two Czech translations from the first half of the 16th century (Náměšť 1533, Melantrich 1556/57) hesitate in the dilemma between the Vulgate and Erasmus (C), most Brethren translations starting with Blahoslav (1564) deviate from the Vulgate (B), only the Six-Volume Kralice Bible from 1593/94 (“Kralická Šestidílka”) hesitates (C), in “Kralická Šestidílka” and in the Kralice New Testament from 1601 the Brethren pay equal attention to both interpretations in a rather extensive note, which, for its balance, is a nice proof of intellectual openness and tolerance. Like Beza, the Brethren add a new aspect to interpretation (B): palingenesis can mean the renewal of the whole world thanks to the Gospel and the first coming of the Messiah. – Considering the dynamic development in the 16th century, it is surprising that all ten tracked Czech translations from around the 20th century agree on only one variant (A). Option (B) seems to be in danger of unfair oblivion. Therefore, in conclusion, we allow ourselves to propose a new, somewhat looser translation or paraphrase of verse Mt 19:28 in the spirit of Kralice: “Amen, I say to you, it was you who followed me when I came to renew the world with the gospel, and when the Son of Man sits on the throne of his glory, it will be you again who will sit on twelve thrones and judge the twelve tribes of Israel.”
Inverse-Heusler Mn2FeSi alloy prepared by powder metallurgy route
Životský, O. ; Skotnicová, K. ; Čegan, T. ; Juřica, J. ; Gembalová, L. ; Zažímal, František ; Jirásková, Yvonna ; Szurman, I.
The Mn2FeSi inverse-Heusler alloy was prepared by solid-state reactions using ball milling for 168 h in n hexane. Two milling procedures varying in the ball-to-powder ratio, namely 4:1 and 10:1, were used to follow \ntheir influence on alloy formation and its physical properties. In both cases no Heusler structure was obtained \ndirectly and therefore the subsequent annealing at 1223 K for 1.5 h in pure Ar was applied. The energy dispersive X-ray analysis resulted in the chemical composition about 49.0-49.9 at% Mn, 25.3-25.6 at% Fe, and \n24.8-25.4 at% Si, in a good agreement with nominal one. X-ray diffraction of both powders confirmed inverse Heusler XA structure of lattice parameters 0.5675 nm (4:1) and 0.5677 nm (10:1), only slightly higher as \ntheoretically obtained 0.560 nm. The presence of minor oxidation phases was also observed in the particle \nstructure of powders. Magnetic properties were analysed at low and room temperatures. Both alloys exhibit \nparamagnetic behaviour at room temperature confirmed by transmission Mössbauer spectroscopy \nmeasurements. Different magnetic behaviour of alloys is observed below 65 K.
Kosher in lexicography. Problems of one religion term
Dvořáková, Žaneta
The Hebrew term košer (‘kosher’) came into Czech through Jewish sociolect / ethnolect. It was originally a religious term for what is ritually pure and appropriate. In addition to the adjective and adverb košer, Czech dictionaries also contain a number of other forms and derivatives, but some of them are no longer used at the moment (e.g. košerný, košerně). Our evaluation is based on an analysis of material base from the Syn2015 Corpus and from a probe into contemporary texts in Jewish magazines or on the websites of Jewish communities and organizations. In Czech dictionaries, we may see a reduction of the meaning compared to how the term is traditionally used in the Jewish community, e.g. the adjective košer is limited to meat, the verb košerovat is associated only with the ritual slaughter. The nouns mentioned in the dictionaries are also very debatable: masculine košer in the sense of ‘butcher’ and masculine košer with the meanings of ‘cut’ and ‘food appropriate for Jews to eat’, these are based on isolated documents from the works of authors apparently unfamiliar with the Jewish community. The question then is whether (and according to what criteria) such words should be included in the dictionaries.
Digital twin skin of the drinking water treatment plant - development and use in practice
Sochorová, Helena ; Andreides, D. ; Hložanka, F. ; Prokopová, Michaela ; Stejskal, O. ; Chalupa, L. ; Šmejkalová, P. ; Dolejš, P.
The contribution presents experiences with the development of a digital twin for optimizing the control of the DWTP within the pilot project in the Železná Ruda locality. The digital twin consists of two components, i.e. digital documentation in the object architecture processed by the reverse engineering process, which is data-connected with the real physical infrastructure (virtual replica of the infrastructure) and a simulation tool based on a mathematical model or a neural network (simulation of the water treatment process). Both components are accessible to the user via a web application, in which one can choose from three operating states (monitoring of the real state - default, simulation of scenarios of operating settings and prediction of pressure loss).
Tombstones of the Švamberks of the 15th – early 17th Century from an Art Historical Perspective
Chlíbec, Jan
Only a fragment has remained from the original collection of the Švamberk family tombstones that was still quite large at the beginning of the 18th century. Surprisingly,\neven though the Švamberks were a prominent noble family, the surviving works usually show average or below-average quality which, as far as the 15th century works are concerned, reflect the prevailing condition of sepulchral sculpture in Bohemia of the time. More factors could have been behind opting for an austere form of the tombstone. In addition to the Utraquist criticism of majestic funeral works and the lack of high-quality stonemasons after the Hussite Wars, it could be the general idea of resistance to pride and accentuation of the confirmation of Christian modesty regardless of the buried person’s confession.\n

The methodology for using wiki software in education is available to teachers who want to use wiki software for teaching purposes. It presents the most important characteristics of a wiki environment, deals with the various possibilities for its active use, provides guidance on the use of the existing wiki environment, and on the selection and installation of the software, including its own modules. Special attention is also paid to analyzing the possibilities for the use, passively and actively, of the electronic encyclopedia Wikipedia for educational purposes. The methodology includes the topic of copyright law with regard to the significant systematic underestimation (especially in the electronic environment) of this issue by most teachers. The methodology also explains in detail the use of free copyright licenses, especially the Creative Commons license. The primary form of this methodology is its electronic form in the Enviwiki (see www.enviwiki.cz), where examples of work by students are also located. The methodology is thus freely available to anyone interested in using it, as it corresponds to the philosophy of teaching in an open information space.

Methodology for co-cryostorage of virus isolates in their host plants and relevant data
Bilavčík, A. ; Bjerkestrand, R. ; Bjelland, B. ; Blystad, D.-R. ; Bobrova, O. ; Fránová, Jana ; Faltus, M. ; Hamborg, Z. ; Hammond Hammond, S. D. ; Koloniuk, Igor ; Zámečník, J.
Currently, the nursery production of plant material emphasizes the production of healthy, virus-free material. For this reason, it is necessary to have reference plant materials in which the virus is present. Some types of viruses are difficult to maintain outside the plant host. Because of the potential transmission to outdoor plantings, it is necessary to keep infected plants in special cultivation areas, and net-houses, which brings considerable financial and labour costs. Therefore, a methodology was developed in which the virus status in plant material is analysed, the infected material can be stored using cryopreservation in liquid nitrogen and then regenerated in vitro, and relevant data in GeneEver cryobox can be stored.
Virus eradication method by cryopreservation tool - a cryoknife
Bilavčík, A. ; Bobrova, O. ; Faltus, M. ; Fránová, Jana ; Hammond, S. ; Koloniuk, Igor ; Zámečník, J.
Currently, in the nursery production of plant material, emphasis is placed on the propagation of healthy material. For this reason, it is necessary to pay attention to the topic of healthy virus-free nuclear stock when preserving collections of genetic resources of vegetatively propagated plants. Some raspberry viruses are difficult to remove from the plant host by existing methods of thermotherapy or chemotherapy. That is why, using the example of black raspberry necrosis virus (BRNV), a comprehensive methodology for testing health status and cryotherapy in raspberry was developed. The resulting methodical procedure makes it possible to eradicate BRNV from raspberry using cryotherapy of in vitro cultures. The user of the methodology is the Ministry of Agriculture, which applies it as part of the 'National Program for the Conservation and Use of Genetic Resources of Plants, Animals and Microorganisms Important for Nutrition and Agriculture'.
Supporting grassland biodiversity through small-scale fallows
Fabšičová, Martina ; Frei, I. ; Jiroušek, M. ; Smetanová, S. ; Šipoš, Jan ; Trnka, F. ; Vymyslický, T. ; Winkler, J. ; Zdražílková, M.
The main goal of the methodology presented here is to propose a system of recommendations for applying fallow in nature conservation based on our experiments. Due to the independence of agricultural land outside specially protected areas with direct payments from the Ministry of Agriculture, it is currently important to apply the methodology, especially in large protected areas where the primary objective is to preserve natural and cultural heritage. Annual and perennial fallows, i.e. biotopes adapted toregular soil disturbance, offer suitable habitats for a number of now-threatened plant and animal species. This is a technically and economically easy solution, but it is challenging to select a suitable location to achieve maximum efficiency. Ploughing must not be used in species-rich grasslands which would lead to ruderalization and degradation of the native vegetation. Furthermore, it must not be carried out on land at risk of erosion. Linear grassland margins, which have been ploughed in the past, recently grassed fields, fallows, and field margins should be selected for fallow management. Nearby populations of invasive plant species can be a problem. Our recommendations for the use of fallow management relate primarily to land in lowland and upland areas with a lower soil nutrient supply.
The methodology of integrated Colorado potato beetle management in potatoes under new agro-environmental conditions
Doležal, P. ; Hausvater, E. ; Baštová, P. ; Kocourek, F. ; Stará, J. ; Sedlák, P. ; Sedláková, V. ; Skoková Habuštová, Oxana ; Nermuť, Jiří
The aim of the certified methodology is to provide updated principles of effective integrated Colorado potato beetle management for the agricultural practice resulting from new findings on biology and population genetics in the situation, when the range of effective insecticides has been reduced, pest resistance to insecticides has been increased and agro ecological conditions have been changed. Colorado potato beetle is a very serious insect pest of potatoes, highly mobile and able to quickly develop insecticide resistance. The methodology was drawn up based on the newest findings on the efficacy of existing insecticides registered against Colorado potato beetle, taking into account newly acquired knowledge in the field of pest resistance to active ingredients and a necessity of ever increasing multiple applications.

Methodology for rapid, comprehensive, independent decision-making on the need, effectiveness and interaction of adaptation measures in river basins under climate change
Fischer, Milan ; Zeman, Evžen ; Vizina, A. ; Hanel, M. ; Bernsteinová, Jana ; Tachecí, P. ; Štěpánek, Petr ; Pavlík, P. ; Máca, P. ; Ghisi, Tomáš ; Rapantová, N. ; Bláhová, Monika ; Janál, P. ; Trnka, Miroslav
The aim of the methodology is to present methods for quantifying the impacts of projected climate change on the water balance when applying adaptation measures in the Czech Republic for the next few decades. Adaptation measures should contribute to the sustainability of the water balance in all major user segments of water use and management in the basin. The main principle is the use of hydrological models to transform climate change scenarios into time series of hydrological conditions and to quantify the overall water balance of the basin using different types of adaptation measures and their implementation over time. Special emphasis is placed on the evaluation of combinations of adaptation measures that cannot be analysed by simplified methods. The methodology is designed to search for the optimal combination of adaptation measures in the assessed catchment. The proposed approach eliminates the shortcomings of effectiveness assessment from the perspective of the exclusive user of the water resource, as the evaluation of the effectiveness of adaptation measures is carried out in the form of a multi-criteria analysis of the evaluation of the outputs of the simulation model for predicting the water balance in the whole basin. This methodology can be used to assess different adaptation measures in all basic segments of water users: agriculture, forestry, energy, water management and others.
