Needs of patients with age related macular degeneration

This bachelor's thesis deals with the area of specific needs in patients with age-related macular degeneration (AMD). Objectives: The aim of this thesis is to provide an overview of the changed needs of patients with AMD. Methods: To achieve the aim of the thesis, a questionnaire survey was conducted. The target group of the survey were patients undergoing anti-VEGF therapy at an unnamed eye clinic. A total of 70 questionnaires were included in the survey. Results: Based on the statistical evaluation of the data, it was found that AMD significantly affects the lives of respondents in many areas. The majority of respondents identified independence and self-sufficiency as the most affected aspect of their lives. More than half of the respondents (55.71 %) have problems with mobility and everyday activities. AMD also negatively affects the mental health of patients, 70 % of respondents are afraid of depression or anxiety. Respondents find meaning and fulfillment in life, especially in the areas of family and friends relationships, leisure activities, spiritual development, education, gratitude, and a positive attitude towards life. This method was applied to the analysis of food supplements based on goldenrod: drops Celík zlatobýl (Valdemar Grešík), Celík obecný BIO (Bylinné kapky), Celík obecný (Bylinné kapky), Zlatobýl (Green Idea), Zlatobýl - tinktura z pupenů (Serafin), Zlatobýl - tinktura z bylin (Serafin), Zlatobýlová směs (Dědek kořenář), Zlatobýl (Inca Botanica), and capsules Celík (Ex Herbis). Current treatments only temporarily delay the progression of disease. The main limiting factor for developing of new therapeutic compounds is the simultaneous presence of multiple pathologies. Thus, a new group of potential drugs called multi-target directed ligands (MTDLs) emerged as an alternative option to combat AD. To this date, dozens of MTDLs have been published. As part of the experimetnal section of this thesis, a series of small molecules based on quinazoline scaffold was rationally designed with the intention to influence the activity of acetylcholinesterase, butyrylcholinesterase, monoamine oxidase A and B (MAO-A/MAO-B), and the GluN1/GluN2B subunit of N-methyl-D-aspartate receptor (NMDAR). Mechanisms responsible for sex bias in the prevalence of autoimmune diseases

Studies have shown that women typically have stronger immune responses than men. This can also make them more susceptible to autoimmune diseases, with females comprising 80% of autoimmune patients. In women, one X chromosome is epigenetically silenced through a process called X chromosome inactivation. However, some immune-related genes located on the X chromosome, such as TLR7 and CD40L, may escape this silencing process in certain cells or tissues. This can result in a sex-biased expression that contributes to the development and susceptibility of autoimmune diseases like lupus, rheumatoid arthritis, and Sjögren's syndrome. Another interesting factor that explains this sex bias is lncRNA XIST, which is a primary regulator of the inactivation process. It plays a crucial role in lupus development and creates an XIST ribonucleoprotein complex with proteins that help establish a repressed chromatin state on the inactivated chromosome. Importantly, many of these proteins are autoantigens in various autoimmune diseases and may serve as clinical markers for the diagnosis of these conditions. The role of infection of HPV and EBV in the etiology of nasopharyngeal and cervical carcinoma

Human papillomaviruses (HPV) and Epstein-Barr viruses (EBV) are common DNA viruses that contribute to the development and progression of tumours. These two groups of viruses are associated with more than 600,000 cases of newly diagnosed tumours of different anatomical locations annually. However, carcinogenesis is not a part of their normal productive life cycle. High risk HPV infections are mainly associated with head and neck tumours and anogenital cancers. EBV infections are associated with a range of acute or chronic diseases and tumours, with Burkitt's lymfomas (BL), Hodgkin's lymphomas (HL) and nasopharyngeal carcinomas (NPC) being the most prominent. In recent years, it has been hypothesized that EBV infections may be involved in the development of cervical cancer in woman and that HPV infections may play a role in the development of NPC. The aim of this thesis is to characterize above mentioned groups of DNA viruses and to summarize currently available information about the potential role of these viruses in nasopharyngeal and cervical carcinomas. Personality predictors of exercise addiction in competitive sport

Exercise addiction is becoming a recognized concern, particularly prevalent in elite athletic environments where the pursuit of perfection is emphasized. Despite growing awareness of problematic exercise behaviors, research investigating the underlying risk factors remains limited. This study aimed to examine personality predictors of exercise addiction in Czech elite athletes. A sample of 168 competitive athletes, aged 18 to 30 years, completed assessments including the Exercise Addiction Inventory, Sport Motivation Scale-6, the Athletic Identity Measurement Scale, Performance Perfectionism Scale-Sport, and the Neuroticism Scale from Big Five Inventory. In terms of prevalence, the findings revealed that 16.5 % of athletes exhibited problematic signs of exercise addiction, while 76.7 % were potentially at risk with low severity of symptoms. No significant differences were observed between different groups of sports. Using multiple linear regression, the study demonstrated a significant overall effect of personality predictors on exercise addiction. Identification of "red flag" items in the S-PMV screening questionnaire

The recognition and identification of developmental delays or developmental disorders in early childhood development can be difficult, and various screenings are often used. The Screening for Psychomotor Development ("S-PMV") consists of ten questionnaires and is designed for children between birth and three years of age. Since 2019, it has been recommended by the Ministry of Health of the Slovak Republic as a tool for paediatricians, which allows them to monitor the development of the child and catch any delays in its progress. The aim of the present thesis is to identify the "red flag" items in motor, communication, cognition, adaptive and social behaviour in the S-PMV at each assessed age of the child, which the majority of children (>95%) of a given age fulfil and to distinguish abnormalities in development as a result of it. The research sample consisted of 60,298 completed questionnaires from children aged between birth and three years. A total of 48 "red flag" items were identified in all questionnaires except for the S-PMV 9 questionnaire for children between 11-12 months of age, which could be due to the fact that children at this age have reached different levels of ability in the domains of interest and therefore the items in the questionnaire may have been chosen unsuitably.
Dispersal and personality

Dispersal of animals is an important area of ecology, the study of which is crucial for understanding population composition and dynamics. The acquired knowledge has broad applications in various fields, whether in the efforts to introduce endangered species or, conversely, to suppress the spread of invasive ones. However, dispersal begins at the individual level, so it is first necessary to determine which traits dispersing individuals possess. One possible aspect in which dispersers may differ from residents is their personality. This bachelor thesis provides a summary of literature that focuses on personality predictors of dispersal, comparing personality traits among each other, both within and across three selected taxa: fish, birds, and representatives of mammals - rodents. The literature suggests that in the context of individual personality, besides others, three main types of dispersal exist. In the first type, primarily observed in early stages of life, dispersal of more submissive individuals of territorial fish and rodents is enforced due to aggression from more dominant individuals. However, in birds, it is necessary to secure suitable territory as quickly as possible, leading to the opposite dependence - earlier dispersal of dominant individuals. Influence of urbanization patterns on the escape behaviour of birds

Urbanization, i.e., the anthropogenic transformation of natural habitats into urban areas, has significant impact on the biodiversity. One of the key factors for successful animal survival in urban environments is the increase in their tolerance towards humans manifested by their decreased shyness. One of the field methods for assessing the level of the shyness across different individuals, populations, and species of animals is measuring their flight initiation distance. In this thesis, I focus primarily on birds, as their responses have been most extensively studied. Previous studies focused on differences in escape behaviour among birds mainly between urban and rural areas, and focused mainly on Europe and North America. However, human settlements vary in their history, size, population density, and other characteristics. Therefore, the observed patterns in bird behaviour may not always be universally applicable across all cities. In this thesis, through a literature review, I summarize how the escape behaviour of birds differs not only between urban and rural environments but also depending on the characteristics of human settlements, such as their size, history, degree of urbanization, the density of the local human population and the ecoregion. The genus Neotinea Rchb.f. in Europe

The thesis deals with the genus Neotinea from the phylogenetic, molecular and ecologic points of view. Most of its species have been classified in the genus Orchis until the use of molecular methods. Within the Orchidoideae, Neotinea forms a sister clade to the genera Anacamptis, Serapias, Ophrys and Himantoglossum. Six species are accepted now. The current taxonomic circumscription calls for a revision under the influence of new knowledge. Especially in N. tridentata, which contains several well-differentiated lineages and is the ancestor of the tetraploid N. commutata. Also the relationship between N. lactea and N. conica remains unresolved. The genome size within the genus ranges from 6.48 pg in N. maculata to 31.14 pg in N. lactea. All species except N. commutata, whose tetraploid origin is debated, are diploid (2n = 42). Partial endoreplication is present in the genus. The range of endoreplicated DNA is relatively narrow regardless of differences in genome size. Species prefer open, slightly basic habitats. In mycorrhizal interactions they appear generalistic with a preference for Ceratobasidiaceae. This research report presents data from the Czech Education Panel Survey (CZEPS) conducted in autumn 2023. The focus is on how first-year male and female students perceive themselves, their academic aspirations, their relationship to school and their level of critical thinking they come to each type of secondary school. Based on responses from more than 22 thousands students from 249 schools participating in the first wave, in this report we focused on outcomes in the areas of wellbeing, choice of secondary school, sense of belonging to school, critical thinking test results and the situation of ethnic minorities.
Basic characteristics of sulfide catalysts for the direct decomposition of H2S

Sulfide catalysts are crucial in the hydrodesulfurization (HDS) of petroleum fractions, facilitating the hydrogenolysis of carbon-sulfur bonds. This process generates significant volumes of hydrogen sulfide (H2S), which is typically processed in the Claus unit. However, excess H2S can overwhelm the Claus process, creating both operational and environmental challenges. Researchers are now exploring sulfide catalysts for the direct thermo-catalytic decomposition of H2S to address this issue. Supported catalysts, like Mo/Al2O3, and commercial catalysts, considered typical monolayer-type well-dispersed catalysts, show the highest O2 uptake, ranging from 41.29 μmol/g for MoS2/Al2O3 (KLO) to 58.70 μmol/g for the commercial 715. Unsupported multilayer catalysts, such as Ni/MoWS2 (KKP), exhibit lower O2 uptake at 2.54 μmol/g, though it demonstrates high HDS activity with thiophene. Moreover, O2 uptake effectively describes the sintering of the Mo monolayer phase during the thermocatalytic reaction of H2S at 800°C. These O2 uptakes clearly correlate with the XRD patterns, where the MoS2 phase is X-ray amorphous with an O2 uptake of about 19.04 μmol/g, while the MoS2 phase in the spent catalyst shows certain crystallinity exhibiting an average particle size by Scherrer equation of about 6 nm and the O2 uptake of 1.63 μmol/g. O2 chemisorption thus serves as a valuable measure of the dispersion of Mo sulfides. The results obtain with the study of CoMo/Al2O3 (KJJ-family catalysts), highlight the correlation between the increasing amount of adsorbed O2 with the increasing nominal loading of MoO3 and the increasing rate constant of thiophene kTH. This research reveals the potential of these catalysts in hydrocarbon refining. The findings underline the connection between catalytic chemistry, process engineering, and environmental sustainability, with the objective to enhance industrial refining processes.

The Czech Statistical Office has been carrying out a statistical survey on the waste generation and management every year, already since 1992. The scope and structure of the survey serve as a basis to ensure reporting duties of the Czech Republic following from the Regulation (EC) No 2150/2002 of the European Parliament and of the Council on waste statistics; the Czech Statistical Office is the administrator thereto. In 2020, the planned process of modernizing the methodology, data collection and processing was completed, which, compared to previous publications, brings the changes in the content of the publication and also in the form of the presented results.

Most of the central banks under review, including the ECB, are keeping their interest rates at low levels, and several are also continuing their unconventional monetary policy programmes, although the ECB, for example, has reduced its monthly asset purchases. A major exception is the US Fed, which, as expected, is continuing to raise its key interest rate and, according to outlooks, will maintain this trend next year. The UK has also seen its first rate hike in ten years. By contrast, the Hungarian central bank’s new instruments are aimed at lowering yields at the longer end of the yield curve. Entrepreneurship during pandemics and crises in the context of gender inequalities. Policy brief.

Whether the pandemic can be perceived as an opportunity to reduce gender inequality in entrepreneurship was the main question of the participatory workshop that we organized at the Institute of Sociology on March 13, 2024. The discussion during the participatory workshop focused on the barriers that women entrepreneurs face, the tools they can use to overcome them, the introduction of a attendance allowance for entrepreneurs, and the availability and usability of gender-disaggregated data. This policy brief write down the views of relevant stakeholders in the business sector on the topic of eliminating gender inequalities in the entrepreneurship that were expressed during the workshop.

Survey expectations, adaptive learning and inflation dynamics

The use of survey information on inflation expectations as an observable in a DSGE model can substantially refine identification of the shocks that drive inflation. Optimal integration of the survey information improves the model forecast for inflation and for other macroeconomic variables. Models with expectations based on an Adaptive Learning setup can exploit survey information more efficiently than their Rational Expectations counterparts. The resulting time-variation in the perceived inflation target, in inflation persistence, and in the sensitivity of inflation to various shocks provide a rich and consistent description of the joint dynamics of realized and expected inflation. Our framework produces a reasonable interpretation of the post-Covid inflation dynamics. Our learning model successfully identifies the more persistent nature of the recent inflation surge.
Financial skills and search in the mortgage market

Are households with low financial skills disadvantaged in the mortgage market? Using stochastic record linking, we construct a unique U.S. dataset encompassing a rich set of mortgage details and borrowers’ characteristics, including their objective financial literacy measure. We find that households with low financial literacy are up to 4% more likely to search less and lock in at 15-20 b.p. higher rates. Upon origination, unskilled borrowers face a 35-45% higher mortgage delinquency and end up with a 30% lower likelihood of refinancing. Overall, for a $100,000 loan, the potential losses from low financial literacy are more than $9,329 over the mortgage duration. To understand how financial education, more accessible mortgages, or mortgage rate changes affect households with low financial literacy, we formulate and calibrate a mortgage search model with heterogeneous search frictions and endogenous financial skills. Our model estimates show that search intensity and financial skill variations contribute to 55% and 10% of mortgage rate variations, respectively. We find that i) more accessible mortgages lead to a higher delinquency risk among low-skilled households, ii) financial education mitigates the adverse effects of increased accessibility, and iii) low mortgage rates favor high-skilled homeowners and, by reinforcing refinancing activity, deepen consumption differences across different financial skill levels.

Tato příručka se zaměřuje zejména na vztah platformové práce a zajištění bezpečnosti a ochrany zdraví při práci, popisuje psychosociální zátěž pracovníků a specifická rizika, která jsou s platformovou prací spojena, a to zejména s ohledem na algoritmický způsob řízení práce. Při zpracování autorky vycházely zejména z odborné literatury, dostupných studií a průzkumů na toto téma, právních předpisů, soudních rozhodnutí a koncepčních materiálů evropských a mezinárodních institucí. Na závěr jsou uvedeny jako příklady správné praxe možné preventivní přístupy k řešení problematiky platformové práce, a to ze strany digitálních platforem, státních orgánů a institucí v podobě legislativních a nelegislativních opatření, i samotných platformových pracovníků a jejich zástupců.

A Review of Relevant Regulations, Requirements and Assessment Methods Concerning Physical Load in Workplaces in the Slovak Republic

This review brings an overview of the Slovak regulations concerning occupational safety and health at work (OSH) primarily considering the parameter of physical load. In addition to regulations, this article focuses on particular requirements at workplaces with an increased physical load and describes the permissible values of the physical load (see tables). Attention is given to assessing the working posture and handling of loads. The main purpose of the evaluation is to determine the parameters of physical load, which is the first important step in the process of health risk assessment. Based on the physical load assessment and the categorization of tasks for the physical load factor in the Slovak Republic, further steps and measures should be taken to improve working conditions, to reduce the workload, number of health problems, fatigue, and ultimately to prevent accidents at work and work-related musculoskeletal diseases.

Jako nástroj pro mapování situace využívá hodnocení pomocí indexů, protože je u nich výrazná snaha o komplexní propojení nejrůznějších témat, která spolu zdánlivě nesouvisí, ve skutečnosti se však navzájem výrazně ovlivňují. Pořadí Česka se v mezinárodním srovnání u jednotlivých indexů liší v závislosti na zvoleném tématu, metodě výpočtu a vstupních datech. Jejich pochopení napomáhá uvedení silných a slabých stránek indexu a popisu, jak jsou jednotlivé indexy konstruovány – které indikátory a vstupní data do jejich výpočtu vstupují i popis metodiky výpočtu.

The article focuses on the impacts of Directive 2019/904/EU on the management of plastic beverage bottles with a volume of up to three liters. It describes how various European countries and the Czech Republic deal with loading and negative effects, describes three main collection methods which are distinguished in European countries and discusses whether to maintain the existing collection system or whether it would be better to introduce a back-up system to achieve the value of sorted collection at 77% by 2025.

Spectral Indices as a Tool for Hop Growth Evaluation

The use of unmanned aerial vehicles (UAV) to monitor crop growth is nowadays a common non-invasive way how to obtain information on the current state of crops. Spectral indices derived from multispectral images obtained in the right growth stage can then serve as a good data source for agro-technical interventions and yield estimation. Hop belongs among the crops where it is possible to scan the individual growth parameters very exactly. In the year 2021, significant precipitation amounts were recorded during the growing season, when it turned out that UAVs are a very powerful tool for determining the quality of production or quantification of vegetation damage compared to the previous year (2020). It was found that the common spectral indices were possible to use for calculation leaf area, structure, vigor and chlorophyll content of hop gardens.

The issue of access to land and buildings within field works of environmental projects implemented in the public interest

Various field activities are also carried out in the public interest, which concerns a few natural science disciplines. In environmental fields, on-site observation or sampling is the basic tool for collecting information, which is usually processed into records of registration databases. In the case of the contaminated site inventory project, the right to visit sites and collect information on site is not specified by law in the Czech Republic. On the contrary, the field worker must respect the legal requirements regarding the protection of property, privacy, and the processing of personal data. In the National Inventory of Contaminated Sites (NIKM) project, a field verification of conditions at known or suspected sites was one of the basic methods of updating and supplementing the registry database of contaminated sites. About 30,000 locations were included in the field verification. The project NIKM was completed by the end of 2021. Subsequently, in the presented case study, we evaluated the extent and types of problems that were associated with site visits and analyzed the reasons for not examining them "in situ". Since the entry of non-visiting and/or non-reconnaissance of the site cannot be completed in a separate section in the Contaminated Sites Registration System (SEKM) and sice there is no prescribed or required exact statement that could be used as a search / filtering parameter, a full-text search in tabular exports in EXCEL format was used for database analyses, according to various relevant statements, words, and parts of words. In the NIKM project, the share of 317 cases of non-visiting the location was only 1.1% of the total number of approved (evaluated and excluded) locations, and the share of 171 unvisited evaluated locations from the number of evaluated locations was 1.7%. At the same time, the number of 171 evaluated locations represents only 0.6% of all approved locations. These are values that can be considered acceptable from the point of view of the quality of the records of the entire SEKM3 database.

Hydrogeological and hydrological data contained in records of contaminated sites in the SEKM database

As of May 2022, approximately 10,200 contaminated and potentially contaminated sites are registered in the database of the Contaminated Sites Registration System (SEKM) updated by the National Inventory of Contaminated Sites project (NIKM, 2018-2021). Each structured record of such site contains a number of hydrogeological and hydrological characteristics, which serve primarily to assess the possibility of contamination migration. The most important synthetic parameter of the hydrogeological and hydrological conditions at the site is the "Possibility of migration of contamination". The basis for the assessment is the assignment of the site to one of the five groups of types of hydrogeological structures of contaminated sites. For the set of localities of the highest priority category A, we demonstrate statistical data on the occurrence of the monitored "hydro" parameters, broken down by five types of migration options, for each subcategory A1, A2 and A3. The summary map of the territory of the Czech Republic shows the location of 451 subject contaminated sites of priority category A.

Tvář české krajiny v prostoru a čase: Mapování CORINE Land Cover 1990–2018 v socioekonomických souvislostech

Publikace pojednává na podkladu dat mapování krajinného pokryvu CORINE Land Cover o fenoménech v české krajině. Zabývá se změnami krajinného pokryvu a využití půdy v Česku během tří desetiletí po sametové revoluci a uvádí změny krajinného pokryvu do socioekonomických souvislostí. Hlavní trendy změn krajinného pokryvu jsou zaměřeny na změny v osídlení, zemědělství a lesních porostech, a jsou doplněny o několik případových studií zaměřených na suburbanizaci, zemědělskou extenzifikaci v pohraničí nebo lesní změny na Šumavě a v Jeseníkách. Interakce mezi ekonomickým systémem a přírodou jsou zkoumány pomocí konceptu sociálního metabolismu a metody Analýzy materiálových a energetických toků s cílem popsat, do jaké míry závisí vývoj změn krajinného pokryvu a následných materiálových a energetických ukazatelů na socioekonomických podmínkách, a zároveň identifikovat hlavní procesy, z nichž tyto změny vycházejí. Analýza navazuje na prioritní oblasti Strategického rámce ČR 2030, které obohacuje o prostorový rozměr. Cílem knihy je oslovit a informovat ty, kdo rozhodují, odbornou veřejnost, pedagogy a absolventy vysokých škol i odborné novináře, jelikož pochopení dopadů změn krajinného pokryvu je předpokladem udržitelného rozvoje území a je důležité pro zachování funkčnosti ekosystémových služeb, zejména pak v reakci na vhodné územní plánování.

Informuje o možnostech řešení doručenek v případě výkonu spisové služby v elektronických systémech a upozorňuje na připravovanou změnu v souvislosti s modernizací Národního archivního portálu. Na dvou příkladech z veřejných zdrojů zde naleznete ukázku dobré a špatné praxe při vyhotovování dokumentů. Measurements of New Particle Formation in the Atmosphere

In this work, the formation of particles and their transformation to larger sizes will be studied by measuring the concentration of molecular clusters. The measurement will take place at a rural background site National Atmospheric Observatory Košetice. At the ground level, a Neutral and Air Ion Spectrometer (NAIS) able to measure both neutral and charged clusters or particles ranging from 0.8 to 40 nm in diameter will be used together with a Particle Size Magnifier (PSM) able to measure neutral clusters as small as 1 nm in diameter up to 12 nm. Larger particles will be measured by a Scanning Mobility Particle Sizer (SMPS). At a height of 230 m above ground level, there will be another SMPS and also a nanoSMPS able to detect particles as small as 1 nm in diameter.
Thin Film Photocatalysts on the Basis of g-C3N4

This study revolves around the synthesis of powder g-C3N4, fabricationof thin g-C3N4 films and their efficacy in degrading pollutants, assessed through experimentation in a batch photoreactor (BPR) and a slit geometry photomicroreactor (PMR)2. The photocatalytic activity of g-C3N4 was tested with 4 different model substances: fluorescent dye, rhodamine B, was used as a reference substance and three separate antibiotics, which are emerging wastewater pollutants, namely tetracycline, trimethoprim, sulfamethoxazole.

The present study focuses on the flow of Taylor bubbles, representing the interaction between the channel wall and the gas phase, and the collision of\na bubble with a vortex ring, serving as a model liquid-gas interaction. Understanding the former case can be essential to prevent the membrane\nfouling or the flow induced corrosion. The dynamics of the latter case is the predictive criterion for whether the initial mother bubble will break up into the daughter bubbles or not2 and this process can therefore be used as an idealized model of turbulent bubble flow.

Investigation of Mechanical Processing of Solar Photovoltaic Panels

This work aims to describe the possibilities of mechanical treatment of solar photovoltaic panels. In the first part, the shredding and milling process for the transformation of the product (whole panel) into material that can be further processed, was investigated. Milled material was sieved and analyzed. It was observed that this shredding also works as a selective grinding because three main products were produced. The second part of this work was aimed at the physical concentration of silver in real samples of milled solar photovoltaic panels by Knelson separator. This device (type of centrifugal separator) showed the ability to significantly increase the amount of silver in the outcome stream.

Synthesis of Fluorinated Methyl β-Lactosides

In my contribution, I will describe the synthesis of a complete series of mono-deoxyfluorinated methyl β-lactosides (1–7). Their affinity towards hgals will be determined by analytical method ELISA. The synthesis relies on monosaccharide building blocks, which serves as key intermediates that are transformed into the target products through glycosylation followed by final deprotection steps.

Ionic liquids (ILs) are emerging as promising alternative solvents in the field of natural substance extraction, offering significant advantages over traditional solvents. Since the last decade, we notice the increasing use of greener solvent alternatives such as Natural Deep Eutectic Solvents (NaDES) or ILs for the extraction of highly valuable compounds from natural sources such as lignocellulose. During this presentation, the influence of water on heat capacity, density, speed of sound and TGA will be discussed and the preliminary results for lignin dissolution willbe presented.

Growth and Phosphorus Dynamics During Tolypothrix tenuis Cultivation – a Cyanobacterium of Biotechnological Potential

Here I investigated the course of biomass growth, phosphorus uptake and changes in intracellular phosphorus content in T. tenuis in a batch cultivation. Cultivation experiment was held in multiple bubble columns during a light regime (alternating light and dark periods 16 and 8h). Thanks to this experiment I was able to obtain a growth curve of T. tenuis. The decrease of phosphorus concentration in the medium was disproportionally faster\nthan biomass growth resulting in significant oscillations of phosphorus content in the biomass, which are typical characteristics for LPU. It was also observed, that after all phosphorus was removed from the medium, the algae growth continues over time, while obviously utilizing intracellular phosphorus reserves. These findings are valuable in course of using this cyanobacterium for efficient bioremediation of eutrophic water based on phosphorus removal by targeted algae cultivation.

Drone Based Vertical Profiling of Black Carbon Aerosols at a Rural Background and an Urban Site in Central Europe

This research aims to assess the vertical distribution of BC aerosols using a drone and micro Aethalometer AE51 at two distinct sites in the Czech Republic: 1. NAOK (National Atmospheric Observatory Košetice) representing a regional background, and 2. MFF (Faculty of Mathematics and Physics in Prague) representing an urban site. At NAOK, a 250-meter measurement tower is accessible, providing a means to validate and calibrate the drone measurements.

The methodology for evaluation of environmental education programs

The methodology may be used by public administration officials who evaluate both formal and informal environmental education programs for various reasons (incl. making decisions on the distribution of funds to support formal and informal environmental education programs)., The results can than be used by those who implemented the program to improve it or by its external evaluators such as the donor to asses its efficiency. The aim of the presented methodology is to define the basic terminology for evaluating environmental education programs, describe the basic stages of the evaluation cycle, characterize the basic alternative procedures for evaluating programs, and finally, to identify the basic requirements for evaluation research. Development of the methodology is based on the assumption that the evaluation research in the medium term will become an integral part of environmental education programs. To achieve this objective requires that officials are familiar with the theme and gain a basic understanding of the opportunities and needs of the assessment procedures.

Methods for the detection of Paenibacillus larvae and Melissococcus plutonius using real-time PCR

The methodology is intended for diagnostic laboratories focused on bee infections. Detection of the causative agents of American foulbrood (Paenibacillus larvae) and European foulbrood (Melissococcus plutonius) is essential to prevent the spread of those highly contagious infections of a honey bee (Apis mellifera) diseases. In case of the suspicion, an examination of bee colonies in protective zones of the disease is indicated. The second possibility of use is the confirmation of the infection based on the detected clinical symptoms in the bee colony. The main target user in the Czech Republic are laboratories investigating animal diseases for the State Veterinary Administration (SVS) (accredited laboratories) in case of suspected disease and investigations in protective zones. Other target users are laboratories investigating bee colonies for reasons of preventive surveillance based on the request of beekeepers. The PCR method enables fast and reliable detection of P. larvae and M. plutonius from different samples taken from the bee colony. Compared to classic microbiological techniques, it allows to provide a result within a few hours of receiving the sample in the laboratory. In this way, it is possible to ensure, above all during the period of intense flight activity of bees, early intervention in bee colonies suspected of the occurrence of the mentioned diseases and to prevent the spread of the disease to nearby habitats of bee colonies.

Development of a procedure for the analysis of the T-cell response against SARS-CoV-2 virus

In 2020, the SARS-CoV-2 coronavirus caused a pandemic of covid-19 disease. To detect the cellular response induced by infection with this virus, a highly sensitive ELISPOT assay has been developed that detects specific interferon gamma-producing T lymphocytes after stimulation with synthetic peptides derived from viral antigens. This assay allows the detection of memory T cells directed against the viral spike (S) protein used for vaccination as well as against other viral proteins. This method can be applied to the analysis of the immune response in individuals after covid-19 and SARS-CoV-2 vaccination and may thus be used to determine the immune preparedness of emergency workers.

An analysis of the needs of rural and small communities in terms of sustainability shows that the preconditions for initiating change are, among others, in the social sphere: these include social capital (trust, developed relationships between actors, joint activities) and education appropriate to the conditions of the region (shaping local identity). The Methodology proposes how relationships and cooperation in a given place can be shaped and subsequently used to improve the quality of life in all its dimensions - social, environmental and economic. It shows practices for shaping local culture and lifestyle, presents the experience of the Movement for Change, recapitulates the principles of social entrepreneurship, and refers to existing forms of cooperation in community projects. It shows that opportunities for education, especially based on locally embedded learning, are lacking so that a shared understanding of sustainability principles in the regions to ensure a general awareness of local UR resources and opportunities is missing.

The methodological document describes the selection of environmental indicators for food items integrated in a single indicator of environmental impact, so-called Nutritional Footprint. The functional unit is one portion of a meal and nutritional values with respect to health are also provided. The document includes theoretical grounding and description of the Nutritional Footprint calculation. Based on a pilot testing of the concept methodological approaches to its practical use are suggested, especially for its use in education (EE and ESD).

Methodology of development of case studies for Higher Education and as Open Educational Resources (OER)

The methodology of development of regional case studies of Sustainable Development for Higher Education is aimed at the promotion of the use of practical cases in teaching; it should lead to the development of research tools relevant to local environmental issues and issues of sustainable development of the region, and thus emphasizes the importance of the regional context and specifics. The publication of the cases in the form of open educational resources (OER) enables access to these materials for regionally oriented education at earlier stages of the education system, and can be used for the presentation of regional specifics, and may also serve to promote dialogue concerning the development of the territory between the different social actors. The methodology outlines the procedure for creating educational case studies and shows how they are used in university teaching. Because the methodology is based on qualitative research methods working with direct experience, discussion also addresses the ways and means of creating a knowledge base as an open educational resources (OER), to facilitate the accumulation of this experience. The methodology takes into account the requirement for continuity of learning at different educational levels and gives a brief overview of similar approaches applied at lower levels of the education system; it also shows how the results can be applied in practice. The methods of case study development and use are based on the authors' own teaching experience, and on its theoretical reflection. The methodology clearly summarizes the assumptions for teaching sustainable development issues from a regional perspective; it offers the possibility of linking university teaching and research with practice, which should lead to an increased competence in addressing local issues of sustainable development in cooperation with universities.

The methodology is intended to provide a framework for the use of open web space as a tool for teaching students at the university level, in e-learning or blended learning courses, and in various disciplines with an active and problem-oriented learning component which is desirable from the perspective of education for sustainable development. The virtual space (Wiki), the so-called "social web", provides an interactive environment and supports social skills such as cooperation, self-expression, and communication. This particular methodology is based on the concept of "academic literacy" where students develop their skills along with the writing process. MediaWiki software was used to stimulate meta-cognitive skills, the organization of the writing process, and critical thinking (with regard to resources); a model for an academic environment was presented there. Instructions available in the wiki support pre-writing, a resource search, the writing of a literature review & annotation, posing a research question, developing of text, and peer review. The method proved to be useful in practice; it provided evidence of student work and the results are available for future generations of students.

The methodology for use of wiki software in education

The methodology for using wiki software in education is available to teachers who want to use wiki software for teaching purposes. It presents the most important characteristics of a wiki environment, deals with the various possibilities for its active use, provides guidance on the use of the existing wiki environment, and on the selection and installation of the software, including its own modules. Special attention is also paid to analyzing the possibilities for the use, passively and actively, of the electronic encyclopedia Wikipedia for educational purposes. The methodology includes the topic of copyright law with regard to the significant systematic underestimation (especially in the electronic environment) of this issue by most teachers. The methodology also explains in detail the use of free copyright licenses, especially the Creative Commons license. The primary form of this methodology is its electronic form in the Enviwiki (see www.enviwiki.cz), where examples of work by students are also located. The methodology is thus freely available to anyone interested in using it, as it corresponds to the philosophy of teaching in an open information space.

The Methodology of Team Cooperation and Team Creation in Higher Education

The methodology provides a basic framework for university teachers who use active teaching methods and support collaborative student work. In CR, these methods are used rather intuitively this methodology provides an overview of basic pedagogical knowledge and experience that can help their inclusion in education in all fields of study. The methodology deals with the theoretical aspects of group cooperation: processes, relationships, group dynamics, possible barriers and risks that are typical in this method of teaching. It provides an overview of the possibilities of selecting team members, the social assumptions of group work, ways of building trust and methods of facilitating group processes. It briefly discusses the principles for planning project teamwork, tools for assessment of its results, and outlines the specifics of the e-learning environment. Using a teamwork method in e-learning enables one to benefit from all the interactive possibilities provided by the Internet to foster communication and collaborative features in the very process of learning; thus teamwork could help to adjust the e-learning method for the needs of practice.

Methodology for co-cryostorage of virus isolates in their host plants and relevant data

Currently, the nursery production of plant material emphasizes the production of healthy, virus-free material. For this reason, it is necessary to have reference plant materials in which the virus is present. Some types of viruses are difficult to maintain outside the plant host. Because of the potential transmission to outdoor plantings, it is necessary to keep infected plants in special cultivation areas, and net-houses, which brings considerable financial and labour costs. Therefore, a methodology was developed in which the virus status in plant material is analysed, the infected material can be stored using cryopreservation in liquid nitrogen and then regenerated in vitro, and relevant data in GeneEver cryobox can be stored.
