Lucie Vybulková The study of transforming growth factor receptor during atherogenesis in mouse model of atherosclerosis Diploma thesis Charles University in Prague, Fakulty of Pharmacy in Hradec Králové Pharmacy Background: The aim of this diploma thesis was the establishment of methodology for detection of TGF-βRI, subtype ALK-5, and studying its expression in the aortic root in apoE/LDLr- deficient mice. Methods: Using female mice C57BL/6J strain with a double deficit of apolipoprotein E and LDL-receptor was performed biochemical analysis of blood specimens, histologic staining with oil red for detection of lipids in tissues and immunohistochemical analysis of segments of tissue from the aorta and the top half of the heart. To detect expression of ALK-5 was used EnVision method with DAB visualization. Results: Using biochemical analysis, we confirmed increased levels of cholesterol in mice fed cholesterol diet as compared with mice on standard diet. At histologic staining with oil red we detected increased formation of atherosclerotic lesions in mice with cholesterol diet. Immunohistochemical analysis of expression of ALK-5 we demonstrated high expression of ALK-5 in the tunica media and in atherosclerotic plaques with no significant difference in the two groups of mice. The protein or peptide substances are not appropriate for oral application. One of the many obstacles, is an aggressive environment of the gastrointestinal tract. One way to protect these substances is the preparation of liposomes. The aim of this work was preparation of liposomes of the different composition, choice of the optimum aqueous media, and stabilization using freeze-drying. All samples consisted of egg phosphatidylcholine(EPC) and cholesterol (Chol), with the addition of one out of the substances: cholylsarcosine (CS), stearoylamine (SA) and vitamin E (TPGS), respectively, in various ratio. Liposomes were prepared using a film method and manual extrusion through membrane with pore size 200 nm. The liposome size and polydispersity was measured by photon corellation spectroscopy. After the extrusion, the liposomes underwent freeze-drying in the presence of selected appropriate cryoprotectants: sucrose and trehalose, respectively. Stability has been established by evaluating the visual characteristics of the product (the appearance), solubility in the water and measuring of liposome size and polydispersity.

This project deals with the optical control of welding glass blank. The blank consists two small parts of made of special glass. This blank is an essential part of the measuring device density of oxygen. Measuring device algorithm is based on paramagnetic properties of oxygen. It is a small and precise piece of the whole mechanism therefore the precision and perfection made strict criteria. For checking compliance with those criteria, developed a computer program written in C# programming language. Its task is to rule on perfection produced parts. It also serves to back up data on individual production units.

Investigation of the β-N-Acetylhexosaminidase Stucture from Penicillium oxalicum.

in English β-N-Acetylhexosaminidase (EC 3.2.1.52) is exoglycosidase, which exhibits the unique properties in the filamentous fungi. Enzyme from these organisms are dimeric, inducible and secreted extracelluary. It is expresed as preproprotein, consists of a signal sequence, a large propeptid and a catalytic subunit. Although the enzyme is widely distributed, its structure differs in varies organisms. Bacteria have only monomeric hexosaminidase. Human enzymes are dimeric as well as fungal, but only hexosaminidase from filamentous fungi have the catalytic subunit noncovalently associated with the propeptide. Propeptide is a essential for the enzyme activity. It exists a homologues model of the catalytic subunit of β-N-acetylhexosaminidase from Penicillium oxalicum, but the structure of the propeptide has not yet been solved. The first part of this diploma thesis deals with the optimization of production and purification conditions. The second part deals with structural studies of β-N-acetylhexosaminidases from the filamentous fungi Penicillium oxalicum CCF 3438. These studies were carried out using chemical cross-linking and high resolution mass spectrometry. The combination of these methods revealed region of the noncovalent interaction of the catalytic subunit with the propeptide.

Fragments of a Love Story (in Photographs)

1 Abstract The diploma thesis Fragments of a Love Story (in Photographs) focuses on visuality of a lover's speech; it follows various possible connections between the media of photography and text. In comparison with the disintegrating lover's discourse, as developed by Roland Barthes in A Lover's Discourse: Fragments, and in a dialogue with it, the thesis highlights love story as a traditional way of expressing a love theme. The elaboration of the topic is based on the interpretation of works written by Vasily Rozanov (Opavshie list'ya), Jakub Deml (Listy Jakuba Demla Otokaru Březinovi), Ludvík Vaculík (Český snář), Bohumil Hrabal (Vita nuova) and others. An appendix that includes the discussed photographs accompanies the thesis.

The Painterly Gesture

Diploma thesis called Brush stroke demonstrates technique of materialization brush strokes in an open space. Center thought of the project is application of a specific technology, when color or another material is not applied on surface by brush, but pressed through surface. It is important, that gesture of painting act remains in the color matter. Project works with reflection of painting as an artistic medium, highlighting visual and haptic elements of the result. Thesis is based on invention of technique, it's demonstration and presentation in form of an exhibition.

Innovation processes in the Czech and Bulgarian language situation (based on media language material)

The problems of the innovation processes in the present-day language systems have been essential to the current linguistic research. In general, this thesis is focused on a synchronic contrastive description of the types of innovation processes producing and inf1uencing the lexical dynamics of the Czech and Bulgarian languages. Doing so, it stems from the generally accepted view that, depending on the criterion applied, the individual innovation processes may be made correlative with various dynamic tendencies of a more complex and universal nature, L e. with signs of intellectualization and democratization, intemationalization and nationalization, terminologization and de-terminologization ofthe Czech and Bulgarian literary languages. Chapter I presents the methodological and theoretical starting points of the research. By means of a model designed by B. Havránek and developed by A. Jedlička, the following parallels between the Czech and Bulgarian language situations are being set in it: a) political and social changes in both societies and new orientation of interests of the members of the "language (communication) community"; b) widening the spheres of use of colloquial and non-literary linguistic devices; c) increase in the number of users of foreign languages, especially English; d) spread of new...

Enterprise architecture

This work analyses the enterprise architectures questions. Enterprise architecture is relatively familiar in the field of informatics today. It covers discipline of modeling and planning development of information technology, especially in wide organizations. Enterprise architecture is an important branch under the conditions that system complexity exceeds brain capacities of workers. The first part deals with analysis of enterprise architecture, primarily structure, content and relationship to whole enterprise. The most important part of this section is a characterization of enterprise architecture frameworks that facilitate entire process of formation. The second part deals with introduction of three essential architecture frameworks: Zachman, TOGAF and FEAF. Subsequently FEAF is compared with two previous frameworks. The conclusion of this work contains short comparison of features and contents all three mentioned enterprise architecture frameworks.

Object-relational mapping on .NET

This study concerns the theme of object-relational mapping on the .NET platform. It briefly explains the differences in the relational and object view on data a therefore it describes design patterns which are in object-relational mapping vastly applied. These design patterns are described in order to inform the reader about their advantages as also disadvantages and they are commonly depicted with an elementary illustration. The description of the design patterns is followed with a specification of the .NET framework and other elements (e.g Visual Studio 2008), which are essential to define for further progress in this study. The specification concludes into description of four products, which serve as modelers and generators of the object-relational mapping layer. The goal of this study is to provide a view of the object-relational impedance mismatch issue, means of its solution to the reader and help him with eventual need to invest into products which aid in object-relational mapping on .NET. For potential need to invest in other products than those which are tested in this study is the reader supplied with well-formed rating criteria which are, in my opinion, ready for being standardised. These goals are achieved by generating the mapping layer with all four tested products and description of the mapping process and the layer afterwards.
