Barbora Svobodová Sunitinib effects on endothelial expression of hemeoxygenase-I in normotensive rats. Bachelor thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Medical Laboratory Technician Background: The aim of this bachelor thesis was to define and describe the expression of hemeoxygenase-1(HO-1) in the aortas of normotensive Wistar Kyoto (WKY) rats and evaluate possible effects of sunitinib treatment by means of immunohistochemical and stereological analysis. Methods: Two groups of WKY rats were used for the experiment (6 animals in each of them). Sunitinib was administered to the first group. The other one (the control one) received water only. Application of sunitinib proceeded by following scheme: 8 weeks of using/ 5 weeks rest period/ 2 weeks of using. After that we performed the immunohistochemical analysis on 60 preparations of rat's aortas which observed the expression of HO-1 by methodology En Vision with DAB detection. Then the stereological evaluation was performed. Results: The immunohistochemical analysis showed the expression of HO-1 in the endothelium only and it was noticed just in several animals especially from the group with sunitinib treatment. Conclusion: The immunohistochemical and stereological analysis demonstrated a significant increase of the...
Effect of gene optimization on recombinant expression of human cytochrome P450 3A4
Svobodová, Barbora ; Martínek, Václav (advisor) ; Bořek Dohalská, Lucie (referee)
Cytochrome P450 3A4 is integral membrane protein residing in endoplasmic reticular membrane. In human the highest concentration cytochrome P450 3A4 is expressed in liver, where it plays a major role in metabolism of many drugs and xenobiotics. The main aim of the thesis was to evaluate the effect of gene optimization on heterologous expression of human cytochrome P450 3A4. At first expression constructs based on vectors pET22b were prepared. Then the efficiency of heterologous expression of optimized vs. natural gene sequence encoding truncated form of the human cytochrome P450 3A4 compared. The results show that the gen sequence optimized for E. coli strains K12 was expressed in significantly higher efficiency than the original human gene based on cDNA sequence. Another aim was to evaluate the suitability of pET22b based expression vectors for recombinant production of native (complete) form of human cytochrome P450 3A4. The amount of native form of the protein found in bacterial membrane was however substantially lower then that of the truncated form. Keywords: cytochrome P450 3A4, heterologous expression, pET22b, gene synthesis
Knowledge of Pupils Secondary School in the Area of Selected Zoonoses
Svobodová, Barbora ; Pavlasová, Lenka (advisor) ; Říhová, Dagmar (referee)
The subject of the dissertation deals with the secondary school pupil's awareness of zoonoses. Zoonoses are the diseases transmited from animals to human beings. Those are one of the topics of biology study currently. The main aim of the dissertation is theoretical definition of the matter in the first place. For knowledge testing have been chosen these four illness: Toxoplasmosis, Lyme boreliossis, Rabies and Creuzfeldt-Jakobo disease. With these chosen illness the main research goal is to provide a comprehensive collection of information about the details such as disease transfer, spread of the disease, disease development, cure and avoidance. The research part of the dissertation tries to find out the extent of knowledge about the chosen kinds of zoonoses among the pupils at specialized secondary schools. As a tool of quantitative research has been used a pupil's didactic test. As based on the evaluated results we may say that the pupil's knowledge at explored schools is relatively satisfactory.
Root system development under drought stress
Svobodová, Barbora ; Soukup, Aleš (advisor) ; Fendrych, Matyáš (referee)
Plants actively react to the environmental conditions in such a way that they can use their resources efficiently and be resistant to suboptimal living conditions (e.g., high salinity, drought stress, high radiation, extremely high or low temperatures, insufficient nutrients etc.). One of the responses to drought stress (DS) is change in root system architecture (RSA). Optimized shape of RSA during drought stress can be under some situations "Steep, cheap and deep" ideotype. Steep - the roots grow in an angle ideally perpendicular to the soil surface. Cheap - most of the resources are spent on growing deeper while having small diameter and lots of aerenchym tissue. Plants with this RSA modulation try to reach deeper parts of the soil with greater water reservoirs and to achieve this, they use a wide range of mechanisms. Another change in RSA in reaction to drought stress, which directs the root to areas with more water is called hydrotropism. The key signal pathway which activates a large variety of drought responsive genes is the abscisic acid (ABA) pathway. Plants also have epigenetic mechanisms, which by remembering a stress factor they have already encountered, are capable of faster and more intensive response.

BACKROUND - The environment of children and adolescents attending the Outpatient Addiction Treatment Clinic for Children and Adolescents has not yet been described in detail. The work deals with the description of various aspects of the life of these clients. AIMS - The aim of the bachelor thesis was to describe the environment from which children and adolescents come to ADDA. Specifically, the research looked at divorce rate, family integrity, and family conflicts. Another monitored aspect of the environment was the presence of a substance user in the common household with the patient. The research should also summarize common aspects from the household of children and adolescent users attending the clinic. METHODS - The research method was descriptive statistics, focused on the description of ADDA patients. The information was obtained from structured anamnestic sheets (SAL), which are filled in by the parents when the child enters the outpatient addiction treatment clinic. The data were clearly divided according to frequency into tables and graphs. RESULTS - 47 children (out of 112 answers) live in a complete family, 24 children live in a household that is supplemented by a partner and the remaining 41 children come from an incomplete family in which one of the parents is missing. Only 7...
