Genetic and morphological diversity of Central European populations of the genus Acanthocyclops

Abstract This study aims to compare morphological and genetic diversity of Central European populations of Acanthocyclops vernalis-robustus species group. To asssess a genetic differentiation on a species level, 12S rRNA gene was used because of suitable evolution rate. Additional analysis of otherwise widely used CO I gene was not successful as the PCR amplification of this gene was successful only for some A .trajani populations, and it failed for the others. Genetic analysis of 12S rRNA validated the recent morphological redescription and establishment of three separate species: A. vernalis (Fischer, 1853) A. trajani and A. einslei (Mirabdullayev and Defaye, 2002; 2004). In general on 12S rRNA, the intraspecific variation was negligible in comparison to interspecific one. All of four analyzed populations of A. einslei shared one haplotype, as well as 12 from 14 populations of A. trajani did. The highest intra-specific variability was recorded in 5 populations of A. vernalis (0.5 - 5.6 %). Morphological determination and morphometrics of all analyzed individuals has been performed without using so called micro-characters, on which the description of A. einslei and A. trajani (Mirabdullayev & Defaye 2002, 2004) mainly relies on. Properties of reclaimed post mining catchments and its effect of run of chemistry

After the end of hydric recultivation from the monitored source of watering, artificially bulit-up lakes are fed with the water from their own basin. This water is not so systematically monitored as compared to the water from the new lake and the sources of its filling. This thesis summarizes findings about the developement of artificially made basin in the mining area and explains differences between various basins. Further, this work deals with the characteristics of water feeding lake "Chabarovicke jezero (Milada)", which was bulit-up during hydric recultivation of the mineshaft in the soft-coal quarry Chabarovice. Monitored basins have different area, slope, vegetation coverage and distribution of dissolved compounds. These factors have significant impact on the character of the water in affluents. We found out, that affluents from the own basin have high concentration of dissolved compounds resulting in the unwanted enrichment of the lake. Big problem was the high concentration of sulphates and nitrates, which in all affluents, except for one, exceed tolerated limits for surface waters. Effecto of surface winning on the freshwater quality

The aim of this work is to show, how much different is the flowing mine water and water, which was not affected by surface mining. The literature on this theme was collected and evaluated. The question was studied in the experimental project, which covered examination of mine landscape water in Sokolovsko. In this research, 17 samples of different flowing water were taken and following values were determined: pH, conductivity, acidity, alkality, hardness, content of nitrates and nitrites, amonnium ions, phosphates, iron, chloride, calcium and manganese ions. These data were compared between samples from sites with different degrees of mining extension. The measured values were compared with previously published values from other sites.

Pro potřeby studie byla vymezena následující kritéria hodnocení variant obnovy těžbou narušených segmentů krajiny: 1. stabilita toku energií, vody a látek v krajině s důrazem na malý oběh (odtoky, evaporace), 2. struktura krajiny, prostorové rozložení krajinných prvků (indikační hodnoty krajinného pokryvu), 3. biotická rozmanitost krajiny podmíněná různorodostí typů ekosystémů (heterogenita), 4. migrace organismů v návaznosti na okolní genofondové zdroje (migrace pozitivní, negativní, prostupnost krajiny), 5. stabilita krajiny (respektování paměti krajiny, schopnost zotavení, resistence a resilience po disturbanci, minimalizace vkládání dodatkové energie k zajištění stability, odnos částic, erozní zatížení) podmínky pro rozvoj diverzity krajiny včetně aspektů časových. Byla vytvořena první varianta srovnávací tabulky pro posouzení rozpracovaných jednotlivých kritérií (bez hodnocení kritérií ekonomických).

Zóny Hornojiřetínské výsypky: zóna zvýšeného návštěvnického zájmu A1, zóny návštěvnického zájmu A2 až A6, zóny zvýšeného zájmu ochrany přírody B1 a B2, zóny zájmu ochrany přírody B3 a B4. U jednotlivých zón je uvedena obecná charakteristika, vegetace, fauna, navržená prioritní funkce zóny, stávající negativní vlivy, návrh opatření a náklady na realizaci opatření.
