A study of the modulation of recombinant and native NMDA receptors by physical and chemical factors
Cais, Ondřej ; Vyklický, Ladislav (advisor) ; Blahoš, Jaroslav (referee) ; Chvátal, Alexandr (referee) ; Šťastný, František (referee)
Excitatory synaptic transmission in mammalian CNS is mostly mediated by ionotropic glutamate receptors. NMDA receptors, one of three subclasses of this ligand-activated ion channels family, are involved in memory formation and learning and also play a role in the pathogenesis of various neurodegenerative diseases. Current knowledge about NMDA receptors function is predominantly based upon results of in vitro experiments conducted at room temperature, far from physiological. The aim of this PhD thesis was to describe the temperature dependence of NMDA receptors. We determined the rate constants that characterise each step in the mechanism of recombinant NR1/NR2B receptors activation in the temperature range 25-45řC. The receptor desensitization, resensitization and glutamate unbinding turned out to be the most temperature sensitive of these processes. In addition to that, we described the temperature dependence of deactivation kinetics in various experimental models of NMDA receptors (both recombinant and native). The second part of the thesis focused on the modulation of NMDA receptors function by steroid compounds derived from pregnanolone sulfate, an endogenously occuring neurosteroid. We tested 21 steroids that showed various degree of ability to inhibit (or, in one case, potentiate) the current...

Influence of personality profiles and social labour conditions on activity of military units in military missions abroad
Dziaková, Olga ; Mikšík, Oldřich (advisor) ; Urbanová, Miluše (referee) ; Kořán, Martin (referee)
This comparing analysis judges differences of personality profiles of the soldiers in military missions in present time and during the first "new-age" Czech military missions abroad. lt describes psychological conditions in actual military missions and the ones during the nineties of 201 h century in its social and labor aspects. lt gives the answers to questions how the change of military missions abroad, military duty and army as whole is connected with the shift of personality profile of the people who apply for military service in present and whom the service in missions abroad does not pose problems and compares collected facts with the era of the initial ones. lt explains how the labor psychological requirements are reflected in personality of soldiers who apply for a military service in missions abroad nowadays. The primary task of this expert work it is to find the answer for four questions: "1. Are there any differences in personality characteristics and in ability of soldiers from different units to adapt to missions environment?, 2. What is the difference in personality direction of military professionals in contingents deployed abroad today and during nineties of 201 h century?, 3. What are the differences among the demands of military profession in foreign military mission and real personality...

Exploring models of work with family whose children suffer from severe forms of epilepsy
Kynclová, Dana ; Goldmann, Petr (referee) ; Procházková, Jana (advisor)
This work deals with families whose children suffer from a serious form of epilepsy. On a general level it describes to what extend the condition adversely influences each child's development stage. Then it focuses in detail on the impacts on the child's mental health, as well as on various constraints imposed by this condition. The study also explores the topics of the parent's acceptance, the consequences on the parent's approach to upbringing, capacities for coping with stress and the common ways of families' adaptation to the situation. The key topic of this study is the impact of the child's illness on the family, which is seen as a system. The essential thesis is that a change of one family component (the sick child) influences the entire system paradigm. Therefore in this sense the functioning and healthiness of a family is for the purposes of this study defined as the ability of a family system to cope with stress and adapt to it. The family stress coping strategies are seen as a key instrument for a successful adaptation. Anything a family does for strengthening and using resources for endurance falls under a stress coping strategy. Since one of the possible strategies is the use of professional assistance services, a part of this study also elaborates on possibilities for an expert support...

Working strategies of nurses on manager posts
MARKOVÁ, Eva
The thesis "Working strategies of nurses on manager posts" deals with issues of the professional role and manager position of a head nurse. With regard to changes in the area of health policy and health care providing, as well as is the relationship with the issue of professional fulfilment of women on the labour market generally, it is important to know views, ambitions, and motivations of nurses, and their working strategies on managing positions. It includes aspects of harmonizing their family and work roles, as these are the moments determining the position of most women in our working conditions to a significant extent. It is to say, managing of the nursing personnel and health care is a typical feminine position in the Czech Republic, adapting to given rules of the work organization and hierarchical structure of health institutions. The purpose of the thesis is to outline such a frame of reference, which would enable to better understand values and attitudes of head nurses. An indispensable part of this outlook is to understand, how their carriers developed, to identify their managing styles and guiding of the nursing team, as well as to map how their personal and professional roles pervade and affect each other. In this process, revealing long-term working strategies of head nurses is an important source of knowledge and at the same time, an attitudinal reflexion of these contexts by nurses themselves. Therefore, the development of the role of a nurse - manager (specifically, the head nurse and the ward sister) was described as the first item in the framework of the professional live of managing nurses; and conclusions of a series of research studies, focused on these issues, were presented. Additionally, the specificity of management in the nursing conditions was analyzed, including changes after 1989, and possibilities of education of nurses - managers. Finally, an empiric probe into the explored points at issue was carried out. Data acquired by means of semi-structured interviews with selected head nurses in faculty hospitals were integrated with the help of one of classical approaches, through so-called Grounded theory method. On basis of the data analysis, a model of professional paradigm was created, demonstrating several phases of the professional career, through which a head nurse passes, which were called for illustration: Seeking of domain, Circle (changing of nursing lines and sites), Virtue of necessity (situation, where a nurse caring of a child chooses a workplace, which enables such care), and Anchoring in the nursing branch. Then, the manager's career of the nurse unwinds form the "Anchoring in the nursing branch" phase. Life stories of head nurses, built on information from qualitative interviews, include predisposing factors for a managing position. Last but not least, it is also about some imprinting aspects from the childhood and adolescence (e.g., number of siblings, reasons for the selected study). After certain time, a nurse becomes a shift leader and a deputy ward sister, and subsequently, a ward sister and a head nurse. According to circumstances, she uses a combination of several managing styles in this position, like authoritative, democratic, and consultative. The theoretical model of professional paradigm of head nurses, construed on basis of empiric data analyses, opens a trail for further investigations, where it may be possible to examine some consequences in more detail, like the course of the professional career of nurses, their managing styles, etc. The acquired and classified evidences contribute to enriching current state of knowledge of certain segments of nursing issues. For instance, the top management of health institutions may use them in planning and implementation of some policies in the area of human resources, or educational institutions may use it for better and more accurate setting of educational concepts of future head nurses.

Possibilities usage base - cognitive procedures in cognitive and personality expansion scholar

This diploma theses focuses on the possibilities of metacognitive procedures use in learning and personal development of a student. It consists of theoretical bases of the topic as well as of empiric research of "minutes of learning", one of the metacognitive strategies which help students to adapt relevant learning abilities. In the theoretical part we can find the principles of education for 21st century and the key skills of a learner, as well as the fundamental curricular documents. In other chapters, the learning and personal development of a student is being focused. There are seven topics necessary for complex development of learner's personality. We get an overview of psychological concepts of the topics, and an insight of different psychological theories which are concerned with metacognitive strategies. The conditions for development of metacognitive dispositions of a learner are mentioned in the last chapter of the theoretical part. The methodological part states the aim of empiric research, describes the method used and defines the chosen sample. The third part - empiric research - examines the influence of the metacognitive strategy "minutes of learning" on the overall approach of a learner towards their own learning process. The empiric part consists of the analysis of research material, ways...

Options assessment of stereognostic hand function in children with hemiparetic form of child's cerebral palsy and in relatively healthy children
Hadravová, Vendula ; Nováková, Tereza (advisor) ; Novotná, Irena (referee)
Child's cerebral palsy is a neuro-developmental disease, its manifestation is not only a motor deficit, but also often a sensitive deficit. Nevertheless we often meet in the physiotherapy treatment of the Child's cerebral palsy with a solution of motor ability, sensitive deficit is often neglected. Failure of sensitivity to the individuals brings distorted perception of the surroundings and himself. The main component is just stereognosis, which does not inform us only about the properties of the stimuli, but helps us to navigate in the space and aware of the different parts of the body. Goal of the thesis: The goal is to work through several tests to assess the difference of the hand stereognostic function by a group of children with hemiparetic form of the Child's cerebral palsy and a relatively healthy group of children. Stereognosis of right and left hand is separately compared within each group in the results obtained. Both groups are comparing against each other. Another purpose of this work is to make a measurement of the time required to detect its own subject and in both of these groups of children. Equally important purpose is the assessment of usability of Petrie's test with pediatric patiens. Method of the solution: This work includes research, conducted in two groups of children aged between 7...

The Recruitment of Artistic Workers in the Sphere of Theater/Musical
Sovová, Andrea ; Němec, Otakar (advisor) ; Stříteský, Marek (referee)
This project deals with personnel recruitment in (drama)theaters and in musical theaters. The thesis consists of theoretical and practical parts. The theoretical part contains the development of social function of the theater as a culture phenomenon (it focuses also on acting and the relationship between stage and auditorium). The practical part of thesis examines the whole acting branch within the context of Czech republic by combining an understanding of the key aspects of the profession: remuneration, legislation, education, ways of recruitment etc.). There is also a description of other critical concerns of the sphere: the personal work, especially given the contracts, working environment, trade unions as well as motivation, adaptation, conducting, evaluation and control of actors etc. Using a mixed methods approach combining interviews with a production manager, psychologist, 1 quantifiable survey (made by Czech Trade Union of Actors), and interviews (and articles) in media, this study compiles accounts and ways of recruitment as well as other factors of this profession(such as psychological aspects). Results from both the surveys and interviews reflect the sphere in the Czech republic and partly in foreign countries (completed with the findings of an international survey made by EuroFIA, European section of International Federation of Actors).

Effect of intravenously applicated lactoferine on early faze; of myocardial ischemia II. Biochemical examinations
Písaříková, Veronika ; Semecký, Vladimír (advisor) ; Mladěnka, Přemysl (referee)
Lactoferrin (Lf) is an 80-kDa iron-binding glykoprotein. Lf is present physiologically in exocrine secretions, eg. tears, saliva, milk, sinovial fluid, seminal fluid and in the secondary granules of neutophils. The precise function of Lf in organism is considered to be very complex and it is still a hot subject of scientific disputation. Lf was documented to act as antimicrobial, antiinflamatory and antitumoral agent. Another of its properties is ability to inhibit hydroxyl radical formation via Fenton reaction. Afinity of Lf to iron is about 260times higher than that of transferrin. Isoproterenol (ISO) has been used as a model compound to induce infarct-like lesions in the rat and various other animal species. Administration of necrotic dose of ISO caused ischaemia followed by damage of the myocardium. Ischaemia alters iron homeostasis and redox-active free iron, which catalyses ROS-generation. The only possibility for myocardial tissue recovery in acute moycardial infarction represents the reperfusion of ischemic myocardium. But the whole process is associated with damage of myocardium due to burst of hydroxyl radical catalyzed by free iron (Fenton reaction). In our experiment we studied the effects of Lf in a catecholamine model of myocardial injury. We had four experimental groups divided according to...
