Study of signal systems with special focus on the role of nitric oxide: gene expression, regulation and pharmacological modulation in hepatocytes and adipocytes

Nitric oxide (NO) is involved in surprising array of vital physiological and pathophysiological phenomena. The expression of nitric oxide synthases, endothelial (eNOS) and inducible (iNOS), was demonstrated in rat liver and white adipose tissue among others. Therefore, the goals of the present thesis were to provide in vitro data about NO in the liver and white adipose tissue and to assess: 1) the effects of selective immunosuppressive drugs, cyclosporin A (CsA) and tacrolimus (FK 506), and nonspecific and specific iNOS inhibitors on NO production and iNOS expression during endotoxemic insult using primary rat hepatocyte culture; 2) spontaneous NO production under various culture conditions with comparison of its influence on functional status of hepatocytes in conventional cell culture and in hepatocyte bioreactors; 3) effects of S-nitroso-N-acetyl penicillamine (SNAP), D- galactosamine (D-GalN), lipopolysaccharide (LPS), LPS+D-GalN and thapsigargin (TG), a selective inhibitor of a sarco-endoplasmic reticulum-Ca2+ -ATPase, on apoptotic/necrotic markers in relation to NO production; 4) the effect of LPS on lipolysis in relation to iNOS stimulation; and 5) the interplay between NO production and β3-adrenoreceptor (β3- AR)/cAMP pathway on lipolysis in rat epididymal adipocyte culture. This thesis concerns with education of talented children in primary school. Teoretic part works up knowledge from literature. The knowledge concerns with talent, talented children, education of talented children, models and forms of education, especially forms of segregation and integration, also with diferentiation and individualization of educational process. Practical part connects teoretical knowledge with practical experience and describes specific education of talented children. Another aim is to work up casuistry of selected pupil with emphasis on social role in the class with segregated and integrated conditions and to analyse possible risks and positives of combined model of education.

The Integration of the Czech Republic to the European Union

My thesis with its topic The Integration of the Czech Republic to the European Union presents the study material for teachers from the Civics and Humanitarian basis. The author draws from the experience and due to this, she created comprehensive material. The emphasis is placed on the connection theoretic information with the practice. The thesis is divided into two parts - theoretic - educational material including historian thoughts of the European integration, the foundation of the European Union and the development of the admission of the Czech Republic to the European Union. The practical contains the analysis of the topic with respect to the contemporary conception of the teaching (General Educational Programme), gathering different types of available educational materials and information sources, its classification and at the conclusion the concrete example of the lesson. The main contribution of the thesis is exactly in this part, which draws the attention at the same time on vast bulk of material. This material does not always correspond, so we have to classify it. My work deals with Soren Kierkegaard's and Martin Heideggr's concepts of dread. The dread plays an improtant role in their philosophical work, because in dread meny important themes are being disclosed. Heidegger is interested in onthological field, Kierkegaard puts emphasis on problems of authentic christian existence. There are also other philosophical questions - freedom, authenticity, etc. - disclosed in dread. Both philosophers were able to approach to these problems from an original point of view.

The role of educational counselling at the lower secondary schools

Diploma thesis The role of educational counselling at lower secondary schools and at the lower levels of colleges describes, in its first part, development of educational counselling, its system and valid legislation in the current schooling, defines in entirety the role and duties of members of the school consultancy team with emphasis on work of educational advisor in the sphere of looking after pupils with specific needs. The theoretical part stems out from the vast amount of studies of the latest professional literature. The second part of the diploma thesis states and evaluates results of a research sample amongst adolescent pupils at the lower secondary schools and their educational advisors.

Thesis "Maria Montessori and her teaching method" deals with main principles of the educational system proposed by Maria Montessori, Italian doctor and teacher. Thesis is divided into two parts, a theoretical part and a practical part. The theoretic part aims at fundamental ideas of Maria Montessori's pedagogical conception with the special emphasis on the teacher's role in this method. The practical part focuses on a comparison of a teacher's role in M. Montessori's method and teacher's role in traditional school system.

Phenotypic plasticity of selected species of aquatic insects

Phenotypic plasticity is the ability of the single genotype to pruduce multiple phenotypes in response to evironmental conditions. There are many factors affecting phenotypic plasticity. The aim of this thesis is to summarize the current knowledge of phenotypic plasticity of aquatic insects with emphasis on the role of temperature and food availability. The review is complemented by a laboratory experiments designed to investigate the effect of temperature and food availability on growth and development rate of diving beetle Acilius canaliculatus (Coleoptera: Dytiscidae). The results are discussed in the light of the ecological concepts of temperature-size rule and developmental isomorphy.

Analysis of current situation of German university librarianship with emphasis on digital libraries

Tématem práce je rozbor a hodnocení současného stavu univerzitních knihoven ve Spolkové republice Německo se zvláštním zaměřením na budování, provoz a využívání jejich digitálních knihoven. Přihlédnuto je i ke stávající situaci německého knihovnictví. Úvodní část obsahuje stručné informace o německém knihovnictví jako celku a způsobu řízení a financování knihoven. Specifické místo je věnováno Německé národní knihovně, která hraje významnou roli pro univerzitní knihovnictví. Práce zahrnuje podstatné informace o německém univerzitním knihovnictví, jeho organizaci, financování a stěžejním činnostem. Práce prezentuje výsledky analýzy digitálních knihoven vybraných univerzitních knihoven v SRN. Pozornost je věnována obsahu, struktuře, typologii zastoupených dokumentů a způsobech vyhledávání informací. Zahrnuty jsou také informace o propojování jednotlivých digitálních knihoven a systémech národní i mezinárodní spolupráce, zejména na úrovni disertací. V závěru práce jsou nastíněny perspektivy dalšího rozvoje digitálních knihoven německých univerzitních knihoven a stručné porovnání se situací v České republice.

The Cyprus question in the foreign policy of the United States of America 1960-1974

As a result of World War II the Eastern Mediterranean became one of the most strategic regions in a rising bipolar system. The United States of America realized the geostrategical importance of Greece and Turkey and therefore they provided them with Truman doctrine financial support as a means of preventing infiltration of the Soviet Union to the region. Greece and Turkey assumed their important role in the security system of the West and therefore the United States had an interest in political and economical stability of the area. Nevertheless, from the fifties deep rooted historical tensions between Greece and Turkey intensified. The main reason for this was the Mediterranean island Cyprus, which was a traditional point of contention between both countries. The Cyprus Republic was founded in 1960 as a way of solving problems on the ethnically divided island and leading to pacify Greek-Turkish relations. However, the independence of Cyprus did not calm the situation in the Eastern Mediterranean. Events on the island created significant crises in 1964 and 1974; Greece and Turkey were in both years on the brink of mutual military confrontation. The United States took responsibility for the maintenance of the southeastern flag of NATO and from the end of the fifties started to be interested in the 'Cyprus...

An anarchism and culture. Tradition of anarchism in Czech lands

My dissertation "An Anarchism and Culture, The Tradition of Anarchism in Czech Lands deals with phenomenon of anarchism and its role in intellectual tradition of Western civilization from the time of its inception in the middle of 19th century till the present time. I was trying to advert ideological disunity of anarchist movement by introduction of the main trends within the scope of anarchism. However the emphasis on freedom and tendency to extricate from the chains of dominancy in political, economical and moral ways is common to all anarchist trends. The principal aim of my dissertation was especially to present the tradition role of anarchism in czech lands with description of its evolution with all its inspiration sources and significance for czech society. Above all I was interested in evolution of czech anarchist movement after the fall of communism and its changing in last years. I was trying to create an image of contemporary anarchist movement in Czech republic in terms of content analysis of anarchistic media and documents and from my own experiences too.
