Holmanová, Magdalena ; Urbanová, Kateřina (advisor) ; Kuchař, Jakub (referee)
This bachelor thesis focuses on studying the effect of seasonality on alcohol use. In the theoretical part, the concept of alcohol use is first defined, including its characteristics, specifics, and long-term effects on the human organism. The risks associated with alcohol use are also described. The second chapter is dedicated to seasonality, the history and importance of its research in the context of alcohol use. Attention is also given to the methodology specific to studying seasonality. Finally, the seasonal patterns to which alcohol consumption is subject are presented. In the empirical part, a research proposal is introduced, aiming to determine whether the seasonal pattern of alcohol consumption is also present in the Czech population. The research focuses on the variability of the frequency and quantity of this use, as well as the variability of the frequency of binge drinking throughout the year. Key words: Seasonality, alcohol, alcohol use, Czech Republic
Media archeology of glass plates
Žáková, Aneta ; Hanáková, Veronika (advisor) ; Šlerka, Josef (referee)
This bachelor thesis focuses on glass plates as a photographic medium that was mainly used from the 1840s to 1880s. The goal of this thesis is to define the period discourse and the influence of this media on socio-economic situation through the research method of media archaeology, and therefore to create a complex profile of glass plates as a semi-forgotten medium. At the same time, this thesis answers the question whether this type of medium can be considered obsolete. The methodological part introduces the applied method of media archaeology through its directions, principles and reasons for its use in the context of this bachelor thesis. The research part uses the previously described method to study the artifact of glass plates. It focuses on its characteristic features, as well as historical development, reflection of the medium in society, financial aspects and finally also set questions. This is done by using specialized literature (Scheufler, 1993; Mišková et al., 2016), period documents (The Photographic News for Amateur Photographers..., 1860) and works of media archaeologists (Hodgson, 2006, 2024; Benjamin, 2004, 1996). These sources are further accompanied by information from the institutions Museum Fotoatelier Seidel and Observatoř Kleť. The obtained information indicates that glass...

Jakubcová, Štěpánka ; Matoušková, Petra (advisor) ; Zemanová, Lucie (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Štěpánka Jakubcová Supervisor: doc. Ing. Petra Matoušková, Ph.D. Title of diploma thesis: Plasmid construction for expression of sdr12 reductase from Haemonchus contortus The barber's pole worm (Haemonchus contortus) is a parasitic nematode causing a severe disease known as haemonchosis, affecting livestock, especially sheep and goats. Anthelmintic resistance represents a significant problem in the agricultural sector, with notable economic impacts associated with decreased animal productivity. Short-chain dehydrogenases/reductases (SDRs) are essential enzymes involved in the biotransformation of xenobiotics and may relate to anthelmintic resistance. The theoretical section describes the barber's pole worm, haemonchosis, treatment resistance, and the biotransformation of xenobiotics focusing on SDR enzymes. The experimental section involves the preparation of a vector (plasmid) carrying the protein-coding region of the sdr12 gene to express and isolate the recombinant protein. The results were achieved by using molecular biology methods such as PCR, ligation of the insert to vector, transformation of competent cells, restriction enzyme digestion, DNA sequencing, agarose gel electrophoresis,...
Coordination of assistance to socially excluded localities
Starý, Matěj ; Sládek, Jan (advisor) ; Mudd, Dana (referee)
Excluded localities present a problem in the coexistence of people in society, where one group is segregated from the rest of society because of its low social, economic or cultural status. In the case of an excluded location, this segregation manifests itself in the physical separation of this group from the others and often results in a reproduction of status. The solution to this problem lies in the cooperation between the non-profit sector and the state, which are crucial in the inclusion of excluded localities. This paper describes the cooperation between these two actors as well as the situation of excluded localities and the effectiveness of addressing the issue with a focus on housing exclusion in the context of the city of Prague based on research and interviews with NGO workers in Prague. Key words: excluded locality; non-profit organization; housing need; exclusion from housing; segregation; Prague

The practical part includes a quantitative research method, which is mediated by a questionnaire survey completed by players from selected extra-league and...
Mythical and fairy-tale creatures in Hungarian folk tales
Adamovský, Vít ; Kolmanová, Simona (advisor) ; Mészáros, Andor (referee)
The presented diploma thesis describes the dominant figures of Hungarian pagan mythology, who have organically transformed over time into protagonists of the genres of folk epics, in this case folk tales and fairy tales. The first part of the thesis is devoted to supernatural beings whose origins are to be sought in the imaginations and superstitions of the simple village folk. These were mainly concerned with rudimentary explanations of natural, meteorological and astronomical phenomena or diseases and problems, which were accompanied in the minds of the people by fear of the unknown or the unusual, and which they could not reasonably understand with their then knowledge of the processes of the world. For each of the supernatural figures, a characterization is made on the basis of the ethnographic literature and then their specific occurrence is illustrated with examples from folk tales and fairy tales. The second part deals with historical figures of the pre-Christian period of Hungarian history, around whom a rich narrative tradition has also developed. The additional part of the work consists in creating the likeness of these characters by means of an artificial intelligence-based image generator, since their depictions have not survived due to the absence of written sources and suitable...
