DESIGN OF FIBER-OPTIC BIOSENSOR WITH NEAR-INFRARED SPECTRAL ANALYSIS

This thesis deals with a measurement and interpretation of NIR spectra of bacterial cells and design of biosensor using this analytical technique. In the first chapter, there is introduction of current state of knowledge in the field of NIR spectroscopy in microbiology and technology of fiber optic biosensors. The summary of this chapter shows that NIR is a suitable technique for direct molecular analysis of bacteria, but it suffers from low sensitivity and insufficient interpretation of bacterial spectra. In the next part of the thesis, there is a theoretical background of spectral analysis techniques and technology of fiber optic sensors. In the practical part of this work, there is suggested the elimination of disadvantages of NIR spectroscopy in microbiology by a series of experiments used for interpretation of NIR spectra of bacteria and design of fiber optic sensor to increase sensitivity of this technique. In this work, spectral regions important for the identification of bacterial strains were determined and partially interpreted and the sensor for bacterial analysis capable of classifying strains based on 105 captured cells was designed. Therefore, the objectives of this work were fulfilled.
Design of Oil Pump for T158 Transfer Case

This master’s thesis deals with a new design of an oil pump for Tatra T158 transfer case. The new solution should replace earlier involute internal gear pump which is not able to provide sufficent lubrication. The main problem of the old design is a poor suction at low speeds of the vehicle. Gerotor type of the pump has been chosen. Its main advantages are a convenient specific flow rate and good volumetric efficency. In an analytic calculation of the pump gearing also important functional parameters have been checked, like a pressure angle, flow rate, flow irregularity, specific flow rate, specific slipping. A design with and without axial clearance compensation is introduced. Measurements on a test rig confirmed good suction ability even in low revs. Generated flow rate and pressure are improved. That is a good prerequisite for faster assembly of transfer case with no testing of each oil pump. The tests have not revealed any unexpected wear of parts of the pump so far. It is supposed an application of the new gerotor design in a serial production of T158 truck and other types in the future.

In this work is situated a brief description of containers structure. Part of the work is devoted to handling equipment at container terminals. It describes their properties and the possibility of mounting containers.

This thesis deal with a calculation and a construction concept of a PTO coupling used in Tatra vehicles. The coupling allows turn on an accessory machinery when the engine is on. It is designed as an multiplate clutch with a wet surrounding. The operating is managed by a pneumatic system. The construction solution was restricted mainly by space needs. The calculation includes torque issues and aslo a part considering a heat question. The construction section includes individual clutch parts design.

Předkládá logickou osnovu zohledňující důležité aspekty související s budovami, jejich vnitřním prostředím, bezpečnostními kritérii a možnými riziky, spojenými se způsoby užívání předmětů kulturního dědictví. Metodika si klade za cíl též ozřejmit možnosti vyhodnocování vhodnosti stavebních objektů pro dlouhodobé ukládání předmětů kulturní povahy i ze strany jejich majitelů či provozovatelů tak, aby bylo možné predikovat jejich vliv na uložené fondy a udržitelnost provozu.

The Order of Malta and its activity in the Czech lands during the years 1874-1938

(in English): The Knights of St. John, also known as the "Johanniter", the Knights Hospitaller or the Knights of Malta, are undoubtedly the most important chivalric order of Christendom, which played an important role in the history of Europe. The Order came to the Czech lands in the 12th century and has been continuously active here ever since, although its activities in Czechoslovakia were almost completely suppressed between 1948 and 1990 and the Grand Priory survived only thanks to its members in exile. The present dissertation describes the activities of this Order in the Czech lands between 1874 and 1938. It focuses on the organizational and social aspects of the Order's activity as well as on its economic side. Nor does the priestly branch of the Order stand aside from the interest of this dissertation. The defined period was a time of revolutionary changes - until 1918 the Order could enjoy the support of the Austro-Hungarian elites, including the Emperor, however after the collapse of the monarchy it had to find its place in the new civil society and even fight for its existence. The Order did not see the successful culmination of these efforts in the Czech lands until the late 1930s, but in the light of the coming historical events, these achievements had only a very short life.

Abrégé Cette memoire a pour le but la présentation de la demission et le licenciement dans le droit du travail français et la comparaison de ceux-ci avec le droit du travail tchèque. La partie préliminaire présente en bref le code du travail français et explique aussi le statut du salarié et de l'employeur dans le droit français et mentionne des problèmes qui s'attachent à cette problématique. La partie préliminaire, qui parle du caractère élémentaire de la relation de travail est suivie par la seconde partie, la partie principale, qui disserte sur des conditions dans lesquelles la relation de travail peut être rompue dans le droit français et compare leurs différences avec le droit tchèque. A la fin l'auteur délibère sur ceux différences et leurs avantages et désavantages.
Regulatory mechanisms of CD47 surface expression

CD47 glycoprotein can be found on the surface of all healthy cells in our body. The interaction of CD47 with inhibitory receptor SIRPα on the macrophage leads to the inhibition of phagocytosis. This makes CD47 irreplaceable for the safe recognition of own cells and removal of aged or apoptotic cells. Apart from this, CD47 plays a major role in several essential signalling pathways, such as cell adhesion and motility or calcium homeostasis. The level of CD47 expression and its presence on the cell membrane depends not only on the type of tissue, but also on the age of a cell. An increased expression of CD47 protein has also been observed in the cells undergoing tumorigenic transformation, allowing them to escape from tumour immunosurveillance. Spontaneous regulation of the CD47 gene expression is achieved via regulatory transcription factors, such as NF-κB or HIF-1. Another mechanism of CD47 regulation includes the 3'UTR of CD47 mRNA, which serves as a binding site for either regulatory proteins, such as HuR, or miRNAs. CD47 expression can thus be regulated on both transcriptional, as well as translational level. Water saws and mills in the district of Čadca in the years 1918 - 1950

The presented thesis aims to approach the development of sawmills and mills in the political district of Čadca in the period between 1918 (the establishment of the Czechoslovak Republic) and 1950 (the closure of most companies). The object of the research is primary water-powered devices, but in a broader context also companies with other forms of propulsion. The work describes their technical development, the process of their modernization, technology transfer. At the same time, it examines the impact of natural and economic conditions on these forms of business in the period under review. Finally, it also aims to map the objects of former sawmills and mills. The information obtained in this way is intended to supplement the inputs from archival research, especially in the issue of the technical equipment of individual plants. Source work draws on map materials I. II. and III. military survey and documents stored in the Čadca archive, a branch of the State Archives in Žilina.
"Homo deformis". The fascination by appearance of deformed human body in the Czech lands between 1526-1620

During the 16th century occurred a new phenomen, that the physically disabled people because of they visage were presented as curiosities or miracles. Some cases were so incredible therefore their appearances gave an impulse for creating an artwork. The images were popular between all social levels, were collected by townspeople, scientists, nobles and kings. The dissertation thesis "Homo deformis". The fascination by appearance of deformed human body in the Czech lands between 1526-1620 aims to explore how the phenomen of human curiosities was reflected in early modern visual art in the Czech lands. This dissertation thesis bases on individual examples of human monstrosity in various art forms as an portraits, allegories, scientific illustrations and pamphlets. It exams images of abnormal disabled people between 1526-1620 and focuses on they symbolic meaning, aesthetic interpretation. It summarizes the knowlege about perceptions of depicted abnormal persons in 16th century. It also shows the Early modern society reactions about "homo deformis", their everydayness and social status. This thesis studies reasons why the disabled body was a kind an inspiration for artists.
