Zastoupení mastných kyselin ve vybraných cereálních potravinách
Kučerová, Kateřina
The diploma thesis consists of theoretical and practical part. In the theoretical part it deals with lipids and their role in human nutrition. I also characterized fatty acids and evaluated their health effects. Subsequently, I studied the theoretical basis of chromatographic methods focusing on gas chromatography, its instrumentation and evaluation. In the practical part I analysed the selected assortment of müsli bars by several producers. I focused on the quality of some müsli bars available on the Czech market, focusing mainly on fat content and fatty acid composition. As far as preparation of samples are concerned, it was necessary to lyophilize them first and then Soxhlet extraction followed. The actual determination of fatty acids was carried out using the method of gas chromatography and the results were statistically evaluated.

Kučerová, Kateřina ; Holas, Ondřej (advisor) ; Vraníková, Barbora (referee)
Charles University in Prague, Faculty of Pharmacy in Hradci Králové Department of: Pharmaceutical Technology Supervisor: PharmDr. Ondřej Holas, Ph.D. Consultant: Mgr. Jana Kubačková Student: Kateřina Kučerová Title of thesis: Preparation and evaluation of lipid based nanoparticles as drug carriers Solid lipid nanoparticles (SLN) and nanostructured lipid carriers (NLC) are promising drug delivery systems. Their capability to encapsulate both hydrophilic and lipophilic molecules, biocompatibility and biodegradability of lipids make them a suitable alternative for well-established drug carries. The aim of this thesis was to determine suitable ratios of composition of nanoparticles with acceptable properties (especially reduced size and polydispersity, high zeta potential absolute values), to investigate status and thermodynamic behaviour of the nanoparticles and lipids used and to examine drug encapsulation efficiency. Nanoprecipitation method was used to prepare nanoparticles from stearic acid as a solid lipid and in the case of NLC preparation isopropyl myristate as a liquid lipid was used. Kolliphor® P 188 as a surfactant and Span® 20 as a co-surfactant were the best choice to meet intended characteristics. It was shown that usually lower the concentration of surfactant and co-surfactant was the...

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Analytical Chemistry Candidate: Kateřina Kučerová Supervisor: RNDr. Lenka Kujovská Krčmová, Ph.D. Title of diploma thesis: Development of HPLC method for determination of vancomycine in clinical research The aim of this diploma thesis was to develop optimal conditions for the chromatographic determination of vancomycin for clinical routine practice. The chromatographic separation was carried out by KinetexTM C18 column, 2.6 μm particle size, 50 × 4.6 mm (Phenomenex, USA) in combination with potassium phosphate buffer (pH 4.5) and acetonitrile (90 : 10, v/v) as the mobile phase. As internal standard was chosen cefuroxime. For analytes determination UV detection at 220 nm wavelength was applied. After optimization of separation conditions, the method was applied to biological material. Samples were easily modified using the protein precipitation. The method was partially validated. Keywords: vancomycin, HPLC-UV, plasma samples
Social Rehabilitation as a Tool to Increase the Independence of Persons with Visual Disabilities
KUČEROVÁ, Kateřina
This thesis looks at social rehabilitation courses designed for visually impaired people. Through social rehabilitation, people learn to do various activities which help them achieve independence, self-sufficiency and independence. The study aims to map out the experience of those who have completed a social rehabilitation course and whether it has helped them achieve a more independent life. It will also ascertain the reasons which led them to undertake the course and compare the opinions of clients with the opinions of those who run the courses. To begin with, the theoretical section of the study will describe visual perception, visual impairments and their sensory compensation. It further defines social rehabilitation and describes specific social rehabilitation courses. Trained instructors lead these courses and are in intensive contact with their clients. As such, a related subchapter describes the relations between instructors and clients. Also looked at is communication with people with visual impairments, which is highly specific in many cases. The practical section utilises a qualitative survey based on semi-structured interviews. The research survey comprises two parts, and I focus on the opinions of those who have completed the course in the first part, having spoken to 6 such people, while the second part of the investigation was undertaken amongst the course instructors (teachers). The interviews were undertaken at two organisations providing services to visually impaired people. The research results suggest that completing social rehabilitation courses has helped people increase their independence and thus improve their quality of life. The courses met the expectations of clients. The opinions of clients and instructors concurred to a large extent, with everyone seeing the courses as of great benefit. The deciding factors for beginning the courses were two fundamental needs the need for independent movement and the need to acquire new information, thus to learn something new. The research results can serve anyone who wishes to find out more about the life of people with visual impairments. They will also find out about special courses with which people with visual impairments can look after themselves and their family relatively independently.
