HPLC in quality control of food supplements based on goldenrod and containing rutin and chlorogenic acid

RNDr. Dalibor Šatínský, Ph.D. Title of the diploma thesis: HPLC in quality control of food supplements based on goldenrod and containing rutin and chlorogenic acid In this diploma thesis, a high-performance liquid chromatography (HPLC) method was developed and validated for the separation and determination of chlorogenic acid, 3,5- dicaffeoylquinic acid, rutin, quercetin, and kaempferol. This method was applied to the analysis of food supplements based on goldenrod: drops Celík zlatobýl (Valdemar Grešík), Celík obecný BIO (Bylinné kapky), Celík obecný (Bylinné kapky), Zlatobýl (Green Idea), Zlatobýl - tinktura z pupenů (Serafin), Zlatobýl - tinktura z bylin (Serafin), Zlatobýlová směs (Dědek kořenář), Zlatobýl (Inca Botanica), and capsules Celík (Ex Herbis). Furthermore, two food supplements containing rutin (Solgar and Walmark®) and a food supplement Zelená káva (Botanic) containing chlorogenic acid were analyzed. Personality predictors of exercise addiction in competitive sport

Exercise addiction is becoming a recognized concern, particularly prevalent in elite athletic environments where the pursuit of perfection is emphasized. Despite growing awareness of problematic exercise behaviors, research investigating the underlying risk factors remains limited. This study aimed to examine personality predictors of exercise addiction in Czech elite athletes. A sample of 168 competitive athletes, aged 18 to 30 years, completed assessments including the Exercise Addiction Inventory, Sport Motivation Scale-6, the Athletic Identity Measurement Scale, Performance Perfectionism Scale-Sport, and the Neuroticism Scale from Big Five Inventory. In terms of prevalence, the findings revealed that 16.5 % of athletes exhibited problematic signs of exercise addiction, while 76.7 % were potentially at risk with low severity of symptoms. No significant differences were observed between different groups of sports. Using multiple linear regression, the study demonstrated a significant overall effect of personality predictors on exercise addiction. Identification of "red flag" items in the S-PMV screening questionnaire

The recognition and identification of developmental delays or developmental disorders in early childhood development can be difficult, and various screenings are often used. The Screening for Psychomotor Development ("S-PMV") consists of ten questionnaires and is designed for children between birth and three years of age. Since 2019, it has been recommended by the Ministry of Health of the Slovak Republic as a tool for paediatricians, which allows them to monitor the development of the child and catch any delays in its progress. The aim of the present thesis is to identify the "red flag" items in motor, communication, cognition, adaptive and social behaviour in the S-PMV at each assessed age of the child, which the majority of children (>95%) of a given age fulfil and to distinguish abnormalities in development as a result of it. The research sample consisted of 60,298 completed questionnaires from children aged between birth and three years. A total of 48 "red flag" items were identified in all questionnaires except for the S-PMV 9 questionnaire for children between 11-12 months of age, which could be due to the fact that children at this age have reached different levels of ability in the domains of interest and therefore the items in the questionnaire may have been chosen unsuitably.
The genus Neotinea Rchb.f. in Europe

The thesis deals with the genus Neotinea from the phylogenetic, molecular and ecologic points of view. Most of its species have been classified in the genus Orchis until the use of molecular methods. Within the Orchidoideae, Neotinea forms a sister clade to the genera Anacamptis, Serapias, Ophrys and Himantoglossum. Six species are accepted now. The current taxonomic circumscription calls for a revision under the influence of new knowledge. Especially in N. tridentata, which contains several well-differentiated lineages and is the ancestor of the tetraploid N. commutata. Also the relationship between N. lactea and N. conica remains unresolved. The genome size within the genus ranges from 6.48 pg in N. maculata to 31.14 pg in N. lactea. All species except N. commutata, whose tetraploid origin is debated, are diploid (2n = 42). Partial endoreplication is present in the genus. The range of endoreplicated DNA is relatively narrow regardless of differences in genome size. Species prefer open, slightly basic habitats. In mycorrhizal interactions they appear generalistic with a preference for Ceratobasidiaceae. Electronic legal transactions with a focus on simple electronic signatures Abstract The thesis is divided into six chapters. The first chapter consists of an introduction. The second chapter presents the general regulation and the meaning of the signature with a brief historical background necessary to assess the possible applicability of earlier conclusions to the current legal regulation. The third chapter is devoted to the current regulation and individual types of electronic signatures, i.e. the division into qualified electronic signature, advanced electronic signature and simple electronic signature and the Czech specificity in the form of advanced electronic signature based on a qualified certificate for electronic signatures, commonly known as recognised electronic signature. The chapter also concludes with a discussion of biometric signature, which is increasingly used in practice. The fourth chapter deals with the written form of legal transactions, i.e. a detailed analysis of the Sections 561 and 562 of the Civil Code, including their interrelation. Furthermore, aspects of digital continuity in higher levels of electronic signatures and their comparison with simple electronic signature are discussed. Virus interaction with the cellular DNA damage response

Maintenance of genomic integrity is an essential mechanism for every cell. Genomic integrity is disturbed by diverse exogenous or endogenous effects influencing the cell and causing damage of its DNA. Cellular mechanisms capable of fixing these disturbances in structure or sequence are indispensable because damaged genetic information can later cause expression of damaged proteins or inaccurate segregation of chromosomes to daughter cells. Therefore, many effective mechanisms for fixing wide range of types of DNA damage have evolved. This thesis focuses mainly on the eucaryotic MRN complex, which plays an important role in detection and repair of double strand breaks. Many viral families try to block these cellular repair mechanisms because they are activated soon after viral infection. One of the reasons for their activation is the resemblance of some viral genomes to the cellular DNA with double strand breaks. Thus, in many cases, the cell ends up inhibiting the life cycle of the virus by attempting to repair viral genomes. However, there are viruses that use cellular repair mechanisms for the replication of their genome, making these mechanisms essential for their own growth. The thesis includes the design, synthesis, and biological evaluation of boronic acid derivatives. These derivatives were prepared as hybrid compounds containing an amide linker, combining mainly pyrazinoic acids with 4-aminophenylboronic acid. Pyrazinamide is used as 1st line antituberculosis drug and 4-aminophenylboronic acid is a bioisoster of 4-aminobenzoic acid, which is a crucial precursor in the folate pathway. Bioisosteric replacement of the carboxylic group with boronic acid could afford the ability of the compounds to form a reversible covalent bond toward a potential biological target. The presented compounds were synthesized in a two-step reaction. The first step was the condensation of 4-aminophenylboronic acid pinacol ester with different derivatives of (hetero)aryl carboxylic acids, that underwent previous activation. André Gide, an engaged writer Bc. Martin Poppr The French author André Gide (1869 - 1951) was one of the most important writers of the end of the nineteenth and the first half of the twentieth century and an important figure of the public intellectual life. This thesis analyses to what extent André Gide was a politically and socially engaged author and the reach of his work beyond the world of literature. The method used in this work is a literary critical analysis focused on the author's personality as the key factor for the interpretation of his work. The initial and second part of the thesis summaries the concept of engaged literature developed by Jean-Paul Sartre in his study "Qu'est-ce que la littérature?" and mentions examples of other engaged writers. In this framework, the thesis also deals with the general question of "What is literature?". The third part of the thesis describes the life of André Gide and presents Gide's works which fall under the definition of engaged literature. These include works celebrating human freedom and ethics which attempt to foster the reader's strength and courage, but also Gide's autobiographic works, in which the author himself serves as an honest and liberated role model. Factorization of quaternion and dual quaternion polynomials

In this thesis we provide basic algorithms for factorization of polynomials over quater- nions and dual quaternions. In the case of quaternions, we translate the art of finding roots of given polynomial into factorization. In the case of dual quaternions, we analyse the case of motion polynomials. We show when it is possible to find a factorization of such polynomial into linear terms and give a workaround if it is not. All of this is supplied with geometric interpretations. More specifically, we use the factorizations to construct mechanical linkages. 1
Leaf physiological and optical properties in relation to the Phytophtora × alni infection of black alder (Alnus glutinosa)

A current problem is the infestation of the black alder (Alnus glutinosa) by the pathogen P. × alni, which causes the loss of alder stands and disrupts riparian ecosystems. The spread of the 'phytophthora disease' occurs through water, the host is infected through the root system and the pathogen attacks the conductive tissues. As a result of the disease, the tree gradually loses its root system, lesions appear on the trunk, defoliation occurs and the individual inevitably dies. The only solution to the situation is preventive measures to prevent further spread by careful removal of infected alder trees. The aim of this paper is to discuss the relationship between black alder and P. × alni in the context of the physiological changes that occur in the foliage of the infested individual. Leaf functional traits, chlorophyll fluorescence and optical properties of black alder exposed to P. × alni (inoculation into soil or confirmed occurrence of the pathogen in natural conditions) were measured using non-invasive methods. The results of the chamber experiments suggest that the mean values of the measured parameters do not change significantly in the initial stages of infection, but there is a widening of the variance of the values in infected individuals.
