Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Kateřina Dvořáková Supervisor: prof. RNDr. Lenka Skálová, Ph.D. Title of diploma thesis: The effect of flubendazole on temozolomide treatment in glioblastoma cells Glioblastoma multiforme (GBM) is one of the most common and aggressive brain tumors in adults. Despite significant advances in treatment, GBM remains to have very poor prognosis and median survival of only 12 to 15 months after diagnosis. Standard treatment consists of maximal possible surgical resection followed by chemo-radiotherapy with antitumor drug temozolomide (TMZ), which use is problematic due to fast developing chemoresistance. The aim of this study was to examine the effect of potential anticancer drug, anthelmintic flubendazole (FLU) and effect of FLU in combination with TMZ on GBM cells. For this purpose, two GBM cell lines were used - A172 and T98G. In general FLU reduced cell proliferation more, especially in T98G cells. Moreover, the use of different combinations of TMZ + FLU showed even higher inhibitory effect on the viability of GBM cell lines. The combination of TMZ + FLU reduced the protein level of -tubulin and III-tubulin, simoultaneously interesting changes in STAT3 and EGFR expression, as well as lower...

Dvořáková, Kateřina ; Stančáková, Zuzana (advisor) ; Svěcená, Kateřina (referee)
The main idea of rehabilitation is a multidisciplinary team collaboration. In my bachelor thesis I tried to highlight the team work of the clinical speech therapist and an occupational therapist in people with language disorders. The main objective was to determine how clo-sely these two professionals work and what is important for a person with aphasia. The re-search within the practical part was carry out by a questionnaire that I sent to clinical spe-ech therapists and occupational therapists in various workplaces. Respondents to the ques-tionnaire often mentioned that the work of speech therapist and occupational therapist blends in some situations. However, it does not mean, they do the same tasks. Both disci-pline look at the issue from their own angle, thus ensuring comprehensive care. The result of quantitative research clearly show the fact that in people with impaired communication is cooperation speech therapist and an occupational therapist beneficial, but in many cases is not the ideal level. The end of the thesis highlights the importance of teamwork.

I chart the situation of legislation in the Czech Republic, I deal with circumstances how the Czech courts make a decision about commiting children into parental care after a divorce of their parents and whether the number of children being involved in joint custody increases. I am really interested in the fact whether the courts make a decision based on the child needs. Following this part I focus on the definition of school successfulness of children and the behaviour of younger learners. I indicate the factors that could have an influence on school successfulness. In the practical part I present the results of my qualitative research. I explore the influence of joint custody into child school successfulness from the point of view of class teachers. I survey the behaviour of children who live in joint custody after the divorce of parents and I also take an interest into the fact whether the class teacher have noticed any features that could connect these children.

Teaching in projects is a great way to understand the issue, which can influence pupils' attitudes towards bio-waste management. The aim of this diploma thesis is to create a project on the topic of biodegradable waste, and then to find out how the knowledge and opinions of students about bio-waste have changed. The created project was verified at the lower secondary school, for which it is primarily intended. A total of 54 6th-year pupils was involved in this project. They were given a pretest before the start of the project, finding out the initial knowledge about bio-waste and their opinion on the classification of bio-waste. After the project, the pupils completed a posttest and by comparing the individual answers, it was determined whether and how the knowledge of the tested pupils and their attitude towards the topic changed. The answers of individual pupils in the pretest and posttest were evaluated...
