Restoration of limestone in Grygov

The subject of this bachelor thesis is reconstruction of historical building of the limestone in Grygov. This project is based on the subject AG034 Studio of Architectural Design IV - Restoration of Monuments. The main topic of the studio work was to think about the appropriate use of a building of limestone with the extant circular limekiln and adjacent area of former site of limestone, and to create an architectural study. The thesis include plan to convert building and adjacent land to the cultural-educational facility with camp. Rooms of building is going to be use for various cultural and non-cultural social events. The whole land is going to be recultivated and rearranged for use of citizen. It´s attempt to return to the original values of the building´s appearance. This values were suppress during the years. At the same time It´s preserve essence of its place. The new interventions are solved with respect to native construction. Together them are create harmonic connection between the old historical part and the new timeless part.

The bachelor thesis is focused on the marketing mix of the company Vranovská pláž s.r.o. The theoretical part gives definitions of basic concepts of both the marketing mix and other analyzes that can be used to improve the company's success. The analytical part contains an analysis of the current state of the company and its marketing mix. The last part proposes changes that will improve the current situation of the company and its marketing mix.

Inclusive summer camp and its significance for intact participants

The thesis deals with the operation of an inclusive summer camp, which is attended by intact children and siblings of children with disabilities, in addition to children with autism spectrum disorder, intellectual disabilities and Down syndrome. The thesis mainly deals with the programme during the week-long stay at the camp and also focuses on interviews with the intact participants. The aim of this thesis is to analyse the preparation, implementation and subsequent detailed description of the inclusive summer camp and also to analyse its impact on the child participants. In my thesis I would like to test the hypothesis that children at the inclusive camp in Bukova get everything they expect from it (quality program, friends, fun, safety, trust in leaders, etc.) even if there are children with disabilities who require a lot of care and attention from other adult leaders and assistants. Immunomodulation of dendritic cells by adenylate cyclase toxin from B. pertussis

Adenylate cyclase toxin (CyaA) produced by the causative agent of whooping cough Bordetella pertussis, is a key virulence factor important for colonization of the host. CyaA targets preferentially myeloid phagocytes expressing CD11b/CD18 integrin. By elevating cytosolic cAMP in the host cells, CyaA interferes with their phagocytic, chemotactic and oxidative burst capacities. Furthermore, CyaA modulates the secretion of cytokines and the maturation state in LPS-stimulated dendritic cells (DC) by affecting the expression of costimulatory molecules. In this study, we investigated the effects of CyaA on the capacity of murine bone-marrow DC to prime CD4+ and CD8+ T cells in response to ovalbumin epitopes delivered by the CyaA-AC- toxoid, as a model antigen. Further, we examined the possible impact of CyaA on the antigen uptake and processing for MHC class I and II-restricted presentation by DC, as we previously observed a decreased T cell stimulatory capacity of CyaA-treated DC in response to soluble ovalbumin. We found out that the high levels of cAMP generated by CyaA in LPS-stimulated DC account for the decreased presentation of ovalbumin epitopes carried by CyaA-AC- toxoid on MHC class I and II molecules, thereby impairing the CD8+ and CD4+ T cell responses. This Magister thesis examines the camp phenomenon. Despite the wide scope of the term I will try to come up with a universal definition or several basic definitions which will after serve as a base for my following conclusions. The main source of my thesis is an essay Notes On a Camp written by Susan Sontag. This essay I will apply to contemporary (20th and 21st century) popular music in which I will search for camp signs and campy gestures in the work of independent popular and mainstream artists and performers. The main focus area for this thesis will be primarily visual aspect of their work - costumes, videos, appearance, … The line between campy and not campy is very thin and indefinite, that is why I will try to draw it demonstrating camp signs on chosen samples. I would also try to point out that in contemporary popular culture we consume some forms of camp without being aware that it is actually camp what we consume. Signalization of adenylate cyclase toxin of Bordetella pertussis in macrophages.

Adenylate cyclase toxin (CyaA) is a key virulence factor of Bordetella pertussis, the causative agent of whooping cough. The toxin targets primarily myeloid phagocytes expressing CD11b/CD18 (αMβ2, CR3, Mac-1) and by elevation of cytosolic cAMP levels it paralyses their macropinocytic and opsono-phagocytic functions. Here, we dissected the cAMP-regulated pathway responsible for the block of macrophage macropinocytosis and characterized the capacity of CyaA-treated macrophages to shut- down Akt (protein kinase B, PKB) signaling; that controls nitric oxide (NO) production by macrophages. By using specific activators of protein kinase A (PKA) and for the exchange protein activated by cAMP (Epac), we show that activation of the cAMP effector Epac inhibits macropinocytosis in macrophages. Moreover, upon transfection of macrophages by the constitutively active and dominant negative variants of a downstream effector of Epac, the small GTPase Rap1, inhibition or upregulation of macrophage macropinocytosis was observed, respectively. It was reported previously that the Epac/Rap1 pathway regulates activity of tyrosin phosphatase SHP-1 as well as of protein phosphatase 2 A (PP2A). Role of cAMP signaling in phagocyte migration

Cell migration plays a key role in a wide diversity of biological processes. Migration enables phagocytic cells to localize into the site of inflammation and to lymph nodes, thereby leading to initiation of innate and adaptive immune responses, respectively. The signal transduction that coordinates phagocyte migration consist of diverse signaling proteins, being often under control of 3'-5'-cyclic adenosine monophosphate (cAMP) and its two effectors, protein kinase A (PKA) and Epac (exchange protein activated by cAMP). Small GTPase Rap is activated by Epac and controls phagocyte migration via activation of RAPL and RIAM proteins. On the other hand, PKA regulates cell migration via modulation of activity of other proteins, which comprise actin, integrins, small GTPases Rho, Rac, Cdc42 as well as protein VASP. A prominent feature of cAMP signalization is its spatio-temporal organization. Therefore, besides description of cAMP-regulated signaling cascades in cell migration, this bachelor thesis also depicts how changes of activity of cAMP effectors in time and place are involved in regulation of cell movement.

Role of small effector molecules in bacterial signalling.

Small effector molecules play an important role in bacterial physiology. There are many types of them in the bacterial cell. One group are small signalling molecules which participate in quorum sensing, enabling bacterial cell-cell communication as part of a extracellular signalling. These molecules mediate information about the cell density and different qualities of the extracellular matrix. Next group of small effector molecules are modified nucleotides. They participate in intracellular signalling pathways which regulate the switch of bacterial lifestyle according to changing environment. Best studied are signalling pathways using the molecules - c-di-GMP and (p)ppGpp. Detail studies were done in case of gram positive (Bacillus subtilis) and gram negative (Escherichia coli, Vibrio cholerae) bacteria, where was proved the connection between c-di-GMP signalling pathways and quorum sensing (Vibrio cholerae, Xanthomonas campestris). Important discovery in field of small effector molecules is the c-di-AMP signalling pathway in Bacillus subtilis. New regulatory mechanisms were determined in well-known small effector molecule cAMP, regarding the signalling pathways connections (Vibrio cholerae). A year in scout in photography

The theoretical part of this bachelor thesis is divided into four chapters. The first part consists of a short description of the history of the scout movement, furthermore, it conveys the circumstances of the origin of Scouting and its spreading all over the world. I pay attention to both the world and the Czech environment. The second chapter explains and clarifies the main ideas and values of the scout movement and its three "foundation stones" - the scout promise, law, and motto plus daily order. The third chapter presents some of the Czech Scouting magazines and their characteristics, however, the area of interest is focused only on currently published titles. I diverted from the thesis by adding another chapter about the media image of Scouting based on selected examples to keep the work up to date. The practical part is a book consisting of photographs that I took in the period from August to December 2020 on a summer camp and other scout events. The images are arranged in such a way as to outline the diversity of scout activities and so that readers can get an idea of what a normal day at a scout camp looks like.
