Antiradical activity of Fagopyri herba extracts.

0 Abstract Petra Mikšátková Fagopyrum sp. (Polygonaceae), especially Fagopyrum esculentum Moench (common buckwheat) and Fagopyrum tataricum Gaertn (tartary buckwheat), which are most frequently used, contain some substances affecting living organism. They have an anticarcinogenic and hypotensive effect. They reduce fragility of blood vessels and also affect the progress of atherosclerosis. For their nutritional value they are used as food. Most of these properties are result of an antioxidant activity of a group of substances, flavonoids. Antioxidant activity of Fagopyri herba was evaluated by two methods using the free radical 2,2,-diphenyl-1-picrylhydrazyl (DPPH) and superoxide. By the DPPH assay, the methanol extract and the lyophilized water extract were evaluated. Their IC50 were compared with standards and the antioxidant activity decreased in the order: hyperosid (IC50 = 0,004 mg/ml) ≥ rutin (IC50 = 0,005 mg/ml) › methanol extract (IC50 = 0,105 mg/ml) › lyophilized water extract (IC50 = 0,2945 mg/ml). By the superoxide assay the lyophilized water extract was evaluated. When compared with standards, antioxidant activity decreased in this order: rutin (IC50 = 0,0242 mg/ml) › ascorbic acid (IC50 = 0,0692 mg/ml) › trolox (IC50 = 0,1192 mg/ml) › lyophilized water extract (IC50 = 0,3169 mg/ml).

Influence of salmonella lipopolysachcaride on gene trascription and secretion of interferon gamma.

The study was performed on infectious model of a gnotobiotic pig infected with gram- negative bacteria Salmonella enterica serovar Typhimurium strain LT2 or its rfaL- mutant. The coincidence as a principle

Náhoda jako princip Chance as a Principle ANOTATION The thesis has a focus in questions of chance in the art of 20. century. The crux of the problem concentrate on artists: Jackson Pollock, John Cage, Merce Cunningham and James Joyce. The opening chapter surveys the use of the chance in etymology, science and organic processes of the nature, watches the chance as an inspiration in a game and artistic production. In the second chapter the chance is typologicly defined from spontaneity as a expression of freedom abstract expressionism by Wassily Kandinsky and free atonality by Arnold Schönberg all the way to the kind of chance as a struggle against conventions and to chance as a spring of imagination in authomatic writing and drawing or psychical automatism in surrealism. The main chapter focuses on the key authors of the top phases of spontaneity connected with the action painting (Jackson Pollock), aleatoric music (John Cage), experimental dance (Merce Cunningham) and similar accidental processes in literature (James Joyce) - in exception of Joyce. Online and Offline Vocabulary Correction while using a Digital Pen

This thesis is a requirement for the completion of my double degree with Charles University and University of Nancy (France) under the Erasmus Mundus masters program in Language and Communication Technology. The topic was provided by Prof. Abdel BelaÄd along with Actimage Company and has been completed and successfully defended at University of Nancy under their supervision. After the defense in France, I came here to Prague and worked with my co-supervisor Dr. Václav Hlaváč to improve the text of the thesis to meet the standards of Charles University in order to defend it in Prague. The aim of this work is to study some helpful handwriting correction marks and propose a system that automatically incorporates the corrections made by the writer while writing with a digital pen. The problem is complex because the corrections have to be at the same time readable by the writer and by the machine. Even detecting a free řowing line of handwritten text has not been understood completely by machines. The spatial relations between the correction marks and the text line are easily detected by the writers and readers and hence systematic to them, yet for machines this detection is a huge challenge. Detached House with Design and Architectural Office

The project involves the design and solution of house with office. This is a new family building on the land plot no. 342/26, which is located in Golčův Jeníkov. The building is intended for housing a family of four. The office is used by the owner/owners of the house for their own business plan and entry to it is barrier free. The object has an irregular shape, which is covered in one part by hipped roof and in second part by gable roof using trusses at an angle of 25° with roofing from concrete tiles. The building consists of basement, two floors and no residential attic. Vertical peripheral constructions are made up of brick blocks Porotherm Profi T, which are filled with mineral wool. Inner load-bearing walls, traverses and ceiling are also from the system of Porotherm. The house is plastered with a light blue tint. The windows and doors are plastic with imitation of wood - pin oak. Paved driveway is connected to the local road through ramp in the inclination of 9°. FH has an orientation of rooms to the southeast, south and southwest side. Entry to the building and the office is located on the east side.

The catering customs of the Jihočeská university´s students

SUMMARY: One of the main aims of the humankind is to obtain and preserve health. To achieve this it is important to prevent illnesses; we talk about prevention. Prevention consists of many important factors; such as right nourishment, reasonable physical and mental activities and optimal vital life. That`s why I selected subject for my bachelor work: {\clqq}The catering customs of the Jihoceska university`s students``, which aims at the right nourishment of the undergraduates. The aim of my work was to find out whether the students of the JCU take meals according to the principles of the right nourishment during their studies and if the standard of their nourishment depends on their financial situation. The result from my questionnaire is clear: Students of the JCU don`t take the right principles of the right nourishment. Negative influences have impact on their right nourishment during studies are iregularity of diet, insufficiency of free time and insufficiency of finance. Students of the JCU prefer light pastry and sweets in opposite to fish, pulse and fermented milk products. It is necessary to implement to their menu more healthy food, such as fruit, vegetable, pulse, dark wholemeal pastry, fish and it is also important to keep regularity in drinking minimally 1,5 l of liquid per day. Students of the JCU should have more physical trainings to keep optimal weight and prevent illnesses. Important factors for electing diet are financial together with taste, smell and appearance of the food. Result from the next questionnaire, which I did in France, is that the French students take more right principles of the right nourishment. Their diet is light and better to consume. It consists mainly from vegetable, poultry, pulse and fish. By my opinion it would be better for students of the JCU to take principles of the French students and to import food of French cuisine to their menu.

The Concept of Science According to Paul Karl Feyerabend

The approach to science to Paul KarI Feyerabend follows this approximate model: The pragmatic goal is to find the solution of a problem. Based on a concrete situation, we elaborate on a random hypothesis which can solve. the problem, and because of that, we expand our knowledge. The approach to the choice of hypotheses or theories cannot be restricted by the scientific point of view, nor by scientific method or scientific demarcation. We are left to elaborate, and let alternative hypotheses compete with each other. Feyerabend provided evidence that this is free approach to the extetion of knowledge more effectively defends against rigid exclusivity, and indoctrination of ideology. Reality is more uncomprehensively rich and methodology binds. In this work a view of the world in which we are now living is presented. It is an approach to the world derived from history. It is about Relativism. This view is presented by the life work of the Austrian professor of philosophy Paul KarI Feyerabend.

Effect of milking with an automatic milking system on selected incides of cattle welfare

The aim of my bachelor thesis was to evaluace the welfar of dairy cows. They are loosely housed and milked by robot. The main indicators were total time spent by cows in the robot and information about the movement of cows after leaving the milking box. The next step was to evaluate a time needed to prepare a dairy cow from entrance into the robot to a successful use of teat handpiece. Of course it was necessary to evaluate effects of stress on selected blood parameters. We can give us an idea of welfare using this loosely housing based on these observation. Next reason is the posibility of free decision in which intervals during the day the cows will be milked. Measured and monitored values were collected by observing dairy cows and precise records of their activities from entrance into the robot. In thirty minutes after leaving the robot it was recorded the movement of animals in housing and then satisfying their physiological needs, rating, drinking and lying down. The measurment was realized in standard operating conditions for 24 hours. The measurment was realized by all milked cows in the herd. Time information of ongoing operations and daily milk production were taken from the milking robot Lely Astronaut. Results of these measurements were evaluated by Microsoft Excel, Statistica 9 and then compared with total performance. Information of stress in all ways of milking were getting from the experimental work of VFU Brno. The observations didn´t show any serious problems in welfare area. Animals were calm and satisfied all the time of evaluating. Milking was carried out without any problems and dairy cows entered discipline into the box of robot. Only five cows (6 % of milked cows) had to be chasing with the help of zootechnician. But in the most of cases we talk about heifers and they have to adapt. During the observation it was detected 147 milking at 58 dairy cows. In diametr that was 2,54 milking per one cow. With the average yield 14,87 kg it means, that the average daily performance of one dairy cow was 37,8 kg of milk. The preparation for milking lasted about 2 minutes and 25 seconds. The time of actual milking was average 6 minutes. After milking 83,7 % of the monitored cows sought food and only 6,8 % of monitored cows lay down. The average daily yield significantly increased in the cows, these are milked three times a day, which means 31 % growth. The activity curve of diary cows in the chart showed strong willingness of dairy cows to work with the milking robot. Cortisone in the blood serum showed no values over the limit.

Block of Flats

Diploma thesis describes the design and processing of project documentation of the apartment building. The apartment building is located in the central part of the city of Brno, in the district of Štýřice. The building has 5 floors. Its basementless with flat roof and it is based on the footings of plain concrete. Load-bearing, peripheral walls and partitions are designed from ceramic blocks POROTHERM. Ceiling construction is designed to be assembled of ceramic ceiling fittings MIAKO built on POT supports. Peripheral walls of all floors are insulated with external thermal insulation composite system ETICS with thermal insulation of mineral wool. In the place of the plinth is wool replaced by XPS polystyrene. In the facade there are anchored wooden lattice constructions using ProPasiv anchors. These constructions are covered by façade cladding sheets CEMBRIT METRO and they devide whole building into separate units. There is housing and communal part of the building. On the first floor there are storage areas, the main service room, utility room and two barrier-free apartments. In the second to the fifth floor there are 9 residential units, one of which is designed as duplex apartment . All floors are connected by staircase and wheelchair lift. Parking for 17 cars is designed in front of the building . Two parking spaces are wheelchair accessible.

Formation of biofilm by probiotic bacteria and its processing to solid drug form.

The aim of present work is cultivation of probiotic bacteria L. acidophilus, B. breve and B. longum in such a way that the culture forms cells clusters or comprehensive biofilm on the variety of free carriers. Biofilm formation of L. acidophilus on the silica from point of view bile and acid tolerance in gastrointestinal tract was studied. While the number of living cells in planktonic form (planktonic form) at pH 1 fell by 30 %, the viability of the biofilm cells was maintained to 90 % under the same environmental conditions. The biofilm culture showed also the protection against environment contained bile. Furthermore, the possibilities of drying procedures of biofilm cultures used as commercial technologies in pharmaceutical industry were studied. The comparison of freeze-drying and fluidization bed drying showed, that freeze-drying is more suitable method, which is able to achieve higher amount of viable cells after drying than fluidization bed drying. The effectivity of freeze-drying method is dependent on the selection of suitable cryprotective medium. In this case, about 90 % higher viability after freeze drying was achieved in comparison with fluidization bed drying. Finally, the industrial processing of probiotic strains into the solid dosage form was studied. Tablets should be produced at hardness between 70 and 90 N and water activity of tablet mixture can be maintained below 0.3. Consequently, the drying step of the tablets in a hermetically closed space with at least 10 % of silica gel must be ensured. Thereafter, the tablets contain (5.4 ± 0.7)109 viable cells after 6 months of drying process. Capsule production technology has no significant effect on the cell‘s viability during production. The triplex blistering foil for primary blistering of probiotic capsules was chosen. The triplex foil, which has low values of water vapour transition rate (0.07 g H2O / (m2 × day) and oxygen transition rate (0.01 cm3/m2 × day), was chosen. Other studied blistering foils commonly used in the pharmaceutical industry are not suitable for long storage of solid dosage forms contained probiotics.
