It shows possible use in the treatment of benign prostatic hyperplasia, tumor treatment, plasmodial diseases, and especially cognitive disorders. The isolated structures were subsequently tested for inhibition of human cholinesterases.1 These substances could be used to treat Alzheimer's disease, which has been increasing in recent years. We are not yet able to stop disease, just slow it down. Lack of iron, as well as an excess of iron, can lead to pathological conditions, so it is necessary to control the level of iron in the blood and in case of overloading the body with iron, its chelation can be considered a possible therapeutic tool. Chelators for these therapeutic purposes should be the body's own substance. Amino acids are therefore ideal candidates for the regulation of iron homeostasis in the body, so this thesis focuses on some of them. The individual amino acids are linked by a peptide bond to form proteins. Proteinogenic amino acids are divided into essential, which must be ingested through food, and nonessential, which the body can create on its own. Plant tissue culture of Juniperus virginiana L. as perspective source of podophyllotoxin

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmacognosy Student: Lenka Srbová Supervisor: PharmDr. Marie Kašparová, Ph.D. Title of diploma thesis: Tissue culture of Juniperus virginiana L. as a promising source of podophyllotoxin. The diploma thesis deals with the cultivation of Juniperus virginiana tissue cultures. A growth was observed at Juniperus virginiana two years old culture (variety 'Glauca' and 'Grey Owl') after adding various concentrations of phenylalanine (1 mmol.l-1, 10 mmol.l-1 and 100 mmol.l-1 ) at selected time intervals. The results show that the highest increase in callus fresh weight was detected at Juniperus virginiana variety 'Grey Owl', particularly on the 14th day after adding 10 mmol.l-1 phenylalanine. Suspension culture was successfully derived from the Juniperus virginiana two years old callus culture (variety 'Glauca').
The influence of elicitation on the production of Trifolium pratense L. suspension culture.

Mgr. Kateřina Kyprová The influence of elicitation on the production of Trifolium pratense L. suspension culture. The proces of elicitation makes use of the kapacity of plants and plant cells cultivated in vitro to react to various stress stimuli by a number of protective reactions leading to increased accumulation of secondary metabolites. The paper examined the effect of four concentrations of calcium ions on the production of flavonoids and isoflavonoids by means in the suspension culture of Trifolium pretense L. which was elicited by an abiotic elicitor - 10 µmol solution of lead dichloride. The culture was cultivated on a Gamborg medium with an addition of 2,4-dichlorophenoxyacetic acid and 2 mg.l-1 6- benzylaminopurine. The maximal content of flavonoids found by a photometric determination according to Pharmacopoeia Bohemica 2005 was demonstrated in the suspension culture Trifolium pratense L. (0,355 %) after 6 hour administration of the elicitor and a 1 mmol calcium chloride solution. In comparison with the control and elicited groups, the production was stimulated by 63 % and 25 % respektively. When excess it develops free radicals and structures of organism are harmed. It can lead to organ failure. Excess is solved with chelators, the most used is deferoxamine. But his terapeutic regimen is disadvantageous and it has a lot of side effects. That's reason for searching for substances with better features. This paper focuses on studium of iron-chelating activity of selected alkaloids from family Amaryllidaceae. Concretely, it's galanthamine, undulatine, buphanisine, caranine, 1-O- acetylbulbisine, homolycorine, tazettine, galanthamine, chlidanthine, ambelline, haemanthamine, haemanthidine, hamayne, 9-O-demethylgalanthine and lycoramine. As standard for comparison of activity was used deferoxamine. Activity was measured by spectrophotometer. As indicator was used ferrozine. Alkaloids from Eschscholtzia californica CHAM. and their biological activity The aim of the thesis was to process the alkaline fraction of quaternary alkaloids in the form of their iodides from Eschscholtzia californica CHAM. (Papaveraceae). By means of common chromatographic methods three alkaloids were isolated in pure form and subsequently subjected to structural analysis (MS and NMR studies). The first isolated substance was a tertiary alkaloid allocryptopine, the second one was identified as a quaternary base (-)-7-O-methyl- etherneocaryachine-N-methiodide and the third one was a quaternary base californidine. All the isolated alkaloids have previously been isolated from E. californica. The isolated substances were tested for their inhibitory activity against human erythrocyte acetylcholinesterase (HuAChE) and human plasma butyrylcholinesterase (HuBuChE). Production of secondary metabolites in plant tissue cultures

Production of secondary metabolites in plant tissue cultures The influence of ammonium cerium(IV) nitrate (0.055, 0.55, 5.5, 55 and 275 mg/l of medium) as a potential elicitor of scopoletin production in cell suspension cultures of Angelica archangelica L. was investigated. The cultures were cultivated in a liquid Murashige and Skoog nutrient medium supplemented with 2 mg/l 2,4-dichlorophenoxyacetic acid and 0.4 mg/l benzylaminopurine in the light or dark. The content of scopoletin was determined by high performance liquid chromatography with fluorometric detection. The elicitor treatment improved production of scopoletin. In the dark-grown cultures, the highest amounts of scopoletin in the medium as well as in cells were reached with a concentration of 0.55 mg/l, in comparison with non-elicited culture. In the light-grown cultures, the content of scopoletin was increased only in the medium with an elicitor concentration of 0.055 mg/l.
Abiotic elicitation of explant culture of Trifolium pratense L.

Abiotic elicitation of explant culture of Trifolium pratense L. Summary The objective of this dissertation was to observe the effect of the abiotic elicitors of mercuric chloride and cadmium chloride on the production of flavonoids, and also of the effect of the mercuric chloride elicitor on the production of isoflavonoids in the Trifolium pratense L. explant culture (variety DO−8). The cultures were cultivated in the Gamborg nutrient media with the addition of 2,4−dichlorphenoxyacetic acid (2 mg.l-1 ) and 6−benzylaminopurine (2 mg.l-1 ), in the temperature of 25 řC and during a 16-hours light/8-hours dark period. The quantity of flavonoids was determined spectrophotometrically according to The Czech Pharmacopoeia 2005. The quantity of isoflavonoids was determined by the HPLC method. We can see from the results of this dissertation that the concentration 10 μmol of both of the used abiotic elicitors had the greatest positive effect on the production of flavonoids. The largest quantity of flavonoids (0,576 %) was detected during the elicitation of cadmium chloride of a 10-μmol concentration and after 48−hour application when the quantity of flavonoids increased by statistically significant 294 % in relation to the control sample.
