Možný vliv genových variant v genu COMT na projevy ADHD a/nebo ASD

This work is focused on examining the influence of the COMT gene in two diseases: ADHD (attention deficit hyperactivity disorder) and ASD (autism spectrum disorder). Autism spectrum disorder is a behavioral syndrome characterized by impaired social interaction, impaired communication, and abnormalities in behavior, interests, and play. ADHD is a neurodevelopmental disease that causes problems mainly in the area of concentration, adjustment of activity and excessive impulsivity. Both mentioned diseases can be genetically conditioned, and therefore this work is focused on researching the frequency representation of gene variants responsible for low or high levels of dopamine in people with autism spectrum disorder and with attention deficit hyperactivity disorder compared to the control population. We performed a genetic analysis on two polymorphisms of the COMT gene, namely the more well-known rs4680 and rs4818. The analysis itself included DNA isolation, PCR amplification, control of the PCR product using gel electrophoresis, purification and sequencing of the PCR product and, in the last step, evaluation of the DNA sequence. Based on our results in the laboratory, we did not confirm the hypothesis that the above polymorphisms in the COMT gene represent a risk factor for ASD and ADHD. Our results were probably influenced by the small number of people in the monitored groups, but even this work helped in a small fragment to understand the cause of the complicated pathology of these diseases.
Pedagogical assistant in a school club and children with behavioral disorders

The main research objective of this thesis is to find out how pedagogical assistants and educators in after-school clubs think about educating children with behavioural disorders and behavioural problems. The first chapter attempts to distinguish between behavioural disorders and behavioural problems, defines ethopedics and behavioural disorders, their etiology, and diagnostics. The second chapter deals with inclusive education for students with special needs and defines the position of teaching assistants in terms of Czech legislation. The third chapter presents qualitative research consisting of four phenomenological interviews. The research aim is to explore and understand the experience of pedagogical assistants and educators in after-school clubs educating children with behavioural disorders and behavioural problems. The findings of the research are compared with the theory presented in the professional literature, and, in conclusion, recommendations for educational practice are suggested.
Life quality of people with attention deficit

This thesis focuses on the issue of quality of life of an individual with Attention Deficit Hyperactivity Disorder (ADHD), with particular attention paid to the impacts of ADHD on important areas of life that significantly affect the quality of life. In addition, the thesis also deals with values and goals and subjective perception of the above syndrome of individuals diagnosed and living with ADHD. The answers to those questions were obtained through semi-structured interviews with five selected respondents and a questionnaire survey focusing on SEIQoL method as a tool to evaluate the quality of life. The results of the research show that Attention Deficit Hyperactivity Disorder interferes with daily functioning in life and influences the quality of life. The most important value in life was identified to be the area of interpersonal relations in the sense of mutual support and mainstay. The respondents have mentioned positive dimension of ADHD syndrome that can be considered as benefit in life. It is very important to consider each person as unique individual who should be treated with respect. Moreover, it is also important to take the symptoms of ADHD into consideration and not make light of them, as trivialization is what negatively affects human relationships, and it may even result in a distortion of one´s relationship with oneself.
Use of Equine Assisted Therapy with children suffering from behavioral disorders and ADHD

Therapeutic horseback riding is one of the most popular and the most used form of animal based therapy. The term "therapeutic horseback riding" itself is very broad and it consists of four basic branches. The most used, yet least described, is the branch that has been called "pedagogical-psychological horseback riding therapy" since 2011. This branch is further divided into two sub-branches, Equine assisted psychotherapy and Horse-based activities. These two sub-branches overlap each other and are usually conducted together. With school-age children they are mostly used in cases of behavioral disorders and ADHD. The goal of the thesis is to evaluate the effectiveness of the methods with children diagnosed as such. The research was realized in "Stredisko hyporehabilitace Pirueta" which operates within the Mental Hospital in Havlickuv Brod. The method of the research was observation of five subjects within the one complete therapy unit. At the beginning and at the end of the unit the tutors of the subjects were given observational scale, which served as supplementary technique for diagnostics. Data obtained from own observation and the data gained from observational scaled matched each other approximately in 90%. Education of pupils suffering from ADHD in elementary school

Thesis consist of theoretical part and empirical part. In the theoretical part of the thesis the author will look into the characteristics of the ADHD syndrome and its subtypes. It will also focus on other issues related to ADHD, such as specific learning disabilities. The author will define the concept of ADHD, including its etiology. The theoretical part will also contain a part that will be devoted to diagnostics, treatment and medication of pupils with ADHD. This part will describe the specific manifestations of ADHD syndrome in each developement stage of a child. There will also be a part dedicated to special education needs and support measures that could be applied to an inclusive approach in education for pupils with ADHD. The research part will use a qualitative research method, including a case study, interview and observation. In this part of the work, the author will focus on two case studies of students with ADHD, supplemented by an interview with the mother of twins. The author intended to include an interview with teachers, but due to GDPR rules, teachers refused to provide one. Comparison of reaction rate in children diagnosed with ADHD and common population of the same age

Title: Differences in reaction rate in children with ADHD and the general population of the same age. Problem definition: ADHD is a neuropsychological disorder occurring in part of the pediatric population (5-8%). The scientific public mentions the existence of the so-called hyperfocus, ie the ability to significantly concentrate in children diagnosed with ADHD, especially in the case of these children being interested in the task. Methodologically correctly compiled and performed testing of the reaction rate of these children could confirm or refute this assumption. Aims: The aim of the work is to find out whether and possibly what effect the diagnosis of ADHD in children has on the reaction rate. Hypotheses: H1: The reaction rate in individuals with ADHD will be on average the same or higher than in individuals of the same age of the healthy population. H2: The difference between the individual experiments will be greater in children with ADHD than in individuals of the same age of the healthy population. H3: The individual with fastest reaction ability with ADHD will have the same or better result as the individual with the fastest reaction of the same aged healthy population.
