Methodology for preparing data from digital libraries for use in digital humanities

This methodology aims to offer libraries and other memory institutions in the Czech Republic a recommended procedure for making large volumes of data available for research purposes. Currently, from this point of view, a more than critical amount of documents from library collections are digitized, while the results of digitization are presented in various digital library systems. When making them available, it is always necessary to proceed from the current version of the copyright law, but it is already possible to prepare for its significant amendment, which implements Directive 2019/790 of the European Parliament and the Council and concerns, among other things, the extraction of texts and data for scientific purposes. The architecture of the newly developed system for digital libraries recommended by the methodology will ensure scalability, easy management and the development of related services. The presented methods of data processing, their enrichment and output formats are based on the requirements of specialists from the entire range of humanities.

Males-females differences in the spectrum of chromosomal aberrations in the group of nanocomposites production workers

An increase in the use of nanomaterials (NM) has been witnessed in many areas of human life. Therefore, assessment of genotoxicity of NM and nanoparticles (NP) is one of the main objectives of genetic toxicology. Despite this fact, human cytogenetic studies following the exposure to NP are still rare. Moreover, no relevant information on possible differences in sensitivity to NP related to gender is available.\n\nIn this study we periodically (in September 2016, 2017 and 2018; pre-shift and post-shift each year) analyzed a group of workers (both genders), working long time in nanocomposites research, and matched controls. Aerosol exposure monitoring of particulate matter including nano-sized fractions was carried out during working shift. Micronucleus assay using Human Pan Centromeric probes, was applied to distinguish, besides the frequency of total MN in binucleated cells (BNC), also other types of chromosomal damage (losses and breaks). Moreover, whole-chromosome painting (WCP) for autosome #1 and both gonosomes (X and Y) were applied in third sampling period (2018) with the aim to identify the particular structural and numerical chromosomal aberrations.\n\nObtained results showed: (i) differences in the risk of exposure to NP related to individual working processes (welding, smelting and machining); (ii) differences in chemical composition of nano-fraction; (iii) no effect of chronic exposure of NP (total MN) opposite to significant effect of acute exposure; (iv) gender-related DNA damage differences (females seem to be more sensitive to chromosomal losses). Additional data from WCP suggested increased frequency of numerical aberrations in gonosomes.

Methodology for assessing the effect of sublethal doses of pesticides on solitary bees\nemploying the OMICs approach

Pollinators are potentially exposed to a number of pesticides in the environment. The\nmost important group of hazardous substances comes from the category of plant protection\nproducts (PPPs), which are applied not only by farmers but also by small gardeners. Each PPP\nincluding the active substance and formulation undergoes a very detailed and thorough evaluation,\nwhich is in the European Union (EU) based on Regulation (EC) No 1107/2009 of the European\nParliament and of the Council. Although the negative effects of PPPs on the environment and nontarget organisms are minimized with proper use, we cannot rule out possible hidden effects,\nespecially those of a sublethal nature. Modern analytical approaches, such as the application of\nhigh-throughput methods, help to identify hidden side effects. This methodology is focused on the\nevaluation of the effect of sublethal doses of pesticides on solitary bees employing the OMICs\napproach. In the risk assessment of pesticides, the honey bee has long been preferred as a model\nspecies of pollinator, while solitary bees stay in the background. The methodology contains\ninnovative aspects for experiments, such as a modified isolator, preparation of plants for exposure,\nthe timing of the experiment, monitoring of active substance distribution or proteomic analysis of\nsolitary bees to identify hidden effects of the test substance or its formulation. In this methodology,\nthe model of the solitary bee, red mason bee (Osmia bicornis), is considered. However, the\nmethodology is also applicable to other, especially related species of solitary bees. The\nmethodology can be used in areas of state administration, private laboratories and research\nactivities in the assessment of environmental risks of pesticides on solitary bees. The\nmethodological procedure can confirm or even eliminate environmental risks when registering new\nproducts or reevaluating existing products. This methodology therefore has the potential for use in\ntesting new substances intended for plant protection before their registration. In an exemplary\nembodiment, the effect of acetamiprid in a Careo sticks formulation was tested. The distribution of\nacetamiprid in the soil substrate and the plant was evaluated. The hazardous metabolite\nacetamiprid IM-2-1 was identified in plant tissues. However, proteomic analysis indicated a very\nlow risk of acetamiprid for O. bicornis. This result is in agreement with the fact that acetamiprid\nhas been assessed by EFSA to be a low risk for bees and is registered in the EU until 28 February\n2033.

Final Report of the Project „Data Acquisition from the Deep Horizons of the Rožná Mine“ presents the results of the study of rock mass properties with respect to the presence of a significant tectonic zone. The study focused on the change of properties in the vicinity of the so-called first zone of the Rožná deposit, but also on the change of these properties with a depth of 12–24. floor of the former mine, 550-1200 m below the surface. Realized works included geological, petrographic, geochemical description of the rock environment, geophysical work (esp. seismics), study of physico-mechanical properties of rocks, transport experiments and deposit revalidation of the Rožná deposit. All work was carried out for the purpose of describing the studied zone and its manifestation with an emphasis on ensuring the safety of the potential location of the deep geological repository.

This report summarizes the data available from the Čertovka site and evaluates the site according to criteria derived by RAWRA staff from SÚJB requirements (Decree No. 378/2016 Coll. On the Location of Nuclear Equipment) and IAEA (NS-R-3 rev.1 Site Evaluation for Nuclear IAEA 2016, SSG-14 Geological Disposal Facilities for Radioactive Waste, IAEA 2011b) on site characteristics that have an impact on long-term DGR security (MP22 v3, Vokál et al. 2017).
