PharmDr. Radim Kučera, Ph.D. Title: Use of HPLC-HRMS for the study of tacrine group compounds potentially useful in the therapy of Alzhemier's disease. High-performance liquid chromatography coupled with high-resolution mass spectrometry (HPLC-HRMS) is an analytical technique very often used in the new drugs development. This technique is widely used, from the confirmation of the proposed structure and identification of present impurities, through the assessment of absorption, distribution, metabolism and excretion to the monitoring of poisoning and intoxications. In the theoretical part, the presented dissertation thesis is focused on the basic characteristics of Alzheimer's disease, its currently available treatment and new treatment strategies. In addition, attention is paid to areas closely related to the experimental part, namely: the study of pharmacokinetics, analytical tools for determining pharmacokinetic parameters, liquid chromatography, mass spectrometry and the specifics of the bioanalytical methods. PharmDr. Radim Kučera, Ph.D. Název: Chirální separace klastrových sloučenin boru Anorganické trojrozměrné klastrové sloučeniny boru se staly předmětem zájmu vědců zejména díky svým zajímavým vlastnostem, např. výjimečné tepelné, biologické a chemické stabilitě, neobvyklé třístředodové dvouelektronové vazbě a trojrozměrné aromaticitě. Podtřídou boranových klastrů jsou karborany, které mají v kleci inkorporován alespoň jeden atom uhlíku, čímž kombinují vlastnosti anorganických a organických látek v jedné molekule. Atom uhlíku umožňuje modifikaci klastru různými organickými substituenty, což vede k existenci nepřeberného množství derivátů. Z tisíců těchto sloučenin byly důkladně zkoumány deriváty aniontového bis(dikarbolidu) kobaltu ([Co(C2B9H11)2]- ), a to zejména ve farmaceutické chemii, chemii materiálů, elektrochemii a analytické chemii. Zvýšený zájem o [Co(C2B9H11)2]- je obvykle vysvětlován jeho rozpustností ve vodě, chemickou a biologickou stabilitou, nízkou toxicitou, skrytým amfifilním charakterem a podobností s chemií ferrocenu. Martin Ferenčík Title of diploma thesis: LC-MS monitoring of selected antimicrobial compounds in waters The monitoring of residues of human and veterinary antibiotics in surface water is gaining more and more importance due to the increasing resistance of bacteria. This work deals with the modificate of an HPLC-MS/MS method to obtain pilot data on the occurrence of antibiotics in surfaces water. The method is focused on the analysis of 30 antibiotics from the groups of β-lactams, macrolides, lincosamides, amphenicols, fluoroquinolones, tetracyclines and sulfonamides. The method was performed on a UHPLC instrument Agilent 1200 Infinity series with MS Agilent model 6495 triple Quad. Separation of antibiotics was performed on reverse phase using gradient elution and tandem mass spectrometry. An electrospray in both positive and negative polarity was used to ionize the analytes. The method was used for the pilot analysis of surface water samples in the Czech Republic. Samples from 15 rivers and streams were analysed. Anthropological reflection of Woman in the Second Half of the 19th Century

The diploma thesis deals with the transformation of self-intimate feminity in the second half of the 19th century. The primary subject matter of the thesis focuses on the transformation of the female subjectivity in the contrast with the prejudiced and culturally influenced role of a female. The thesis focuses on the biological determination of a female from the masculine perspective in the discourse of the 19th century and on the influence of socializing factors on the change of understanding of the female subjectivity. The primary subject is the role of a mother as a non conceptprovider of specific information about the matrimonial cohabitation. The methodology of the thesis concerns historical, culturally - anthropological, sociological survey and the gender and a culturally and socially created construct. The framework is: the biological determination of feminity, female psyche, juvenescence, and enlightenment of church, female readiness for matrimony, bachelor mother. Molecular probes are mainly used to quantify multiplication in RT-PCR and identify mutations in PCR analyses. There are typically two different types of probes - mono-labeled molecular probes and double-labeled molecular probes. The mono-labeled molecular probes are constructed from fluorophore- labeled oligodeoxynucleotide sequence or quencher-labeled oligodeoxynucleotide sequence. Whereas double-labeled molecular probes consist of an oligodeoxynucleotide sequence with a fluorophore attached at one end together with a quencher attached at the other end of the chain. Amphetamines (especially methamphetamine) belong among the most used hard drugs in the Czech Republic. Amphetamines and their structurally similar substances synthetic cathinones are psychostimulants and have a stimulating effect on the central nervous system. They increase the level of dopamine, norepinephrine, and serotonin at the synapses through their reduced reuptake. Drug use during pregnancy does not harm only the mother, but also the fetus and can lead to abnormalities that may occur malformations of fetal organs, delayed fetal development, low birth weight of the newborn, premature birth, or risk of hypoxia. There is also a risk that a newborn will have neonatal abstinence syndrome (NAS). Prenatal drug exposure not only risks complications during pregnancy and childbirth, but also later consequences. Ondřej Horáček Title of Thesis: Separation of molecular probes by capillary electrophoresis Oligonucleotides are organic molecules which find their application in fields such as biotechnology, molecular biology, diagnosis and therapy. For the use of oligonucleotides (ON's) as molecular probes, it is necessary to covalently modify the ON's for example with a fluorescent dye and a quenching moiety. The products of this labelling reactions are the required double labelled ON probe, but also side products like the unlabeled ON and the ON's labeled either with the fluorescent dye or the quencher. Consequently, the products of this reaction must be separated, which represents the focus of this diploma thesis. The separation of these double labeled ON probes has been so far only performed by chromatographic approaches. Ondřej Horáček Title of thesis: The influence of experimental conditions on chiral separation of borane clusters in capillary electrophoresis Keywords: chiral separation, borane clusters, capillary electrophoresis, cyclodextrins Abstract: Boron compounds also include borane clusters, in which boron atoms form triangular configuration and 30 such sides are presented as a so-called cage. These clusters can be synthetically modified into various forms (open or closed) and can be substitued, for example by carbon atoms. This substitution can lead to the asymmetry of these molecules and thus to their chirality. Currently, their potential is also being studied in medicinal chemistry, where they could contribute to the design of new drugs.
