The molecular mechanism of CSL protein participation in oxidative stress response in Schizosaccharomyces pombe

Redox homeostasis maintenance is important for proper organism and cell function, for while relatively low amount of reactive oxygen (and nitrogen) species contributes to the fine tuning of signal transduction, excessive concentration of ROS (oxidative stress) has demonstrably harmful effects and is tightly connected to many pathological states. Cells therefore evolved broad palette of antioxidant mechanisms that express striking level of conservation among different species. Large, intricate stress response signaling networks have been already described; nonetheless, novel molecules employed in stress-related signaling are still being discovered. Several studies recently suggested transcription factors CSL, proteins essential for regulation of metazoan development as effectors of Notch signaling, are also involved in response to oxidative stress. The fission yeast Schizosaccharomyces pombe, well established model of response to various stresses, comprises two paralogs of CSL proteins - Cbf11 and Cbf12. We have found cells depleted of cbf11 are highly resistant to hydrogen peroxide. This resistance appears to be caused by upregulation of important stress responsive genes including ctt1, gst2, pyp2, and atf1. Cbf11 is therefore negative regulator of these genes, which suppresses their expression...

Surgical treatment of diabetic macular edema

Diabetic macular edema is the most common cause of visual impairment in diabetic patients. Precise pathophysiology of diabetic macular edema is unclear and seems to be multifactorial and includes pericyte loss, microaneurysm formation, basement membrane thickening and focal closure of the capillary bed, vitreomacular traction, and ultimately breakdown of the blood-retinal barrier with increased vascular permeability. Risk factors for clinical significant diabetic macular edema are hyperglycemia, hypertension, hyperlipidemia, duration of diabetes, and pregnancy. The increasing number of individuals with diabetes worldwide suggests that diabetic macular edema will continue to be major contributors to vision loss and associated functional impairment in the working-age population of most developed countries. Although eyes with diffuse macular edema carry a particularly poor prognosis despite laser photocoagulation, laser treatment is still the first choice of treatment for diabetic macular edema as it is safety and less invasive that other surgical options. Left ventricle remodeling in patients with primary aldosteronism and essential hypertension

Myocardial damage is one of the most serious consequences of arterial hypertension. Changes in the heart structure and function develop not only due to pressure overload itself, but many other hemodynamic and neurohumoral factors contribute to their formation. Our work has compared echocardiohraphic strucutural anf functional changes of the left ventricle, caused by essential hypertension and hypertension associated with primary aldosteronism (PA) as the most common reason for secondary hypertension. The first part of our work focused on the differences in left ventricle geometry in men with PA and essential hypertension after separating it's low-renin form (where, similarly to PA, the plasma volume expansion was considered to have the dominant effect on left ventricle remodelation). In men with low-renin forms of hypertension including PA, we observed greater both endsystolic and enddiastolic diameter of the left ventricle, lower relative wall thickness and more frequent eccentric type of hypertrophy when compared to essential hypertensives with normal renin levels. Whereas left ventricle cavity diameters were positively correlated to aldosterone levels, wall thicknesses were associated mainly with hypertension severity expressed as an average 24hour blood pressure and number of antihypertensives....

The Rise of Protest Parties in Southern Europe

This thesis analyzes the rise of populist parties in southern Europe - italian Five Star Movement, greek SYRIZA and spanish Podemos. The aim is to define the key factors, that contributed to the perception of these protest political formations as political alternatives. The thesis chronologically follows development of examined political parties. Further it defines their strategy in terms of levels of protests and programme. Finally it outlines probable transformations of party systems, which is caused by the rise of protest parties.

The Communication Strategy of Sládkova limonáda

This diploma thesis is focused on the communication strategy of the Staropramen brand Sládkova limonáda. The thesis is aimed on the description of the marketing strategy of the brand with main focus on communication strategy. The emphasis is placed on current trends affecting the market and the strategies of companies presented on the market. The major goal is to create a communication strategy plan for Staropramen Sládkova limonáda. The communication strategy is based on theoretical knowledge, information and data collected through a questionnaire survey, situational analysis and subsequent evaluation of the strategy of launching the brand on the Czech market.

The usage of algae in biogas transformation

Using of algae in a biogas transformation is still in the beginning. However, the microalgae have large potential from the perspective of growing demands on biogas quality and trend of using,natural resources. First of all, it is their ability to fix carbon dioxide (CO2) using photosynthesis and presumed ability of some algae to metabolize hydrogen sulphide (H2S). Biogas contains not only required methane but also components causing its worse quality such as mentioned CO2 and H2S. Therefore, the algae are potential biological systems for biogas-conditioning. The microalga Chlorella pyrenoidosa Chick (IPPAS C2) was used for fixation of CO2 and H2S in our experiment. The microalgae were cultivated in a medium BG 11. The algal suspension was aerated with the biogas during two weeks. Different values of CO2 concentration measured in the input and output confirmed decrease of CO2 caused by intensive growth of algal culture. Decline of H2S was not confirmed.

Art therapy with children with learning disabilities

This thesis describes art therapeutic work with children suffering from learning disabilities (LDs) and Attention Deficit Hyperactivity Disorder (ADHD). The theoretical part of this thesis is based on specialized literature and focuses on the definition of learning disabilities and ADHD syndrome. Furthermore, possible causes of these disorders and also their consequences on the personality and life of an affected child are being discussed. In this theoretical part I also describe a care program for these children and include application of art therapy. I also focus on specific features at art works of children suffering from LDs and ADHD. In the practical part I present two case interpretation studies where art therapeutic procedures used in work with affected children are being described. Furthermore I describe art work progress of two boys. Here I focus on specific points connected with their disabilities and impact of these disorders on their personalities.

Effect of Expansion of Modern Information Technologies on the Aviation Industry

This diploma thesis is focused on modern information technologies and its direct effect on the aviation industry. Comparing the state of the industry without technologies and nowadays; describing changes in consumer behaviour as well as in the matter of providing services by companies; pointing out that these changes were caused mainly by growing number of Internet and mobile technologies users all over the world. The last chapter is dedicated to projection of future trends regarding aforementioned area.

Analysis on status of CSR in the Czech Republic

This master´s thesis is focused on analysis of state of CSR in the Czech Republic. The theoretical part covers the basic concept of CSR associated with its development, benefits, methods of its measurement and reporting including current trends in the field. The practical part is based on author´s own research conducted among national and multinational companies operating on the Czech market using quantitative and qualitative research methods. The research´s results reflect the differences between these companies following up the individual aspects of CSR and other variables. Therefore the thesis portrays the evaluation of the current state of CSR in the Czech Republic, including related causes and recommendations for improvement.
