Herman Joseph Tyl
Feranc, Daniel ; Royt, Jan (advisor) ; Šebek, Jaroslav (referee) ; Petráček, Tomáš (referee)
The work called Herman Joseph Tyl offers a total lack on the life of this important religious figure, the abbot of one of the oldest Norbertine monastery founded by Czech nobleman blessed Hroznata in Teplá, which experienced the suffering of two totalitarian regimes of the 20th century: Nazis in concentration camps and prisons in Communist labor and uranium camps, as well as various interventions in the persecution of his life in the form of withdrawal of government approval. The work is given to the period of his life, which has so far lacked the processing. This is particularly Tylova childhood, adolescence, studies and life after 1958, when he worked as a warehouseman at Cakovském collective farm and returned to the priesthood, which he re-intervention prevented Communist totalitarian power by removing state approval, to the end of his life again to priesthood returned and was even elected abbot of the monastery Tepla. The work also offers a historical and critical evaluation of the data known from the Tyl autobiography affecting in particular the period from his arrest by the Gestapo in 1942 until his release from prison and their communist completion in the light sources. Processing of the dissertation was based on archival sources collected relating to the person of Herman Joseph Tyl: his...

Role of endocytosis and endosomal acidification in TRAIL-induced apoptosis
Hradilová, Naďa ; Anděra, Ladislav (advisor) ; Kovář, Jan (referee)
TRAIL (TNF-related apoptosis inducing ligand) became known for its ability to selectively eliminate cancer cells. This ligand is a member of the TNF (tumor necrosis factor) ligands family and triggers extrinsic apoptotic pathway by binding of its death receptor 4 or 5 (DR4/5), and subsequent formation of death-inducing signalling complex (DISC). This signalling complex is required for successful transmission of apoptotic signal and activation of proximal caspases. However, regulation of the initial steps leading to activation of caspases is still not fully understood. Endocytosis of a TRAIL- DR4/5-DISC complex can be one of modulators of the initiation of extrinsic apoptotic pathway. Recent studies show controversial data documenting that endocytosis of TRAIL receptosomes can in cell type specific manner either positively or negatively influence TRAIL-induced apoptotic signalling. In this study, we focus on the analysis of a role of endocytosis and acidification of endosomal compartments during TRAIL-induced apoptosis in human colorectal cancer cell lines. Our results support the view that both clathrin-dependent endocytosis of TRAIL receptosome and endosomal acidification positively affect activation of caspases during the early stages of TRAIL-induced apoptosis. Inhibition of endocytosis or endosomal...

The Czech economic transformation process compared to Hungary and Poland
Perková, Martina ; Loužek, Marek (advisor) ; Vlček, Josef (referee)
This bachelor writting is focused on analysis and comparison of economic transformation process of three central european countries. These selected countries - Czech republic, Hungary and Poland - have comparable starting conditions. The aim of the thesis is not only a general description of theoretical difficulties connected with the transformation process in the economical realm (and partially also in the political and institucional realm), but also to discover similarities or dissimilarities during the development of these economics. The analysis and comparison is performed mainly with the best known and the most used macroeconomic indicators. The conclusion includes an over-all summary of the transformation process up to now.

Prescription of Antibiotics and Antibacterial Chemotherapeutics of Selected Doctors in the Vysocina Region
Barochová, Jana ; Vlček, Jiří (advisor) ; Matoulková, Petra (referee)
Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Department of Social and Clinical Pharmacy Candidate Mgr. Jana Barochová Consultant Prof. RNDr. Jiří Vlček, CSc. Title of Thesis Prescription of antibiotics and antibacterial chemotherapeutics of selected doctors in the Vysocina Region This work concerns the problems of prescription of antibiotics in the out-patient care. It analyzes the possibility to use pharmaceutical database for monitoring and evaluation of medical prescription at regional level. It is well known that the portion of antibiotics in the total prescription in Czech Republic is higher than would be optimal and therefore methods for minimization of excessive prescription of antibiotics are now intensively being searched. In this work we focused on the evaluation of prescription of antibiotics during years 1995 to 2008 for 35 general practitioners for adults and for 20 general practitioners for children. The data for prescription analysis was taken from the pharmaceutical database Medico, from 15 pharmacies in Vysocina region. For evaluation of the prescription patterns we used so called prescribing indicators. We have found out that during years 1995 to 2008 the drugs of the first choice in the treatment was gradually substituted by the alternative...

Characterisation of potassium cation transport systems in the yeast Zygosaccharomyces rouxii
Stříbný, Jiří ; Hodek, Petr (advisor) ; Schierová, Michaela (referee)
Characterization of potassium cation transport systems in the yeast Zygosaccharomyces rouxii Potassium has become absolutely necessary cation for living cells, including yeasts, because it plays several important roles in physiological processes. Intracellular concentration of K+ in yeasts is usually between 200 mM and 300 mM, while the external K+ concentration is ranging from molar to micromolar. To adapt to environments with low K+ content, yeast cells employ various K+ high-affinity uptake systems, e.g. Trk, Hak and K+ -ATPase, that provide cells with the sufficient amount of potassium. The recent release of the complete sequence of the osmotolerant yeast Zygosaccharomyces rouxii genome allowed us to search homologues of the known yeast potassium uptake systems. We have found just one gene encoding a putative potassium transporter homologous to the S. cerevisie TRK1. For the characterisation of transport properties and physiological roles of the product of this gene, named ZrTRK1, three approaches have been used. First, the IT tools serve to analyse sequence characteristics, phylogenetic relationships etc. The second approach involves cloning of the gene and its expression in a S. cerevisiae mutant strain lacking its own two Trk systems, characterisation of transformants' growth phenotypes and...

Segmental structure of Mandarin syllable
Třísková, Hana ; Heřmanová, Zdenka (advisor) ; Volín, Jan (referee) ; Uher, David (referee)
This dissertation concerns itself with the segmental syllable of Mandarin, i.e., of standard Chinese as codified in P.R.C. and known as putonghua. While it leaves aside the aspect of tone it explores all major topics related to the phonological and phonetic description of the isolated Mandarin segmental syllable and the ways in which they are treated in broad literature, both Chinese and Western. We approach the subject from the perspective of teaching Chinese as a second language (TCSL), where mastering the syllabary (cca 400 segmental syllables, 1300 tonic syllables) represents a crucial basic step. Poor command of the pronunciation of isolated syllables will inevitably have a detrimental effect on the spoken language competence of a student. Because the pinyin romanization system (developed in P.R.C., officially approved in 1958) assumedly is the only feasible phonological system for the paedagogical purposes, the present dissertation does not aspire to establish any new phonological interpretation of the Mandarin syllable (although some attempts will be made). After making a broad comparison of existing analyses and their pros and cons, we strive to explore the solutions most convenient for the purposes of teaching pronunciation.Along with accepting pinyin as a phonological background, we advocate for a...

Palaeoecology, sedimetology and biostratigraphy of the Chýnice limestone at Čeřinka near Bubovice
Ferrová, Lenka ; Frýda, Jiří (advisor) ; Budil, Petr (referee)
The diploma thesis presented deals with paleoecological, biostratigraphical and sedimentological research in the most important locality of the Chýnice Limestone - Čeřinka near Bubovice village. The research was carried out during the years 2006-2010. Multidisciplinary approach has been applied during the fieldwork as well as in the subsequent laboratory research. Such an approach brought a lot of valuable knowledge. The contact between the Zlíchov and Chýnice Limestone has been described and characterized for the first time within this thesis in this locality using sedimentological, biostratigrafphical and geochemical data. Among others, microfacies analysis has been carried out and the sedimentary succession the of Chýnice Limestone has been described and sedimentary environment has been interpreted. During my research I was able to gather the largest collection of fossils with the exact stratigraphic position, which was ever collected within the Chýnice Limestone (2500 pcs). Fifty new taxa in total have been recorded from the Čeřinka locality during my research, representing almost 80% increase of taxa known from this classic locality. Numerical analysis of the fauna diversity development of the Chýnice Limestone revealed an increase of the total diversity in time and in the same time decrease of...

Industrial melanism in the context of ultraviolet reflectance
Stella, David ; Reif, Jiří (referee) ; Kleisner, Karel (advisor)
This work focuses on the topic known as industrial melanism. The main aim of this thesis is to summarize the knowledge published on this topic. The first mentions about industrial melanism appeared in England in the end of 19th century. Many different hypotheses emerged during the last one hundred years. The work starts with introduction of the fundamental theory of industrial melanism in the context of environmental change during last three hundred years. Furthermore, the historical development of ideas on industrial melanism is given and contemporary hypotheses are discussed. Significantly, work deals with the biological role of UV reflectance in the context of industrial melanism.

Flow-shop Problems
Dyntar, Tomáš ; Tegze, Miron (advisor) ; Červinka, Michal (referee)
A description of flow shop and lot streaming including an example is given. Methods and algorithms ofr solving various modifications of the single job two-machine flow shop transfer lot sizing problem are shown. It starts with the calsulation of two lots (discrete), three lots (continuous) problem and then Trietsch s polynomial algorithm or an arbitrary number of sublots is introduced. This alogithm is then used for solving modifications with finding the proper number of sublots for given makespan, with constant or linear transfer times including variants with limited number of transfer machines or their limited capacity, with constant or linear set up times. A domed with both transfer and setup times is then developed and Trietschs algorithm modified. A theoreme by R. G. Vickson for solving multiple job lot streaming problem by treating the lot streaming problem for each job separately and then using Johnsons alogorithm is proposed to extend the results of the single job cases to those of multiple jobs. A known way to extend results to the three machine problem by treating each pair of successive machines separately or developing Trietschs algorithm for three machines is shown. Most of the procedures are illustrated in examples.
