The study aims on effects of mercury on haematological and biochemical parameters in blood plasma on common carp (Cyprinus carpio L.). The theoretical part of study is focused on phenomenon of occurrence of mercury and its forms in aquatic environment and fish. Practical portion of study includes the effects of administration to mercury and also the effects of co-administration of sodium chloride. In the first experiment we carried out the exposure of Cyprinus carpio L. by selected mercury concentrations for 5, 10 and 15 days. In the second experiments were carps exposed to 1.5 micro g.l-1 mercury with the addition of various concentrations of sodium chloride (29 mg.l-1, 300 mg.l-1 and 1000 mg.l-1) for 72 h. For the evaluation of haematological and biochemical biomarkers in blood plasma of carps, we exploited the Konelab T 20xt and Easy Lyte machines. The obtained data were statistically processed and evaluated.

Phylogeography and dispersal routes of the Proasellus coxalis s.l. (Crustacea: Isopoda)

Dispersal of freshwater invertebrates is affected by many factors. Recently, human activities played a very important role in their dispersal. Among others, constructions of new shipping canals damages natural boundaries and the species can spread into new areas. This Master thesis deal with non-indigenous species P. coxalis s.l. On the genetic bases assess a possible dispersal routes into the Central Europe (and the Czech Republic), and phylogeography of this species in the Czech Republic. I amplified and sequenced DNA of the mitochondrial cytochrome oxidase subunit I (COI) gene and 28S nuclear rDNA gene. 96 individuals from 27 localities in the Czech Republic were analyzed. I also included 40 sequences of the COI gene and 7 of the 28S gene from the GenBank database. In the Czech Republic, there are two different genealogic lineages of P. coxalis (with 6% genetic distance between sequences of the COI gene). The first lineage comes from recent invasion of this species from Mediterranean area (through Western invasion corridor and other shipping canals). The second lineage is probably indigenous in the area and it can probably be assigned to the subspecies P. coxalis septentrionalis. The results are discussed. This work deals with study of thiol compounds (metallothionein) and the determination of its quantity in the tissues of rainbow trout (Oncorhynchus mykiss). This tissues of the fish were exposed to several concentrations of arsenic. Concentration of metallothionein was measured elektrochemically by using differential pulse voltametry (DPV) by using Brdička's reaction. Metallothionein, which is among the low molecular weight thiols has very strong detoxication effects. Determining of the concentration of this protein could be used to determine the quality and also the health of fish meat.
Molecular biodiversity inventory of the ichthyofauna of the Czech Republic - presentation of the started project

The study based on international cooperation would like to contribute in a significant way to identification and description of the species diversity of the complete Czech ichthyofauna using a comprehensive approach. The inventory and subsequent cataloguing concentrate on recent native and non-native species of fishes and lampreys living in natural waters of CR. The obtained results are used for inter-continental comparison of all fish species using the "DNA barcode" within the BoLD platform. The final summary evaluation will contribute to updating of the data for monitoring within the system Natura 2000 and will provide vital information and recommendations to the ANCLP CR. The new system of species collections of type specimens designed on the basis of a comprehensive approach and creation of a detailed voucher will mean significant contribution to the Czech museums.
