Significance of the S1 subsite of rhomboid intramembrane proteases for catalysis and inhibitor design

This thesis focuses on the development of specific inhibitors of rhomboid intramembrane proteases. These inhibitors are needed for the cell-biological investigation of rhomboid proteases and their potential pharmacological targeting, as rhomboid proteases have been associated with various diseases, such as malaria, Parkinson's disease, cancer or toxoplasmosis. The thesis advisor's laboratory has recently discovered the first such group of compounds, the peptidyl ketoamides. To exploit them fully, it is necessary to examine their properties and the possibilities of their modifications. In this work, synthetic fluorogenic substrates and enzyme kinetics were used to examine the possibilities of exploiting the S1 subsite in the rhomboid active site for rhomboid inhibitor design. Furthermore, using variants of these substrates modified by unnatural amino acids in the P1 position, the mechanism of water transfer to the rhomboid active site was investigated. Comparison of cleavage rates of ten fluorogenic substrates modified in the P1 position revealed that the E. coli rhomboid protease GlpG strongly prefers side chains -CH2-CH3 and -CH3 in the P1 position (i.e. binding into the S1 subsite). Keeping records about nursing care at medical department

The thesis is focused on the characteristics, purpose and principles of medical documentation. The theoretical part describes the partial activities of keeping records of the nursing process in practice. The practical part is divided into two chapters. The content of the first part is collection of informations about possibilities of keeping the documentation from 1976 to 2010. The second part deals with qualitative research in the form of ethnographic research. For the collection of data, was chosen the method of direct observation of the nurses from the department of neurology at the regional hospital. The attention was focused on measuring the time consuming of administrative tasks. The aim of the thesis was to find out the time-consuming task of keeping the documentation in the individual activities, what part of the working time is devoted to the administration. Role of expression of cellular prion protein in the differentiation of neuronal cell lines

Cellular prion protein (PrPC ) is a membrane bound glycoprotein. The protein is expressed in all vertebrates, mainly in the nervous system, but it is present also in the cells of gastrointestinal tract, bone marrow, germ cells and heart. PrPC is necessary for pathogenesis of prion diseases, which are deadly and without the possibility of therapy. The pathogenic isoform of prion protein is formed by changing of secondary structure of PrPC and it's the main constituent of infectious prion particles. Pathological form of prion protein accumulates in brain of infected patients and this process is associated with neurodegradation. Physiological function of PrPC is poorly understood. Knock-out of the PrPC gene (PRNP) is not connected with any noticeable phenotype. Potential functions of PrPC are dispersed, protein may have antiapoptotic effect, it can be involved in ions metabolism or in protection against oxidative stress. Latest results show, that PrPC can play important role in cell differentiation. During the differentiation PrPC can influence the development of cells and their typing. Evaluation of ecological education in Ecocentre Vlašim

The main topic of this thesis is the CUNC basic organization unit and Podblanické Ecocentre CUNC in Vlašim. The first part of the thesis presents the Czech Union for Nature Conservation (CUNC) followed by an overview of its Environmental Centres. The next section contains a summary of all the activities carried out by the CUNC basic organization unit and Podblanické Ecocentre CUNC in Vlašim from their establishment to the present and recapitulates the formation of these organisations themselves. The thesis also covers a characteristic of all the one-day as well as residential teaching programmes for secondary schools, technical schools and grammar schools carried out by the Ecocentre. The greatest emphasis is laid on four selected one-day teaching programmes, for which a research was conducted by using a questionnaire method. Its aim was to determine the influence on pupils and students of the educational programmes from secondary schools, technical schools and grammar schools.

1 ABSTRACT Kučerová J.: Biological activity of secondary plants metabolites VI. Alkaloids of Vinca minor L. Diploma thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of pharmaceutical botany and ecology, Hradec Králové 2016, 67 pages. Isolation of alkaloids was done from extract of Vinca minor, family Apocynaceae. Individual substances obtained were examined for their ability to inhibit human acetylcholinesterase and butyrylcholinesterase. The basic extract was after an initial extraction with 95% ethanol pre-purified by extraction with chloroform. This chloroform extract was divided further by column chromatography on a total of 531 fractions. The fractions obtained were, based on the similarities identified by thin layer chromatography, interconnected in a final number of 17 fractions. The mobile phase for column chromatography were mixtures of chloroform and medical gasoline, chloroform and ethanol, and ethanol itself. For further testing were used fractions number 2 and 5. From the fraction 2 was by preparative thin layer chromatography and subsequent crystallization isolated substance number 1. Role of prion proteins at differentiating cells

Cellular prion protein (PrPC) is well known for its pathological isoform PrPSc, widely believed to be the infectious agent of the prion diseases, which include Bovine spongiform encephalopathy (BSE), scrapie and Creutzfeldt-Jakob disease. The physiological role of PrPC is poorly understood, but its involvement in the regulation of apoptosis, adhesion molecules, antioxidant, or signal molecules, has been described. Despite of these findings, it hasn't been proven, that the protein is necessary for normal development of mice. However, the protein was shown to be essential for regeneration of hematopoietic stem cells after exposure to lethal stress conditions. Expression of PrPC may have an effect on the proliferation and differentiation of cells by helping them keep the proliferative activity, or slow spontaneous differentiation. The quantity of the protein correlates positively or negatively with expression of transcription factors such as Oct4/Nestin, which are essential for development in embryogenesis. Its expression also regulates transition of cells from G1 phase to S phase of the cell cycle. This bachelor thesis is focused on published results describing the influence of PrPC on cellular proliferation and differentiation.
The role of the mediation in pre-divorce and post-divorce care in the Czech Republic

The dissertation thesis "The role of mediation in pre-divorce and post-divorce care in the Czech Republic" is about identification possibilities that the mediation brings to families, husbands and wives, and partners with solving difficult life situations like a divorce or a break up between partners. After it describes the impact of these interpersonal conflicts. Theoretical base of this thesis is: family crisis theory, individualized society theory, theory of gender inequality, but for all concept of harmonizing working and family life and changes on labor market. Main source of information was qualitative research of mediation clients and interviews with experts. In my thesis I analyze causes and consequences of conflicts in a partner life and through mediation I suggest a solution of consequences in the incurred situation, which seems like desired in wide society context.

Legal regulation of advertising of medicinal products in the Czech Republic Abstract This thesis deals with the advertising of medicinal products for human use and its development and legislation. The introductory part is set the objectives of this thesis and the reasons for choosing this topic. The first chapter is devoted to advertising, its legislation, the definition of advertising, history of advertising and this chapter also highlights the institution supervising advertising. The second chapter is to set the legal regulation of advertising in the Czech Republic. Regulation can be divided into the regulation of advertising public and private, together are in this dividing developed individual laws regarding advertising and is not omitted non-legal regulation of advertising.
