Buněčný prionový protein PrPC je membránově vázaný glykoprotein, který se běžně vyskytuje u obratlovců, především v nervové soustavě, ale také například v buňkách gastrointestinálního traktu, v kostní dřeni, v pohlavních buňkách či v srdci. PrPC je zároveň nezbytným pro patogenezi prionových chorob, smrtelných onemocnění bez možnosti léčby. Patogenní izoforma PrPSc , vznikající změnou sekundární struktury PrPC , je základním stavebním kamenem infekční prionové částice, jenž se akumuluje v mozku postižených jedinců a tento děj je spojen s neurodegenerací. Samotná fyziologická úloha PrPC v organizmu však nebyla dosud objasněna, při vyřazení genu pro PrPC (PRNP), nebyl pozorován fenotypový rozdíl. Potencionální funkce PrPC jsou spojovány s anti-apoptotickými účinky, metabolismem iontů či ochranou před oxidačním stresem. Výsledky z poslední doby současně naznačují, že by PrPC mohl hrát důležitou roli v buněčné diferenciaci. V průběhu diferenciace by protein mohl mít vliv na vývoj a typizaci buněk, ovlivňovat průběh buněčného cyklu a podílet se na utváření nervové soustavy. Cílem této práce je zmapování míry dopadu snížení exprese PrPC na buněčnou diferenciaci a vlivu prionové infekce na diferenciaci neuronální buněčné linie (CAD 5), která je schopná propagovat priony. V rámci této práce byly porovnávány linie... Cellular prion protein (PrPC ) is a membrane bound glycoprotein. The protein is expressed in all vertebrates, mainly in the nervous system, but it is present also in the cells of gastrointestinal tract, bone marrow, germ cells and heart. PrPC is necessary for pathogenesis of prion diseases, which are deadly and without the possibility of therapy. The pathogenic isoform of prion protein is formed by changing of secondary structure of PrPC and it's the main constituent of infectious prion particles. Pathological form of prion protein accumulates in brain of infected patients and this process is associated with neurodegradation. Physiological function of PrPC is poorly understood. Knock-out of the PrPC gene (PRNP) is not connected with any noticeable phenotype. Potential functions of PrPC are dispersed, protein may have antiapoptotic effect, it can be involved in ions metabolism or in protection against oxidative stress. Latest results show, that PrPC can play important role in cell differentiation. During the differentiation PrPC can influence the development of cells and their typing. It could affect cell cycle and have an influence on formation of nervous system.
