Kryoprezervace a transplantace spermatogonií kapra obecného

Cryopreservation and transplantation of germ cells in fish provides a suitable tool for preserving genetic information. By method of surrogate reproduction, the offspring with characters of the chosen donor can be obtained. In this case of our commercially important species common carp. However, for the successful cryopreservation of the germ cells, a suitable protocol for each species must be established. Several cryoprotectants were tested. The best of them, Me2SO, regarding the viability of spermatogonia, was tested for its different concentrations depending also on the rate of freezing. Further testing, related to the effect of tissue size, incubation time and added sugar, was performed. The result of the assay identified best cryomedium composed of 2.5M dimethylsulfoxide, added sugar of 0.3M glucose, 1.5% BSA and 25nM Hepes dissolved in PBS. The most suitable size of tissue was 100 mg, incubation time was 30 min and coolig rate was -1 ° C/min. This protocol ensures the highest viability rate of cryopreserved spermatogonia of common carp. The second part of the work was to verify the success of the transplantation of cryopreserved and fresh spermatogonia into a suitably chosen recipient, the goldfish, which shares similar reproductive characteristics with carp, but also offers reduction of space requirements or resistance to koi-herpes virus. The transplanted germ cells colonized the germ line and started gametogenesis in 42.5% (cryopreserved spermatogonia) and 52.5% (fresh spermatogonia) goldfish recipients, which demonstrated that the transplantation of cryopreserved spermatogonia of common carp can be successfully achieved.
The eye of a poet: Antique figures in the work of Jean Cocteau

The thesis is concerned with the instances of the figures of mythology of the antiquity in the works of Jean Cocteau and their application. It focuses on the character of Narcissus, which is examined in connection to Oedipus and Orpheus, while concentrating mainly on the topic of mirroring and the related motives of the eye and sight. This mirroring is then not examined only on the level of an aesthetic object, but also in relation to the recipient and in mutual interferences between various media. The thesis is methodologically based on selected philosophical theses by Maurice Merlau-Ponty and the theoretical concepts of Pierre Brunel, Mieke Bal and Mary Ann Caws. Within the works of Jean Cocteau the thesis is primarily concerned with the analysis of the novels The Holy Terrors (Les Enfants Terribles), Le Livre Blanc and their illustrations, the poem "Tombeau de Narcisse", the play La Machine infernale and the films Blood of a Poet, Orpheus and The Testament of Orpheus.

This work consists of three parts: the translation theory of drama and the specifics of school theatre, the translation of a play by David Llorente, Los árboles dormidos, and a commentary discussing the translation. The theoretical part focuses on problems and characteristics of translations of this genre. The commentary to the translation includes a translation analysis, a discussion of the translation problems and their solutions, and a typology of translation shifts. Viewers and other recipients are in the center of attention of all media platforms. The environment is constantly developing and consumers of media content are developing with it. They are much more involved in the process of creation of content, which they now not only change and edit, but create as well. The borderline between author and public is gradually fading. Technological convergence, which gives the public new opportunities, creates a new type of recipient. Prosumer, mix between producer and consumer, defines the directions of media activity all around the world. Public has decided to take advantage of the opportunities and power of internet and developed from simple consumers to full-bodied creators of content.

The impact assessment of water treatment plants on water quality in the recipient

Watercourses influenced by anthropogenic activity increases in recent years and streams lose their natural character. This situation is critical particularly in small streams with minimal flow rates. The present work deals with the assessment of streams affected by discharges of treated wastewater from small waste water treatment plants. The results showed that in case of average flow in the recipient none of the monitored waste water treatment plants do not negatively affect the stream. Problems can occur when snow melts, when the lower temperature of the flowing water in the wastewater treatment plants negatively affect biological cleaning processes.
Art Brut as a question of Aesthetics

This bachelor thesis deals with the phenomenon called art brut that brings interesting aesthetical questions related to the status of art piece, author or perceiver. The first part of the work presents the insight into history and origin of this specific artistic expression, as well it follows up its forms and significance. Every subject is based on basic theoretical sources. The second part focuses on matter of author and authorship as the relevant question of aesthetics and also focuses on significance and importance of author´s pole within art brut´s creation. This significance is searched in key esseys of art theory which analyse author as an separated entity of the aesthetic situation.
Pragmatic and Stylistic Analysis of the Horoscopes in Periodicals

The thesis entitled "Pragmatic and stylistic analysis of the horoscopes in periodicals" deals with the horoscopes from linguistic point of view; the greatest attention is paied to the pragmatic and stylistic aspects. The goal of the thesis is to identify the typical features of horoscopes on the basis of pragmatic and stylistic analysis. The research material comprises of 1752 astrological predictions taken from sixteen different periodicals. The author and the recipient of horoscope are characterized in each chapter and selected attributes of the target group are studied in each chapter as well - age, gender, social roles and status. It is dealt with the intention of horoscopes; its impact on recipient and strategies used by authors of horoscopes are analyzed too. The thesis presents the way vocabulary and other language tools are used in horoscopes to communicate intention of their authors. The analysis also deals with themes of horoscopes.
Media marketing strategy analysis of Prima Cool

Aim of this thesis is to map how they can live on their own media carrier marketing strategy and media promotions. Through case study - Prima Cool TV - I dissert on the monitoring of its anchorage in the media environment through their own television channel, but also the cooperating contact channels created by the proprietor of the station. The thesis presents couple of theoretical insights from marketing and media strategy, particularly in the application for the media. It analyzes in detail the concept of strategy and position in the marketing, various forms of strategy - thinking and management. It elaborates forms of strategic management, monitors media strategy, as a form of marketing and the usefulness of the media (media providers) in their own media in particular. To obtain an objective view on the subject work brings several known methods. Expert theories are confronted with (by) the Prima Cool TV case study. In this tracked subject the thesis analyzes the usefulness of its own media space for marketing and media strategies. The analysis subject is the TV audience, program structure, communication with recipients and fostering awareness of the TV station in the Czech media market. Above all it confronts theoretical knowledge to specific steps of FTV Prima, in the frame of examined issue....
