The effect of Goeckerman therapy on selected immune markers in patients with psoriasis
Kondělková, Kateřina ; Andrýs, Ctirad (advisor) ; Posová, Helena (referee)
1 Summary The effect of Goeckerman therapy on selected immune markers in patients with psoriasis Psoriasis is a systemic inflammatory disorder with skin manifestation. Classical approach to treat psoriasis is Goeckerman therapy (GT) which is based on daily application of pharmaceutical coal tar on affected skin with subsequent exposition to UV light. The aim of this study was to observe selected immunological markers and cell subpopulations in patients with psoriasis and to elucidate the effect of GT on these parameters. We evaluated T regulatory cells (Treg), membrane CD163 expression on monocytes, serum levels of CD163 (sCD163), membrane TLR2 and TLR4 expression on monocytes and serum levels of TLR2 (sTLR2). The immunofluorescent analysis was performed on erythrocyte lysed peripheral blood using a flow cytometer FC500 Cytomics (Beckman Coulter) and FACSCalibur (Becton Dickinson). The level of soluble markers was detected by sandwich enzyme-liked immunosorbent assay (ELISA). There was no significant difference in the relative number of Treg cells in the peripheral blood of healthy blood donors and patients with psoriasis before initiation of GT. The relative number of Treg is significantly increased in psoriatic patients after GT compared to the pre-treatment level. In comparison with levels in controls...

Švidrnochová, Michaela ; Novotná, Eva (advisor) ; Krátký, Martin (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Mgr. Michaela Švidrnochová Supervisor: RNDr. Eva Novotná, Ph.D. Title of Thesis: Influence of benzoxazinedione derivatives on selected mycobacterial enzymes Tuberculosis is an insidious infectious disease with more and more increasing incidence in certain regions. It is difficult to treat by the available means because of increasing cases of multidrug resistance, which is defined as a resistance of M. tuberculosis at least to two primary antituberculotic drugs - rifampicin and isoniazid. That's why is necessary to search for new substances which interfere with the metabolic pathways of Mycobakterium tuberculosis. Enzyme inhibitions in different levels of these pathways are considered to be a target of the new potencial antituberculotic drugs. In this work, influence of monosulphur and disulphur derivatives of 3-(4-alkylphenyl)-2H- 1,3-benzoxazine-2,4(3H)-dione on enoyl-ACP-reductase, dihydrofolate reductase, isocitrate lyase and FtsZ was tested by thermal shift assay, spectrofotometric method and light scattering. The results showed that the dihydrofolate reductase is not a drug target of selected inhibitors. Isocitrate lyase was the most inhibited by sulphur derivative 3-(4-terc-...
The impact of selected sesquiterpenes on the activity of antioxidant enzymes in the differentiated Caco-2 cell line
Verešová, Erika ; Svobodová, Hana (advisor) ; Szotáková, Barbora (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Erika Verešová Supervisor: PharmDr. Hana Bártíková, Ph.D. Title of thesis: The impact of selected sesquiterpenes on the activity of antioxidant enzymes in the differentiated Caco-2 cell line The cases of cancer diagnosis rapidly continue to grow across the population. It is a modern trend to buy more and more often a variety of herbal food supplements in order to strengthen or restore human health. Many scientists have been researching Myrica rubra plant for its potential therapeutic effects. In our research we used sesquiterpenes isolated from this plant, in order to determine their effects on the activity levels of antioxidant enzymes in a differentiated Caco-2 cell line derived from intestinal epithelium tumor that have maintained characteristics of healthy cells. Caco-2 cells are being used as a basis for testing of substances and their effects on non-cancerous cells. The cells were incubated for 24 and 48 hours with a concentration 25 µg/ml of tested substances. Essential oil from Myrica rubra and selected individual sesquiterpenes (α- humulene, β-caryophyllene and trans-nerolidol) were chosen to be tested. Of the antioxidant enzymes, we determined the enzymes glutathione-S-transferase,...

Exiled Periodical Nový život [New Life] (1949-2001): Introductory Study and Bibliography
Benáková, Marie ; Novotný, Vojtěch (referee)
The core of the licentiate thesis The Exile Periodical "Nový život" [New Life] (1949-2001): Introductory Study and Full Bibliography consists of a bibliography of all fifty three volumes of the Czech exile periodical, which had been issued in London originally as "Věstník Cyrilometodějské ligy akademické" [Journal of the Academic Association of SS Cyril and Methodius] and from 1950 as "Nový život" [New Life]. From 1954 the journal was taken over by the "Křesťanská akademie" [Christian Academy] in Rome. The bibliography is complemented by an introductory study, which summarizes the current state of research, clarifies the identification of "Nový život" [New Life] and presents its content and authors (the institutional background, editors, administrative assistants, and authors). The chapter about the institutional background describes the first years of existence of the Academic Association of SS Cyril and Methodius on the basis of the articles from "Nový život" [New Life] and comes with more precise facts in comparison to previous works.
European Federations and European Union
Uhlova, Diana
188 Summary This submitted dissertation thesis provides a constitutional comparative analysis of the European Union and the European federations, primarily the Federal Republic of Germany, Swiss Confederation, Republic of Austria and also basic features of the United States of America. I will attempt to determine whether EU is already a federation, or how much closer it has shifted to a federative type political arrangement. In this dissertation thesis I will try to methodically analyze, which characteristics of a federal state the European Union already meets and if we can put the EU under the category of federations, confederations or international organizations. To achieve better understanding of the context of this dissertation thesis, a comparative analysis of the European Federations is studied from historical, sociological and political science perspective. However the main emphasis here is focused on the analysis of the constitutional and legal arrangements. The first part of this thesis starts from the scrutiny of the origin and development of the theory of federalism in historical perspective, followed by determining the concept of federalism and federation in constitutional terms. Subsequently provided here are definitions of various types of federations and the reasons for their creation and...
