Trees Have the Power
Mikušková, Nina ; Fajnor, Richard (referee) ; Kovářová, Marta (advisor)
The project "Trees Have The Power" is being developed since 2013 and revolves around the strength of interpersonal connections experienced throughout our lives. It points out the reality, that we all create our own micro-cosmos around us, shaped and expanded by every new person we meet. First phase of the project involved exhibition of a tree within a gallery, which served as a metaphor of an ever expanding organism. For the second phase an online social platform was developed, to map out and connect my personal contacts. The third outcome offers space to present the "tree". By involving people from my own personal tree of connections, we are creating lectures, interactive events, concerts, film screenings, and discussions focused on multiple aspects of life. The aim is both to test the skills and initiative of a specific community as a driving force in sharing and accumulating knowledge and experience; and at the same time to demonstrate the magnitude of impact of such activity. This unorthodox framework of encounter offers opportunity to generate new perspectives within the community, and enrich the audience.

Study on pretreatment and hydrolysis of selected lignocellulose materials
Kovářová, Markéta ; Dvořák, Miloš (referee) ; Obruča, Stanislav (advisor)
This diploma thesis is focused on study of chemical and enzymatical hydrolysis of raw wood material. The aim of this work was to find the suitable method for pretreatment of selected lignocellulose materials. The theoretical part deals with characterization of lignocellulosic material and its components. There are also subscribed various pretreatment methods and their effect on hydrolysis of sawdust. In experimental part of the work the most appropriate approach of pretreatment and hydrolysis of sawdust was studied. Criteria for the selection of suitable method was concentration of saccharides as desired product of hydrolysis and also concentration of the most important microbial inhibitors - polyphenols. Application of 96% ethanol or 5% H2O2 were identified as the most promising pretreatment methods which enhanced yields of fermentable sugars about 30 %. Further, we also performed cultivation of bacteria Burkholderia cepacia and bacteria Burkholderia sacchari using solution obtained by hydrolysis of lignocellulose material.
Enzymatic hydrolysis of wastes associated with coffee production
Kovářová, Markéta ; Skoumalová, Petra (referee) ; Obruča, Stanislav (advisor)
This bachelor thesis is focused on study of potential production of extracellular hydrolytic enzymes by microorganisms – bacterium and moulds, which have been cultivated on spent coffee grounds. The theoretical part deals with characterization of coffee and utilization of coffee by-products. There are also subscribed microorganisms and enzymes which have been noticed. In experimental part coffee ground was used as the sole substrate for production of extracellular hydrolytic enzymes. Productions of protease, cellulase, mannase and lipase enzymes were observed. None-identified isolate of mould spontaneously contaminating spent coffee grounds was identified as the best producer of these enzymes. Subsequently the conditions of cultivation such as water content and shaking vs. static cultivation of this moulds were optimized. Further, we performed partial purification and pre-concentration of the enzyme cocktail by ultrafiltration, ultradialysis and PAGE-SDS characterization of extracellular enzymes was performed as well.
Cable-supported bridge
Kovářová, Marika ; Bezručová, Kristína (referee) ; Nečas, Radim (advisor)
The theme of the thesis is to design a pedestrian bridge over a river. Out of two possible designs, the cable-stayed construction was chosen. The deck of the bridge is designed as a monolithic construction supported by cables from two inclined pylons. Two models were developed for the calculation of internal forces: a strut model for the longitudinal direction and a plate model for the perpendicular direction. These calculations were checked up by Finite Element Method (FEM) in SCIA Engineer 2017 software. Considerable part of the thesis was dedicated to the creation of so-called initial state. The design of the prestressing force and the evaluation of ultimate limit state and serviceability limit state was made in accordance with the codes currently in force. Small part of the theses is dedicated to the verification of the load bearing capacity of the pylon on the combination of normal force and bending moment with a subsequent verification in the software IDEA StatiCa9. The pedestrian bridge of 64,150 m length is considered as a perpendicular.

The theoretical part mainly focuses on the addictive behavior of teenagers on the Internet and social networks. It examines the specific effects of this phenomenon in relation to adolescents and their upbringing. The work also deals with the possibilities of prevention and treatment. The practical part examines the influence and impact of the Internet and social networks on adolescents and their upbringing. The aim of the diploma thesis is to analyze the issue of non-substance addictions among adolescents and the influence on their education. A questionnaire survey was chosen as the research method. Keywords: netholism, non-substance addiction, prevention, education
Lhota Rapotina bypass - location study
Kovářová, Marika ; Efenberk, Tomáš (referee) ; Radimský, Michal (advisor)
The subject of the diploma thesis is the design of the bypass of the village Lhota Rapotina, road II/374, processed in the details of the study. The village of Lhota Rapotina is located in the Blansko district in the South Moravian Region. The task of the diploma thesis was to examine possible alternative solutions of the bypass with connection to the existing and planned surrounding infrastructure related to the construction of the Boskovice – connecting road. The aim was, among others, to propose a solution that would reduce the traffic intensity in the village (especially the high number of trucks transporting material from the adjacent quarry). Three variants of the bypass were developed; eastern and western. From the developed variants, the western bypass variant designed in the S9.5/90 category was selected for closer examination, which respects the location of the planned Boskovice - connecting road and which also serves as a flood protection measure for the village.

The effect of spoil-mining subtrate with heavy metal contents on leaf physiological status with emphasis on phenolics content.
Kovářová, Monika ; Lhotáková, Zuzana (advisor) ; Soudek, Petr (referee)
5 ABSTRACT Heavy metals abundance in the environment increases via natural and anthropogenic processes, mainly mining and industrial activities. Spoil mine substrates of the Sokolovsko Region characteristic by high heavy metal contents are recultivated by tree planting, e.g. of Scots pine (Pinus sylvestris L.). The diploma thesis is a part of a larger project aimed on interpretation of hyperspectral remote sensing data for monitoring of vegetation physiological state. That is why it is focused on non-specific indicators of stress by heavy metals, which can be detected by foliage spectral analyses. The experimental part of thesis comprises 1) field research on P. sylvestris needles and 2) model pot experiments on tobacco plants. Field research was accomplished in 2009 and 2010 on one control locality and three spoil mine banks localities with different heavy metal soil contents (Hg, As and Cu). Photosynthetic pigments', phenolic compounds'and lignin contents were determined spectrophotometrically. Model pot experiments with tobacco plants (Nicotiana tabacum cv. Samsun) - pilot experiment and experiments 1 and 2 using the spoil mine substrates in 2011 and experiment 3 in 2012 also included treatments with different mercury concentration in irrigation (10 and 15 ppm HgCl2). Gasometric measurements (net...
