Determination of Se in selected food supplements using electrochemical generation of H2Se and AAS

The aim of this bachelor thesis was to verify the efficiency of electrochemical generation of H2Se in connection with atomic absorption spectrometry used to determination of selenium in real samples, specifically in selected food supplements with the declared content of selenium. Firstly, working conditions of apparatus for electrochemical generation of H2Se with atomic absorption spectrometry were optimized. Using these optimized conditions, calibration dependences for SeIV a SeVI were measured. It was found that for the quantitative determination of selenate, the prereduction step is needed. Therefore, prereduction of SeVI to SeIV was also optimized. After prereduction step, new calibrations were measured and figures of merit of determination of these selenium species were achieved from them. In conclusion, concentrations of Se in real samples of selected food supplements with the declared contents of SeIV and SeVI were determined and interference effects for such determination have been discussed.

The Role of Lipid Rafts in Translocation of the Adenylate Cyclase Toxin of B. pertussis across Cytoplasmatic Membrane

The bacterium Bordetella pertussis is the causative agent of pertussis or whooping cough. The main virulent factor of this gram-negative bacterium is adenylate cyclase toxin, a member of the RTX protein family. After secretion, the toxin binds to CD11b/CD18 integrin receptor of myeloid phagocytic cells and consequently translocates its adenylate cyclase enzyme (AC) into the cytosol of the host cells. In the cytosol, the AC is activated by calmodulin, followed by rapid conversion of ATP into the signaling molecule cAMP, resulting in paralysis of bactericidal functions of phagocytic cells. Recently it was shown that translocation of the catalytic AC domain into the cytosol proceeds in two steps. After binding of CyaA to the receptor, the influx of calcium ions into the cell occurs. High local concentration of calcium induces the translocation of the CyaA- CD11b/CD18 complex into the lipid raft, where the translocation of adenylate cyclase enzyme into cytosol occurs. This work is aiming on the description of the new established paradigm dealing with membrane translocation of RTX toxins.

Manager decision making

The basic topic of this presented bachelor thesis is the manager decision-making. The main theme of this text is to present the decision-making process. The first part of the thesis summarized basic knowledge about the manager decision-making. There are described and explained main elements of the decision-making process and are mentioned several types of common manager decision-making process. The main part of this paper characterizes the decision-making process and gives information about particular steps of this process, from the identification of the problem to the evaluation of realized decisions. Further part is focused on description, explanation and giving reasons for the importance of quality manager decision-making process and necessity of information for decision-making process. The final part is concentrated on differences, advantages and disadvantages of the intuitional and rational decision-making.

Expression of adaptor protein PAG/Cbp in non-neoplastic and neoplastic lymphoid tissue

New notions about cell development, cell cycle, differentiation and cell signalling are now being used for refinement of classification of malignant lymphomas (ML) and for development of new drugs specifically targeting deregulated proteins some of which play a role in immune signalling. The first step in most of the signalling pathways is transduction of the signal through the plasma membrane mediated by a plasma cell receptor. Certain proximal signalling molecules such as a family of Src-kinases (SFK) are segregated in particular plasma membrane compartments called lipid rafts or glycosphingolipid-enriched microdomains (GEM). I studied expression of PAG in non-neoplastic and neoplastic lymphoid tissue in order to determine its expression pattem and to investigate its utility in routine diagnostic haematopathology.

Sedimenty jako dynamická zásoba fosforu ve stratifikované nádrži

The ecosystem of the small, dimictic, and hypertrophic reservoir (Jordan Reservoir, Czech Republic) was studied from March 2001 till March 2004 to evaluate rates at selected pathways in the P-cycle. The rates were inferred from measurement of P mass-balance, sedimentation of seston, and P-release from sediments. A shift between seasonal changes of inflow and outflow P concentrations caused by P accumulation in the hypolimnion and the sediments was demonstrated. An important amount of P was temporally retained at the sediment surface during the summer stratification but a part of it was released back to water during the autumnal mixing, due to pore water „flushing out” and fast organic matter decomposition.

Hamiltonian cycles in cubic graphs

In this work we study the complextity of Thomasson's algorithm over a special class of cubic graphs. This algorithm nds another hamiltonian circuit, if it starts with a rst one. An open problem is the complexity of this algorithm, but a class of cubic graphs has been found, where for each graph in this class, there exists a hamiltonian circuit and an edge on it, for which Thomasson's algorithm needs exponential number of steps. The goal of this work was to gure out, if Thomasson's algorithm needs exponential number of steps for any hamiltonian circuit and any edge on graphs from this class. We succeeded in proving, that indeed is so.

The role of nuclear weapons in the 21st Century U.S. security strategy : comparation of documentation Nuclear Posture Review from the years 2001 and 2010

The present thesis aims to compare the approach of the Bush and Obama administrations towards the role of nuclear weapons in the United States security strategy. The author focuses at the Nuclear Posture Review (NPR) reports from 2001 and 2010, employing a detailed comparative analysis of their respective content as well as their implications. The analysis concentrates at broader conceptual issues as well as very concrete steps related to specific elements of the United States strategic arsenal. The author of the thesis argues that the current political discourse which attributes a nearly revolutionary character to the approach of the Obama administration to the United States nuclear policy does not fully match the actual dimension of change between the NPR of 2001 and 2010. In fact, the evolution of the United States nuclear strategy maintains its own dynamics in many aspects. Additionally, the present thesis also tries to analyze both documents within the context of the evolution of the United States nuclear strategy since the 1990s which allows the author to identify the shifts as well as the patterns of continuity in the US nuclear policy in the post-Cold War period.

Monitoring of water eutrofization in model reservoir of Czech republic by remote sensing data

Present study compares possibilities of monitoring water eutrophication in the Czech republic using remote sensing. The theoretical part describes basic spectral characteristics of water and their change caused by eutrophication. Chlorophyll-a was chosen as the quantitative indicator of trophic rate. Methods of obtaining value of chlorophyll-a content from scanner TM and ETM+ Landsat satellite data and from in situ measurements data are described. Three regression equations, that employ variety of channels or channel ratios of Landsat data were utilized. The regression equations were applied on three satellite images of model area of Vltava's steps (water reservoirs of Lipno, Hnevkovice, Korensko and Orlík) from the years 2001, 2004 and 2007. The regression equation ln(chl-a) = a.TM2 + b.TM3 + c.TM4 +d gives the best results, where the determination coefficient varies from 0,245 to 0,422. Maps showing distribution of chlorophyll-a concentration in water reservoir Lipno and in the part of water reservoir Orlík were created based on obtained results. Powered by TCPDF (www.tcpdf.org)

SOA Governance

This bachelor thesis is focused on the processes which are used in the service oriented architecture (SOA) enviroment of IT for its governing -- the SOA Governance. That thesis deals with the reasons for an implementation of the service oriented architecture as a resource for fast and effective encompassment of dynamicaly floating conditions of an entrepreneurship. Further the thesis analyzes characteristics of services which are necessary for availing the service as the part of the service oriented architecture. The attention is paid to the ITIL library and ITSM. In that thesis are described the processes that are upheld within the ITSM in the service design phase, in the service transition phase to the productive operations and in the run-time phase of the productive operations of the service. Further is paid attention to the process of continual service improvement. Further the thesis deals with the SOA Governance - Reference Model and the SOA Governance - Vitality Method in more detailes. The thesis analyzes processes of the SOA Governance feasible in the service design phase and in the service run-time phase. The thesis adverts to direct realitonship and synergy between individual phases of the service life-cycle governing of the service oriented architecture and the service life-cycle governing in accordance with ITIL and further adverts to the need of the integrated governance. There are pointed out the basic steps of the implementation of the SOA Governance including the time progress in the example of the practical realization.

The Economy of Marriage in the Czech Republic

The goal of my work is to explain the recent trends in the development of the economy of marriage in the Czech Republic. One of the inseparable parts of my investigation of reproductive decision making using the cost-benefit analysis is the comparison of Gary Becker's economy of marriage, i.e. of the application of Chicago School's theory of consumer demand at the given problematics, with the Pennsylvanian theory of relative income. This comparison is to be done in the conditions of the Czech Republic. Another part of my analysis is formed by the investigation of the much discussed view of children as long-term consumer goods or capital goods. Confronting Chicago School's theory of fertility with its critics is very important. The sub-goal of my work is to prove the connection between marriage rate, birthrate, and economic cycles and also the connection between economic growth and population growth in the Czech Republic. I also concentrate at the change of the trends in reproductive decision making which occurred in the Czech Republic after the fall of communism. Another chapter is devoted to marriage markets and their connection with job markets. The last part of my work concentrates at the effects of pro-natalist politics in the Czech Republic, possibilities of regulating population, and the effectiveness of population politics. At the end of my work I am going to use all my findings to try to answer the question whether it is or is not possible to use the economy of marriage to explain the development trends in the Czech Republic and if necessary I am also going to try to find all the aspects causing deviations from the economic theory.
