Isoflavonsynthasa: přítomnost a aktivita v bobovitých a nebobovitých rostlinách

Isoflavone synthase (IFS; CYP93C) plays a key role in the biosynthesis of the plant secondary metabolites, isoflavonoids. These phenolic compounds, which are well-known for their multiple biological effects, are produced mostly in leguminous plants (family Fabaceae). However, at least 225 of them have also been described in 59 other families, without any knowledge of orthologues to hitherto known IFS genes from legumes (with the single exception of sugar beet - Beta vulgaris, from the family Chenopodiaceae). In view of these facts, this masters thesis has focused on two main objectives: (1) to identify isoflavone synthase genes in selected leguminous and non-leguminous plants exploiting the PCR strategy with degenerate and non-degenerate primers, and (2) to find a system for the verification of the correct function of these genes. Our methodology for the identification of IFS orthologues was successfully demonstrated in the case of two examined legumes - Phaseolus vulgaris L. and Pachyrhizus tuberosus (Lam.) Spreng, in the genomic DNA of which the complete IFS sequences have been newly identified. To design a procedure for ascertaining the correct function of these genes and others once they have been completely described, a pilot study with IFS from Pisum sativum L. The Development of Local Government in the Statutory City of Olomouc.

This master´s thesis deals with issues of  a local government. The research will focus on the city Olomouc. The goal will be to characterize main socioeconomic indicators of the city and evaluate their impact on the development and functioning of the local government. The next step will be identify the actors of the political process in the village and analyze their relationships and define the nature of the decision-making process of local governments on the basis of a comparison of the results of municipal elections. Research approach, that tries to take into account differences in the content and decision-making of local authorities in cities with different Population sizes. The work is proposed  as a case study. Two basic engineering approaches such as study of documents and interviews will be used for collecting of data.Other standard methodological procedures, especially comparison will be used in this work too

Water column separation under hydraulic turbine runner during unsteady operating regimes.

In this diploma thesis called Water column separation under the hydraulic turbine runner during unsteady operating regimes are solved the pressure pulsations of the reverse water hamer. In the thesis is deduced a mathematical relationship of elaboration the numerice model which is based on equations of continuity and equations of forces equilibrium. Numerical model is created in MS Excel uses for computation the numerical method Lax-Wendrof that allows consideration of variable sound speed as function of static pressure and allows variable lenght step in computation domain. Reverse water hammer is in the thesis solved with consideration of rotating flow behind shut-off valve, where we expect forming of vortex rope. This situation can be applied on the closing water turbine which has vertex rope under turbine runner. Specifically for this thesis was carried out the experiment of the reverse water hammer. Constants going into numerical solution are optimalized with using experiment and pressure pulsation are compared between numerical solution and experiment.

Foreign direct investments and their impact on the Czech Republic

The theoretical part of the dissertation is dedicated to the balance of the payments as well as to the theory of the direct foreign investments with the linkage to new methodology of the balance of the payments. The determinants of the direct foreign investments are consequently introduced in the economic theory. The attention is to the higher extent dedicated to the industrial and territorial break-down. The incentive system and the Agency for the support of incoming direct foreign investments (CzechInvest) are described and introduced in the connection with the system functioning in Czech Republic. The incoming direct foreign investments in the period from 2004 to 2014 are analyzed in the second part of the dissertation. The time series were set up on the basis of the annual reports and the databases of Czech National Bank; the evolution phases of direct foreign payments is consequently analyzed with the regard to the respective industries and territories. The development of the direct foreign investments and their impacts on Czech Republic is assessed in the conclusion of the dissertation. It is concluded that the development of the incoming direct foreign investments is in a close correlation with the economic situation in Czech Republic and neighboring countries. The dissertation confirms that the economic and also non-economic factors of each country are key determinants for the final level of the incoming direct foreign investments.

Conflicts at work and the method of nonviolent communication as a way of coping them

Most people have lived through a situation in their work life, where dealing with a conflict was required. Topics connected with human relations and use of communication are delicate and it is often difficult to orientate oneself in them. At the same time, people sense these delicate topics as very important. This thesis introduces a compendium of the conflict issue. It deals with topics like the classification of conflicts, the evolution of the view of a conflict, the problems of a functionality of a conflict, and the process of a conflict. The issue also includes ways of solving conflict and emphasises the importance of an efficient communication in order to prevent conflicts and their constructive processing. Then, the thesis introduces Nonviolent Communication, a popular-psychological method founded by M. B. Rosenberg in the 1960s in USA. The aim of this method is to facilitate people with the means to cope with conflicts peacefully, to break patterns of thinking that use judging or demanding which lead to negative feelings and functionless relations with others, to support mutual respect and to develop colaboration.

Navier-Stokesova equation - solution of the real liquid

This thesis deals with the Navier-Stokes equations for real, compressible fluid with first and second viscosity. The method of expansion into a series of eigenmodes of vibration is chosen to solve the Navier-Stokes equations. The general relations of the expansion coefficients and the eigenmode shapes are given for 1D-, 2D- and 3D-flows. The specific formulae of eigenmodes of vibration are determined only for 1D-flow. The final form of the pressure function is analysed using Matlab software.

Study of diaphragm pump with linear motor

The diploma thesis is aimed at the constructional design of diaphragm pumps with a linear motor. This pump is supposed to be used in medicine for pumping the blood or as an artificial heart. The basic facts, which are focused on the function of the heart and artificial heart, are mentioned in the first parts of the thesis. The next parts describe the diaphragm pumps and the constructional ways of their diaphragm fixation. Furthermore, the basic calculations, describing the function of the pump, are derived there. The basic parameters of the pump are also calculated. The thesis also contains the description of the body pump construction, whose working room is optimized by CFD calculation before the pump is manufactured. The final part of the thesis is concentrated on the measurement evaluation of produced pump model.

Aplikace optimalizačních metod na problémy výroby elektřiny

This thesis deals with application of optimisation methods based on linear and mixed-integer linear programming to various problems in the power sector related to electricity production. The thesis goal is to test the applicability of such methods to formulating and solving various instances from the class of real-world electricity production problems, and to find the advantages and disadvantages associated with using these methods. Introductory chapters describe the main characteristics of power markets, including the historical and regulatory context. Fundamental properties of power markets on both demand and supply side are also described, both from a real-world and a modelling point of view. Benefits of optimisation and modelling are discussed, in particular the solution feasibility and optimality as well as insights gained from sensitivity analysis which is often difficult to replicate with the original system. In the core of the thesis, optimisation techniques are applied to three case studies, each of which deals with a specific problem arising during electricity production. In the first problem, the profit of gas-fired power plant in Slovakia from selling power on the day-ahead market is maximised. The model is set up using both technical and commercial constraints. The second problem deals with the problem of representing a two-dimensional production function which primarily arises for a hydro generator with large variations in the level of its reservoir. Several representations of the original function using piecewise linear subsets are presented, compared, and characterised by their computational intensity both theoretically and practically. In the third problem, the prices on the German day-ahead market in 2011 are modelled. Contrary to the previous two models, the model does not capture an optimisation problem faced by a single producer, but incorporates a large subset of the whole market instead. Consequently the model is formed out of generic constraints relevant to all power plants whose parameters are estimated. By combining information about the aggregate availability of power plants with the estimated efficiencies a full supply curve for each day is created. Different scenarios are analysed to test the impact of uncertain inputs such as unknown or estimated constraints. The choice of the investigated problems stems from the attempt to cover electricity production problems from the point of view of multiple criteria. The three investigated electricity production problems span a broad range from the decisions of a single power plant to the modelling a power market as a whole. Formulations of the production function with different level of detail are presented ranging from a simple linear relationship to several bivariate function formulations. While each problem answers a specific question, they all illustrate the ease with which various electricity production problems can solved using optimisation methods based on linear and mixed-integer linear programming. This is mainly due to the ability of these methods to approximate even non-linear functions and constraints over non-convex domains and find global solutions in reasonable time. Moreover, models formulated with these methods allow sensitivity and scenario analyses to be carried out easily as is illustrated in each of the case studies.

Luxury economics: Comparative study of conspicuous consumption among students of The University of Economics in Prague and Technical University of Liberec

The aim of this thesis is connect to BA František Staněk (who analysed conspicuous consumption on students at The University of Economics in Prague) and verifying hypotheses on students of Technical University of Liberec. Thesis is extended to analysis of the impact of technical education. Main conclusions of this thesis were obtained by statistical and regression analysis of data from a survey. Each brand of consumer electronics are perceived very differently in terms of social status which they express. There is a strong relationship between price of a product and and its signaling value. Students of technical disciplines show a dependency between product functionality and its ability to indicate social status. Others have no correlation.

Signalling of protein kinase B and expression of cyclooxygenase in early phases of diabetic nephropathy

Introduction: Renal hypertrophy, extracellular matrix accumulation, aItered apoptosis as well as changes in regional hemodynamics have been implicated in the pathophysiology of nephropathy in diabetes mellitus (DM). On the molecular level the detailed mechanisms for development of diabetic nephropathy (DN) have becn intensively studied. Insulin induces a variety of biologicaI effects in a number of cell types via phosphatidylinositol-3 kinase (PI3K)/Akt kinase signaling pathway. Considering multiple function of Akt that incIude potentiaIly hannful pro-growth effects mediated by mTOR and cyclooxygenas-2 (COX-2), as well as protective effects mediated by endotheliaI nitric oxide synthase (eNOS), it is possible that aIterations in activities of Akt may play role in the pathophysiology ofDN. Renal corticaI activity and expression of Akt, its down-strearn effectors mTOR, eNOS, and "C<JIX-:Z. as well as PTEN, an endogenous Akt inhibitor, were investigated in streptozotocin (STZ): diabetic rats as a model of Type 1 DM with different levels of glycenůc control, and in Zucker ~d.iabetic fatty rats, a model ofDM2, and in nondiabetic rats as controls.
