Characterisation of recombinant cathepsins B of the bird schistosome Trichobilharzia regenti

This study focuses on the recombinant cysteine peptidases - cathepsin B originating in the bird schistosome Trichobilharzia regenti that is unique across the whole family for its ability to migrate through the nerve tissue to the final localization. For invasion, migration, degradation of nutritional proteins and/or evasion of host immune responses, schistosome employs peptidases. This study follows the research done by researchers of Department of parasitology, Faculty of Natural Sciences, Charles University. The main goal of this study was to deepen the characteristics of recombinant cathepsins B originating in T. regenti. In T. regenti, two cysteine peptidases - cathepsins B1 (TrCB1) and B2 (TrCB2) - have been previously characterized. TrCB1 is located in the gut of schistosomula and involved in digestion. TrCB2 occurs in post-acetabular penetration glands of cercariae and probably facilitates penetration. The recombinant pro-cathepsin B (isoforms TrCB1.1, TrCB1.4 and also TrCB2) were expressed in Pichia pastoris yeast system. Financing of promoting physical activity by the towns with extended scope

Title: Financing of promoting physical activity by the towns with extended scope Aims: Purpose of this work is to explore and analyze condition of financing of promoting physical activity in the towns with extended scope in Czech Republic. I am focusing on budget of towns with extended scope, analyzing of sport financing in these towns, analyzing of profit or loss of sport facilities, utilization of grants, the citizen attitude with this problem and the functionality of departments of sport. Methods: The work is descriptively-analytical. Collecting of data is done by recherché and questionnaire. Analysis and comparison will be used for evaluation of data. I will research whether the number of population is correlated with the amount of money put into the promotion of physical activity and with the number of workers in the Sports Department etc. Results: The towns with extended scope are recognized the importance of physical activities and they are very active to seek financial resources to support them. However, in many smaller municipalities with extended scope is not personnel involved in the sport at all, or this issue is passed on someone as a part-time. Psychosocial Effect Of Bariatric Surgery and the Patient's Satisfaction with the Surgery

The doctoral dissertation is focused on the evaluation of the effect of bariatric surgery and on identifying the parameters that are related to the patients' satisfaction with the surgery. Telephone survey was used to collect data. 122 bariatric candidates were inquired, 86 of them were operated. The average time after the surgery was 20,2 months. The average weight loss was 26.6 kgs. There was a significant improvement in the monitored domains - physical appearance, general health, self-esteem, marital satisfaction, occupational functioning and social functioning. The negative effects of the surgery (post-surgery vomiting, difficulties in dietary adherence, other complications) were also experienced by significant number of patients. In total, 87% respondents were satisfied with the surgery and 13% were dissatisfied. The chi-square test was used to identify factors influencing subjective satisfaction with the surgery and the results were recorded at 95% and 99% significance levels. The satisfaction is associated with the improvement of general health and self-esteem (p<0.01), physical appearance, marital satisfaction, occupational functioning and social functioning (p<0.05). Issues in education of officials of territorial self-governing units in Ostrava

This diploma thesis deals with the education of officials of public administration on the level of territorial self-government. The aim of this diploma thesis is to evaluate a system of education of officials of territorial self-governing units on example of a particular town district authority in Ostrava, to identify problematic spheres and to propose the possibilities of their solution. After introductory determination of the problem in the beginning of the thesis there are theoretical solutions stated and there are contemporary trends of development in education of officials in Europe outlined. Afterwards there is an origin of contemporary system of education of officials of territorial self-governing units in the Czech Republic described in the context of reform of public administration which is followed by the determination of legislative framework, by organisation of education and by overview of participants engaged in this sphere. In empirical part of the thesis there is a research of education of officials at the town district authority in Ostrava implemented and the results of the research including identification of problematic spheres are compared with results of similar researches. In the first part of the paper, the overview of traditional methods used in self-concept research is presented, and the critical analysis of their contribution and limits is given. Further, the attempt to map the new methods and research approaches thhat were created in eighties and nineties in connection with new topics in the field of self-concept research (the complexity of self, the stability of self-concept, the dialogical-self and others) is done.

Media on media : self-representation and representation of competition

This thesis called "Media on Media: Self-Representation and Representation of Competition" describes how, and under which circumstances Czech media project themselves into their news content. It also defines attributes typical for such content, and what are the initiative situations that cause its production. The thesis also determines which of the analysed media types produces the highest figure of selfrepresenting content, and how this amount evolves quantitatively over time. The second part of this work focuses on methods that particular media use to inform on their competition. As a result of quantitative content analysis, which had been used in both parts of the research, it was not only possible to state the overall figure of such contents, but also to specify characteristics usually present in this content. The thesis also shortly analyses the topics and types of these contents. By using the above specified methodology it was also possible to identify particular media that refer about others the most and those that are on the contrary the most mentioned in the news coverage of others. This should clarify functioning of agenda-setting of Czech media in this field.

Factors influencing how popular mathematics is among primary pupils

This thesis focuses on the popularity of mathematics among pupils of a lower secondary school and factors affecting it. Specifically, pupils' self-concept in mathematics and pupils' initial feelings towards mathematics, the importance of mathematics for their future, the personality of mathematics teachers and his/her teaching skills, class climate and pupils' parents' feelings towards mathematics. The theoretical part deals with the processing and interpretation of the various factors that may affect the popularity of mathematics for pupils, as given in literature. Research results of this area on both international and national scales are presented. The practical part is devoted to my research conducted on factors influencing the popularity of mathematics. The research was carried out via a quantitative questionnaire method. The pilot study served to verify the suitability of designed questionnaires. The results of the main study are carefully discussed (given an opportunistic sample of pupils) in terms of my hypotheses and compared to the results of the international research by the OECD/PISA 2003. Among the factors that influence the popularity of mathematics among pupils most is the influence of teachers' approach and of parents and a pupil's self-concept.

Preclinical assesment of phthalocyanine for photodynamic therapy of rectal carc inoma

The present work includes research photodynamic therapy (PDT) colorectal carcinoma. The actual experiment was conducted in collaboration with the Institute of Biophysics and Informatics 1st in 2005-2008. Colorectal cancer is the most common malignancy of the Czech population. Risk of disease of colorectal cancer in the Czech population reaches 5.9% and still growing. Still is a large group patients with advanced cancer (approximately 30% of patients), which is indicated only palliative therapy. One of the most common procedure in patients with advanced cancer is the foundation of the stoma, which significantly extends the life of the patient. Photodynamic therapy is already used by default. We focused on the possibility of treatment of colorectal cancer phthalocyanine, which is among photosensitisers second generation with a rapid uptake in the tumor specimen. The aim of current research is to improve screening and to design new treatments that would achieve better results than existing procedures. PDT can improve the quality of life and in patients with advanced malignant disease and prolong survival interval.

Effectivenes of cognitive-behavioural treatment applied in courses of weight reduction related to cognitive and behaviolal factors

The Czech Republic is from the early 90 years, reducing rates of overweight in CBT applications, which the company operates STOB Mgr. Iva Malkova. According to available information has not been done until now no research in these courses chartedabove the influence of selected cognitive factors on changes in body weight and also to monitor the change in energy intake (behavioral factor). The aim of this study is to determine the relationship between selected cognitive factors (self-efficacy - and with respect to the general dining behavior and physical activity, self-esteem, personal causal orientation restrictive way of eating behavior) and changes in body weight. At the same time the respondents will track changes in energy intakeduring therapy. The study is to verify the reliability of the Czech version of the WELquestionnaire (Weight Efficacy Life-Style Questionnaire), PSQ (Physical Self-Efficacy Questionnaire) and extended range of restrictive behaviors related to food, based on a questionnaire TFEQ (Three Factor Eating Questionnaire).

Supporting the client´s selfreflection in following a diet regimen

Thesis on the theme Client{\crq}s self-reflection support during observing dietary regime is divided into two parts. The problems of dietary regime and self-reflection is mapped on the basis of studied literature in a theoretic part. Practical part deals with a research which is focused on nurses` supporting client{\crq}s self-reflection during observing dietary regime or whether clients use a self-reflection during observing dietary regime and whether their family members support them. The research was made with a method of questioning, questionnaire technique, in hospitals in České Budějovice, Písek and Příbram and was aimed at three informant groups.
