South Bohemian sacral architecture of the late Gothic1450 - 1550 in the Rosenberg domain Mgr. Roman Lavička A group of late-Gothic churches, which keeps attracting our attention, was built in the Rosenberg domain during the reign of the Jagiellon dynasty. Limited archival material, results of dendrochronologic analyses and preserved buildings suggest that it was mainly building maintenance that was in progress in the Rosenberg domain in the second half of the 15th century. In the early 1480s the building of the new pilgrimage church in Kájov (1471/1474- 1485) was coming to its end. And it was only then that big town parish churches in Trhové Sviny, Dolní Dvořiště, Hořice na Šumavě, Chvalšiny and Nové Hrady started being rebuild or constructed anew almost simultaneously. Around 1485 almost twenty religious buildings were being built in the Rosenberg domain. They reflect the increasing economic prosperity and self-confidence of the town and village municipalities, corporations as well as individuals manifesting their wealth, success and strength. From a formal and ideological point of view it is possible to trace down a group of related buildings directly in this initial phase. Struggle for the right to the city: Alternative spaces in post-socialist Prague

The dissertation focuses on spaces in post-socialist Prague which transcend the hegemony of capitalist social and spatial relations and provide alternative spaces for non-profit culture and grassroots activities used and operated by students, artistic and creative communities, alternative subcultures, and NGOs. The author presents 14 case studies focused on these spaces, whose existence in the city is threatened. Referring to Marxian urban theory and the concept of the right to the city, the author critically investigates the democratic character of the social structures which are based on the contemporary socioeconomic model, and interprets the way in which the imperative of capital-accumulation, combined with the legacies of the totalitarian past, constrain the development of open democracy and civil society, and the creation of diverse, vibrant, progressive, and socially inclusive urban environments. The empirical part of the dissertation outlines the process of creating and operating alternative spaces in Prague during a changing political-economic context, and describes in detail, alternative spaces that existed in Prague in the early 21st century. The Forest of Laughs (Xiaolin): mapping the offspring of self-aware literature in ancient China

Literary history, as established in 20th century China, mostly believed that Confucian conservatism had always oppressed and marginalized practices of "humour" in China.1 This view, formulated in early 20th century when anti-traditionalism prevailed among Chinese intellectuals, regarded entertaining practices as suppressed and suffocated by Confucian moralizing and at that time even the notion of "humour" itself was introduced to China using an English word (humour-youmo ).2 As a result, in sinology until recently the topic of "humour"3 in literature was - with only few exceptions - 4 perceived as marginal to the understanding of ancient Chinese society and culture (as very few works have been published on the topic of Chinese humour, which, though, do not bring valued insights to the topic).5 However, in early sources there are evidences of entertaining practices linked to humour, which can be traced back to Warring States period. ABSTRACT This dissertation deals with the issues in motor skills in pre-school children. The first part of the dissertation introduces basic theoretical background in relation to motion, refers to developmental stages and principles in motor development, and deals with the functioning of the motor system. Motor ability and skills are defined as basic preconditions for successful motor performance. The issues related to the developmental disorders in coordination (dyspraxia) are also discussed. The second part of the dissertation introduces the actual research aimed at detecting motor weakening, especially in the area of coordination ability, in pre-school children. Therefore, a diagnostic screening tool was made in order to map the state of coordination abilities in pre-school children. The screening test helps to detect deficits in motor skills at an early stage.

It focuses in detail on individual pharmacotherapeutic groups and their ability to influence the progression of the disease and patient's quality of life. It also refers to treatment during pregnancy and lactation. Further, the thesis describes essential non-pharmacological interventions and means of surgical treatment. Biological treatment has been a breakthrough in the therapy of rheumatoid arthritis and we may anticipate its growing importance in the future. The key to successful treatment are early diagnosis and correct assessment of the disease activity as the prognosis of the disease depends mainly on early start of the therapy.

Early care of children with the compound disablement, focused on care of caring parent

In my thesis titled {\clqq}Early care for children with combined disability with the focus on the care for caring parents`` the following issues are dealt with: disability problems, the system of the early care in the Czech Republic, possibilities, techniques and methods used in the early care for children with combined disability. The thesis also refers to the role of the caring parent as the main component of the team of early care providers, the process of coping with the child disability and the danger of the burn-out syndrome. For the purpose of this study I used the educational projects prepared by the association Alfa Human Service {\clqq}Education and practice of persons caring for a dependent family member (CZ.04.3.07/2.2.1.01.1/2004)`` and the project {\clqq}Support for the persons potentially endangered by unemployment (CZ.04.3.07/1.1.01.3/1088)``. The objective of my thesis was to survey the needs of caring parents, their expectations and the effectiveness of educational activities aiming at the social integration and the access to the job market.

Frege and Husserl on Objectivity

This bachelor thesis is concerned with early works of significant logi- cians and philosophers Gottlob Frege and Edmund Husserl at the turn of the nineteenth and the twentieth centuries. Most importantly, it deals with their solution of the objectivity of arithmetic and the objectivity itself. At first they both started in the same direction and they har- shly rejected psychologism. They also introduced similar differentiation of sense and reference. According to Frege, the reference of a sentence is a truth value, however, according to Husserl, it is a state of affairs. The sense is the way of referring to an object, also called intentionality. The sense in a sentence is a thought according to Frege and it is a noema according to Husserl. They both put emphasis on objectivity of number but they gradually went in different direction. Frege identified number with extension of concept, however, this subsequently let his system to dispute. Husserl derived number by abstraction, which exposed him to Frege's harsh criticism. Early eneolithic in central Bohemia with reference to site Prague - Krč

Úkolem této práce je v prvé řadě shrnutí současné koncepce časného eneolitu na území Čech. To bylo provedeno rozdělením kultur do kapitoly začínající pozdně-lengyelským horizontem a končící michelsberskou kulturou. Sledovány byly typické znaky, charakterakteristické nálezy i diskutované otázky jednotlivých kultur a příslušných nálezových skupin. Dalším cílem práce je vytvořit seznam středočeských časně eneolitických lokalit, což bylo provedeno sepsáním katalogu nalezišť podle okresů s následným vyobrazením v mapách podle kulturního zařazení. Poslední částí této práce je zveřejnění post-jordanovských (shussenriedských) keramických nálezů z lokality Společenská zahrada - Praha-Krč, jejichž materiální základna se v oblasti středních Čech stále více rozšiřuje. Analýzou prošel pouze keramický materiál, další nálezy čekají na zpracování. Kriticky jsou zhodnoceny i dva post-jordanovské objekty (palisádový žlab, kostrový pohřeb v sídlištním kontextu).

The knights Kaplíř of Sulevice on the treshold of the early modern age

This thesis focuses on the history of the the family of knights Kaplíř of Sulevice. It covers a period from the first reference to this family before the half of the 14th century until the death of the last male member in the lineage at the beginning of the 18th century. The family Kaplíř of Sulevice belongs among significant members of the estate of Czech knights. In the course of the 14th and at the beginning of the 15th century the members of the family built up their property structure and that time there were 5 descents formed: descent of Sulevice, descent of Ostrý, descent of Solany, descent of Skalka-Řehlovice and descent of Vimperk. Thanks to the Hussite period the knights of Sulevice, the men of great enterprise, succeeded in penetrating more significantly even beyond boundaries of their own estates. The half of the 15th century was marked with certain stagnation (except the descent of Vimperk) which, on the contrary, was in their prime.

Laboratory diagnostics of micrometastases in breast cancer patients

Introduction: The presence of circulating tumor cells (CTC) in the peripheral blood has been associated with worse prognosis and early relapse in breast cancer patients. CTC determination in the peripheral blood has been considered as a liquid biopsy. The aim of this project was to analyze the presence of CTC followed by their molecular characterization with the potential use not only as a new biomarker for real-time monitoring of therapy efficacy but also as a suitable tool for patient's stratification and individualization of treatment for breast cancer. Methods: A total of 54 patients with diagnosed early breast cancer were enrolled into a prospective study. Ten millilitres of peripheral blood were sequentially collected to test for the presence and characterization of CTC during the follow-up of patients. CTC isolation and detection was performed by AdnaTest BreastCancer™ (AdnaGen AG, Germany), which is based on the detection of EpCAM, HER2 and MUC1 specific transcripts in enriched CTC- lysates. cDNA from isolated CTC has been further used for newly optimized qPCR assays for breast tumor and therapy resistance associated genes: TOP1, TOP2A, CSTD, ST6GAL, KRT19 and reference gene actin. qPCR results have been analyzed by Genex software (MultiD Analysis).
