Yeast as a tool in development of new antivirals
Dubská, Jitka ; Janderová, Blanka (referee) ; Mašek, Tomáš (advisor)
The increase of infectious chronic diseases is the main health problem of modern civilization. These diseases, which are spread both in developing as well as industrial countries, cause more than 59% of all life ends. Statistically, the most frequent infection is hepatitis C virus (HCV) infection. Incurable hepatic illnesses such as cirrhosis and hepatocellular carcinoma are the outcomes of the HCV infection and the liver transplantation is the only successful therapeutic option for the patients in industrial countries. It is estimated that about 170 million people worldwide are chronically infected with this virus. HCV is a small coated virus with single stranded RNA genome of positive polarity. Lots of research has been done since it was discovered twenty years ago but the efficient drug is still missing. There is a strong tendency to focus the research on searching for translation initiation inhibitors in last few years, because the whole life cycle of HCV, including RNA replication, new virions assembly etc., is dependent on translation products. Nowadays there is a small group of human proteins (so called ITAFs) known for up- or down-regulate HCV and other viruses translation initiation. It is believed, that this group is much more comprehensive. The aim of this diploma work was to construct yeast...

"Art" and its definition. Between function and procedure
Ciporanov, Denis ; Kulka, Tomáš (advisor) ; Haman, Aleš (referee) ; Zuska, Vlastimil (referee)
The thesis explores the problem of definition of "art" as it has been systematically discussed since the second half of the 20th century - mainly in the Anglo-American analytical aesthetic tradition. Despite the complexity and diversity of this discussion, it is possible to indicate two opposite definitional approaches. The first is the so called proceduralism which is established on the conviction, that the art status is connected with certain performative procedures (social, theoretical or historical in character), and that its attribution to an artifact essentially affects its artistic/aesthetic function. The second is a functional strategy, traditional in character, which presupposes on the contrary, that ascription of art status comes always after the indication of functional efficiency of the artifact in question. In four parts of this thesis we want to argue for the superiority of functional view of the concept of art. To do that, we made one exceptional step out of the borders of analytical tradition in order to see how this traditional problem is solved in the theoretical context of Prague Structuralistic School, domain of European functional thinking. The comparison of Jan Mukařovský, the main representative of functional approach to art, with Nelson Goodman, one of the most prominent American...

The ability of new firms to enter the monopolistic enviroment
Diblík, Petr ; Ježek, Tomáš (advisor) ; Dočkal, Dalibor (referee)
The present thesis deals with topic of monopolies and ability of new entities to enter mar-kets where such monopoly is present. For a deeper understanding of this field various views on monopolies in history of economic thinking are mentioned in introduction. In connection with possibility of new companies establishing themselves on the market there is analyzed creation and current practice of anti-monopoly legislation. In this part, certain arguments favoring protectionist policies are impugned. On particular examples it is dem-onstrated that more than privileges from the part of the state it is ensuring of free access to a given field which helps to a starting company. The state should be a guarantor of law enforcement and oversee observation of generally binging rules. In the conclusion of the work a difference is also shown between a situation where a monopoly originates in a natu-ral way as a result of exceptionality, which the clients positively value and a monopoly which exists due to support by state. A new company, as a continuous potential competitor, presents a good-quality feedback for a dominant entity and a driving engine for enhancing its efficiency. A free economic competition is shown also here as an effective tool for de-velopment of business, and it is the consumer who derives major benefits from such situa-tion.

Bone metabolism, infection and ischemia in diabetic foot syndrome
Bém, Robert ; Jirkovská, Alexandra (advisor) ; Štěpán, Jan (referee) ; Broulík, Petr (referee)
The diabetic foot (DF) is one of the most frequent late complications of diabetes, which often leads to limb amputation and effects the quality of life in patients with diabetes. Ischaemia and infection play important role in pathogenesis of DF. One form of DF is Charcot neuroosteoarthropathy (CNO), defined as a noninfectious destruction of bones and joints in the foot due to diabetic neuropathy. The subject of the dissertation thesis was divided into 3 parts: bone metabolism and CNO, ischaemia and infection in DF and new methods in the treatment of DF. (...) In the third study, new quantitative scintigraphic parameters for assessment of CNO activity were defined and confirmed their satisfactory correlation with other parameters of CNO activity. In fourth and fifth study, good reproducibility was verified and efficiency of power spectral analysis of heart rate variability as a new method for diagnosis of autonomic neuropathy in patients with CNO. Sixth study refered to rare causes of atypical localization of CNO in the knee joint. Ischaemia and infection in DF were resolved in one study. In multivariate analysis of seventh study we approved, that risk factors for difficulty healing after minor amputation of the foot were low level of transcutaneous oxygen tension and high level of C-reactive protein before...
