Isolation and fractionation methods for determination of humic substances content in South Moravian lignite
Černý, Pavol ; Kalina, Michal (referee) ; Enev, Vojtěch (advisor)
This bachelor thesis focuses on isolation and fractionation of humic substances (HS) from the natural matrix of lignite, which comes from southern Moravia from the mine Mír Mikulčice. In the theoretical part, it informs the reader about the issue, starting with information about lignite and HS. In this work, the standard procedure of isolation and fractionation of HS was used as according to the International Humic Substances Society (IHSS). The content of HS in the natural matrix of lignite was determined, and then isolated HS were subjected to physicochemical characterization – elemental analysis (EA), thermogravimetric analysis (TGA), UV/Vis spectrometry, infrared spectrometry with Fourier transformation (FTIR) and the acidity of humic acids (HA) was determined by potentiometric titration. EA was used to detemine the content of biogenic elements, moisture and ash content were determined by TGA. By using FTIR, various functional groups and building blocks that are present in supramolecules of HS were indentified and absorption coefficients EET/EBT, E2/E4, E2/E3 a E4/E6 were determined by UV/Vis spectrometry. Results represent good characterization of HS isolated from lignite and are in agreement with scientific literature.

The Effect of Accumulation on Distribution Grid Operation
Jestřáb, Tomáš ; Černý, Pavel (referee) ; Toman, Petr (advisor)
This diploma thesis introduces the reader to the possible types of electric energy storage technologies, their comparisons and utilization. After its introduction, it follows the description of individual technologies and principles that are currently meaningful and useful in practice. The thesis continues with the part characterizing the ways of using the accumulation in the grid. However, the main points of this thesis include the analysis of the influence of the photovoltaic installation on the specified distribution grid operation at the low voltage level and the analysis of the influence on the grid with connected accumulation. Another important objective was the model for the calculation of steady-state, which was programmed on the basis of the theory supported by the principles of production from intermittent sources and the principle of accumulation function. The outputs of the model in the form of the calculated voltage ratios at the individual supply points and current flows at the distribution transformer point were used to evaluate the analyzes mentioned above. The courses of the calculated parameters are broken down by different scenarios (according to the period, the degree of integration of production and the accumulation or the power output of the production), graphically processed and compared from the point of view of voltage fluctuations and changes in the power (or current) balance of the supply transformer.
Prosthetic Solutions to Transmetatarsal Traumatic Amputation
Hráček, Karel ; Pavlů, Dagmar (advisor) ; Černý, Pavel (referee)
Title: Prosthetic solution of traumatic transmetatarsal foot amputation Objectives: The main goal of my work is to design a prosthetic solution for a patient after a traumatic transmetatarsal amputation of the leg with the intention of restoring the function of the leg with respect to its natural appearance, maximally reducing the risk of injury to the patient so that he can use normal sports shoes. A partial goal is to increase the mechanical resistance of the prosthesis during increased physical activity and to create a production procedure that can be used by other active patients. Methods: The bachelor thesis consists of a theoretical part and a practical part. In the theoretical part I primarily draw on professional publications and articles related to the given issue. The practical part consists of the actual case study of the patient with a focus on the production of a prosthesis according to his disability, which is based on the data obtained from the analysis of posture and gait. Results: The case report was processed within two months in the non-state medical facility Proteor CZ s.r.o. Ostrava, based on the specific needs of the patient. The resulting prosthetic product met all the goals we set together with the patient. The manufactured prosthesis improved the patient's movement...
Viral proteins that facilitate RNA replication
Černý, Prokop ; Bouřa, Evžen (advisor) ; Faltová, Lenka (referee)
In recent years, the pathogenic human coronavirus SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) has caused a global pandemic of highly infectious disease COVID--19 (coronavirus disease 2019). According to the World Health Organization, as of August 20, 2023, there have been nearly 770 million confirmed cases and nearly 7 million deaths associated with the disease. However, only a small number of effective antiviral drugs are currently known to help treat the disease in already infected patients. Therefore, in this work, the interaction of host 14-3-3 proteins with the nucleocapsid protein of SARS-CoV-2 was studied as a potential biological target for the rational design of new antiviral drugs. The importance of this interaction in the virus life cycle has not been fully elucidated, but it probably has an important regulatory function affecting multiple processes in the infected cell. The SARS-CoV-2 N protein contains several binding motifs for 14-3-3 proteins in its sequence, two of which (at positions S197 and T205), most likely responsible for the protein- protein interaction, were studied in this work. Microscale thermophoresis was performed to determine the affinity of each of these phosphorylation sites on the nucleoprotein for the 14-3-3ζ protein. The key phosphorylated residue...

Importance of Kind Approach in Care of Seniors
ČERNÝ, Petr
The work deals with a kind approach in the care of seniors. The first part of the work addresses issues such as ageing and old age, division of old age, psychic and social aspects of old age and old age in the Bible. The second part defines the concept of {\clqq}kind approach`` and demonstrates such a kind approach on the examples from the Bible and personalities of Christian charity and the Catholic Church, who became known by their kind approach. The third part discusses the kind approach in perspective of social work, of psychotherapy and importance of kind approach in the care od seniors. The kind approach in the care of all those in need stems from Christian love agapé, caritas), it is also an important therapeutic instrument of Rogers´ psychotherapy and in this meaning relevant in social work, especially in social work with seniors.
Are Women More Risk-Averse than Men? A Meta-Analysis
Černý, Patrik ; Havránek, Tomáš (advisor) ; Bauer, Michal (referee)
Are women more risk averse than men? While a large portion of economic literature confirms this phenomenon, the link between gender and risk aver- sion has not been found to be consistent. We investigate the difference in risk aversion between genders employing meta-analytic methods on 147 estimates collected from 25 primary studies, converted to partial correlation coefficients. We find positive publication bias in our dataset, suggesting that the reported estimates in primary studies are exaggerated. After adjusting for the publica- tion bias, men do not seem to be more risk-taking than women. In addition, we employ Bayesian Model Averaging to examine heterogeneity among the es- timates, controlling for additional 68 variables reflecting the design of primary studies. Based on the heterogeneity results, we find that the publication bias in our dataset is driven by 'lower quality' studies based on the RePEc ranking of economic journals. This finding also aligns with our robustness checks on sub- sets divided according to the quality of journals publishing the primary studies.
