The effect of adjustment of sleep patterns on mental stress and body weight
Mutinský, Petr ; Manková, Denisa (advisor) ; Nekovářová, Tereza (referee)
This study aimed to investigate the impact of sleep pattern modification on sleep quality, circadian preference, mental strain, and body composition. Sleep pattern modification was implemented through a 14-day intervention involving three groups with varying levels of engagement. All participants received recommendations compiled into a sleep manual. The primary objective was to examine the relationship between sleep pattern modification and changes in sleep quality, circadian preference, stress levels, and body composition, including body fat and skeletal muscle mass. Additionally, the study sought to determine whether the level of engagement in sleep pattern modification within each group influenced post- intervention sleep quality. The study employed an online questionnaire battery comprising the PSQI, MEQ, and PSS-10 questionnaires, completed by N = 95 physically active participants aged 18-53 years. Body composition measurements were obtained using an InBody device. Repeated measures ANOVA analysis revealed a statistically significant improvement in sleep quality following the intervention as measured by the PSQI questionnaire (F(1, 77) = 144,31, p < 0,001, ηšₚ = 0,66). However, no significant interaction between the intervention and individual groups was found (F(2, 75) = 1,29, p = 0,28, ηšₚ...
Lifestyle changes in young adults in the context of the COVID-19 pandemic
Baroňová, Martina ; Kolářová Veverková, Martina (advisor) ; Malvotová, Pavla (referee)
This bachelor's thesis evaluates the lifestyle changes of young adults caused by the covid-19 pandemic. The aim of the thesis is to understand how the change in lifestyle associated with the pandemic was experienced and evaluated by two groups of young people - students and workers - in the Czech Republic and what permanent changes it brought to their lifestyle. From a theoretical point of view, the work is based on Pierre Bourdieu's concept of habitus. It mainly focuses on two areas of lifestyle - healthy lifestyle and free time. As one of the very significant problems of a healthy lifestyle, the sedentary lifestyle that the pandemic has promoted is considered. The main core of this work is semi-structured in-depth interviews with seven communication partners. Interviews with communication partners showed that almost half of them encountered a major impact on their life path during the pandemic, in the form of, for example, loss of housing or employment. As a result of the pandemic restrictions, they began to prefer home food more and to limit eating at fast food restaurants. Students were more affected by social isolation and a sedentary lifestyle than working individuals. The restriction of outdoor sports fields has caused a change in physical activities for some young adults, with young people...
Repeated low-dose flubendazole treatment and its effect on glioblastoma cells
Namyslová, Marie ; Szotáková, Barbora (advisor) ; Králová, Věra (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Marie Namyslová Supervisor: prof. Ing. Barbora Szotáková, Ph.D. Title of diploma thesis: Repeated low-dose flubendazole treatment and its effect on glioblastoma cells Glioblastoma (GBM) is one of the most aggressive and common primary brain tumours in the adults. GBM is resistant to current treatment and the median survival of treated patients with this disease is around 15 months. The very poor prognosis of this disease has prompted the research of new therapeutic strategies. A promising direction is represented by drugs already registered for other indications, such as the anthelmintic flubendazole (FLU) with potential antitumour activity. The aim of this work was to study the effect of FLU after repeated low-dose treatment of GBM cells. Two cell lines, U87MG and U87MG-IDH1mut, were used for this work and the effect of repeated doses of FLU on cell viability (using the WST-1) and on cell morphology (via phase contrast microscopy) was investigated. Furthermore, changes in the expression of cell cycle markers, proliferation, growth, and resistance markers after FLU treatment were evaluated at the mRNA level (RT-PCR) and protein level (Western blotting). Subsequently, the activity of initiator and...
The ways to aestheticize consumption and advertising in three chosen modern novels
Averina, Aleksandra ; Janišová, Míla (advisor) ; Voldřichová - Beránková, Eva (referee)
The aim of this thesis is to analyze the means of aestheticizing consumer society and advertising in three works: Frédéric Beigbeder's "99 Francs," Christophe Léon's "Black Friday," and Gauz's "Debout-payé." This selection of works allows us to see the image of consumption from three different angles. In Beigbeder's work, we find the people who produce advertising, Christophe Leon opens up the world of consumption and purchasing, and in Gauz's "Debout-payé," we see people working "standing up" so that others can consume. The narrator in all the novels attempts to convey the image of consumer society, with each narrator choosing their own social level. Our analysis will be based on Jean Baudrillard's theory of consumption and Umberto Eco's thoughts on mass media and communication. Given the position of this work at the intersection of literature and sociology, one chapter will be dedicated to the literary styles that authors choose to aestheticize or "de-aestheticize" consumption. In the other chapters, we will analyze the social aspect of purchasing in the novels.
Selected Characteristics of Adolescent Female-to-Male Gender-affirming Medical Care Applicants
Schönauerová, Dominika ; Weiss, Petr (advisor) ; Kolečková, Tereza (referee)
The aim of the bachelor's thesis was to analyze selected anamnestic characteristics of adolescent FtM (female-to-male) applicants seeking gender-affirming medical procedure, in order to provide a clearer understanding of this population within the Czech Republic. This purpose was served by data from patients of the Institute of Sexology, General University Hospital in Prague (N = 78), collected in 2022-2023, who were referred to care at the age of at least 15 and less than 19 years (M = 16.3, SD = 1.00). Within the sample late-onset gender incongruence with the first self-recognition at the mean age of 13,2 years (SD 1,98), was typical, as well as complete social transition at the time of first assessment, initiated at a mean age of 15,1 years (SD 1,29). Minority sexual orientation and the desire for complete gender reassignment surgery were also characteristic. Almost half of the FtMs had experienced a romantic relationship, but only one third were sexually active. Half of the patients had a history of psychiatric pharmacotherapy and more than one-fifth of psychiatric hospitalization. A number of psychiatric comorbidities were present. The most common comorbidity was the mixed anxiety-depressive disorder present in one-third of the FtMs. More than half had a history of self-harm, suicidal...
Integration of Arabic loanwords into Czech
Columby, Aleksandar ; Zemánek, Petr (advisor) ; Pospíšil, Adam (referee)
Arabic loanwords are present in many Slavic languages. Arabic words have also entered the Czech language, and still do. Arabic loanwords in Czech have been studied previously, however to a small extent. This research focuses on the contemporary layer of Arabic loanwords, which previous research did not deal with. Specifically, the work deals with their semantics. The author asks the question whether contemporary Arabic loanwords in Czech undergo changes in meaning, and if they have undergone changes, the author asks what changes. He bases his answer on a comparison of their meaning deduced from the collocations that they form in corpora. Using the Czech National Corpus (ČNK), the author first created a list of contemporary Arabic loanwords and then examined their collocations in the Czech corpus SYNV12 and the Arabic corpus The Arabic Web Corpus (artenten18). The research shows that most contemporary Arabic loanwords in Czech (12/18) have not undergone a significant change in meaning. All the words that have undergone a change (6/18) have narrowed their meaning. In addition to the meaning being changed by narrowing it down, two words (šarīᵓa and ğihād) have acquired a negative connotation. Despite certain limitations, this research brought new knowledge about the current layer of Arabic loanwords...
The expression analysis of aldo-keto reductases in Haemonchus contortus
Babičková, Markéta ; Matoušková, Petra (advisor) ; Szotáková, Barbora (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical sciences Candidate: Bc. Markéta Babičková Supervisor: doc. Ing. Petra Matoušková, Ph.D. Title of diploma theses: The expression analysis of aldo-keto reductases in Haemochus contortus Haemonchus contortus is a blood-sucking nematode found mainly in livestock. This nematode causes a disease called haemonchosis. During its evolution, it has developed emerging resistance to anthelmintics, a is growing global problem despite good strategies. Aldo-ketoreductases (AKRs) belong to the super family of NADPH-dependent oxidoreductases catalyzing oxidation-reduction reactions of aldehydes and ketones in various organisms from bacteria to mammals. In mammals, they are abundantly expressed in various tissues. Their essential role is mainly in the metabolism of hormones, xenobiotics, eobiotics, oxidative molecules or a role in drug resistance. Some species, such as aldosareduktasa (ADR), are also capable of damaging the organism. Unfortunately, very little has been described about these enzymes in H. contortus, and therefore this thesis deals with the investigation of putative AKR genes in this nematode. Genome sequencing identified 22 genes from the AKR superfamily. Expressions of all AKRs were detected in eggs, larvae...
