Barbora Szotáková, Ph.D. Název diplomové práce: Opakované nízkodávkové podání flubendazolu a jeho vliv na buňky glioblastoma multiforme Glioblastom (GBM) patří mezi nejagresivnější a nejčastější primární nádory mozku u dospělých. GBM je odolný vůči dosavadní léčbě a medián přežití u léčených pacientů s tímto onemocněním se pohybuje okolo 15 měsíců. Velice špatná prognóza této nemoci podněcuje ke studiu nových terapeutických strategií. Nadějný směr představují léčiva již registrovaná pro jiné indikace, jako je např. anthelmintikum flubendazol (FLU) s potenciální protinádorovou aktivitou. Cílem této práce bylo studovat účinek FLU po opakovaném nízkodávkovém ovlivnění buněk GBM. Pro tuto práci byly použity dvě buněčné linie - U87MG a U87MG-IDH1mut a byl studován vliv opakovaného podání FLU na jejich viabilitu (metodou WST-1) a na morfologii buněk (mikroskopie fázového kontrastu). Dále byly po podání FLU hodnoceny změny exprese markerů buněčného cyklu, proliferace, růstu a markerů rezistence na úrovni mRNA (RT-PCR) a na úrovni proteinů (Western blotting). Následně probíhalo stanovení aktivity iniciačních a efektorových kaspáz (pomocí luminiscenčního měření) a stanovení... GBM is resistant to current treatment and the median survival of treated patients with this disease is around 15 months. The very poor prognosis of this disease has prompted the research of new therapeutic strategies. A promising direction is represented by drugs already registered for other indications, such as the anthelmintic flubendazole (FLU) with potential antitumour activity. The aim of this work was to study the effect of FLU after repeated low-dose treatment of GBM cells. Two cell lines, U87MG and U87MG-IDH1mut, were used for this work and the effect of repeated doses of FLU on cell viability (using the WST-1) and on cell morphology (via phase contrast microscopy) was investigated.
