Bachelor thesis expands an architectural study elaborated within the framework of the course AG33 to the stage Documentation for permission of construction and Documentation for building permission. The topic of the thesis is a Culture Center in Brno, for which was selected a lot on the street Údolní in front of square Obilný trh in a slope under the Špilberk. Close to the lot is located University hospital Brno – maternity hospital and the building of Public Guard of Rights and historical build-up area from 19. century, which strictly keeps a street line, this street line was disrupted by gap site during the 2. World War, due to the bomb fallout. New building of the cultural center is designed in this gap site between two historical houses and completes original frontageline. The main idea of design was to keep the fromtage line and at the same time interconnect by designed project an opposite square with a park and castle Špilberk. Object comprises three parts. The main object is in the shape of a letter L and its back part is partly built in the slope, in bigger wing of the building is located a foyer, a big concert hall, performers, staff and technical facilities and storage area in the first floor, small conference hall with foyer in the second floor and gallery in the third floor, in smaller wing café and gym are located. Second part includes sheltered penthouse in the shape of letter L which is designed in form of steel structure, which is adjacent to the main object and thus creates square-shaped yard. The cover at the same time creates sheltered passage from Obilný trh onwards to Špilberk. Third object is the smallest one, it has cuboid shape and it is defined as a commercial space especially for free-time activities.
Use of cytogenetical methods in taxonomy of Arachnida (Arachnida)

This bachelor thesis deals with the use of cytogenetic methods in taxonomy of Arachnida. To understand this matter, we need to analyse available information about inter and intraspecific karyotype variability of individual orders. At the beginning of the thesis there is a description of cytogenetic methods used with Arachnida.They are used to gather the karyotype's fundamental information, such as the diploid number of chromosomes, chromosome morfology and possible occurence of sex chromosomes. Main part of the thesis is focused on describing karyotypes of selected orders (Amblypygi, Palpigradi, Pseudoscorpiones, Scorpions, Opiliones and Araneae) and considering the convenience of use of cytogenetic methods for their taxonomy based on these information. The thesis includes current number of described genera (species) of the orders, as well as a number of cytogenetically analyzed genera (species) for comparison.
A study of the HCV IRES variability: An experimental approach coupled with design of a large-scale mutation database

Translation initiation in the hepatitis C virus (HCV) occurs through a cap- independent mechanism that involves an internal ribosome entry site (IRES) capable of interaction with and utilization of the eukaryotic translational machinery. We focused on the structural configuration of the different HCV-IRES domains and the impact of IRES primary sequence variations on secondary structure conservation and function. For this purpose we introduced into our laboratory, methods such as denaturing gradient and temperature gradient gel electrophoresis for screening the degree of heterogeneity and total amount of HCV-IRES variability accumulated in HCV infected patients over a period of time. The selected samples showed variable migration pattern of the HCV-IRES (from all the patients) visualized in DGGE and TGGE, were sequenced and evaluated for translation efficiency using flow cytometry. In some cases, we discovered that multiple mutations, even those scattered across different domains of HCV-IRES, led to restoration of the HCV-IRES translational activity, although the individual occurrences of these mutations were found to be deleterious. The Inflation-Output Variability Relationship in the CEE countries: A Bivariate GARCH Model

This thesis examines the output-variability relationship and causal relationships among the inflation, the output growth and their uncertainties for the Central and Eastern European region during the period of time that covers the economic crisis of 2008. We apply the bivariate GARCH(1,1) model with the constant conditional correlation covariance matrix to obtain conditional variances that proxy the two uncertainties and use Granger causality test to determine the causal effects among four variables. We come up with a number of interesting results. First, we did not find statistical evidence neither for the inflation-output variability relationship nor for the Phillips curve. Second, we uncovered support for the positive causal effect of the inflation on its uncertainty and negative causal effect for the reverse direction. Additionally, we also found some support for the indirect negative causal effect of the inflation on the output growth. These results support the policy of low and stable inflation in the countries. Variability of grave goods in female burials in the middle La Téne period in Bohemia

Presented diploma thesis is devoted to the analysis of the female graves of the La Tène period in Bohemia, more particularly it relates to quantity, material and placement of every objects of the grave goods. Observed variability indicates time, regional and in some mesure social differences which influenced the composition of the grave set. Within the comparison, the graves from Moravia, Slovakia and Austria have been also taken into consideration.
Sociophonetic study of dysfluent behaviour in native English speakers

The present thesis focuses on the study of the influence of two social factors, age and gender, on the frequency and variation of seven different types of dysfluencies (filled pauses, silent pauses, repairs, repetitions, false starts, vowel and consonant lengthening) in the spontaneous speech of native English speakers from England. The speakers were divided into four different social groups according to their age and gender. The first part of the present thesis provides a general characterization of the relevant types of dysfluencies, together with the main issues concerning the production of speech dysfluencies. The empirical part presents the analyses of the recordings of 32 native English speakers from England. The overall results considering general influence of age and gender show that the only significant difference is between age groups, with older speakers producing more dysfluencies than younger speakers. Gender, on the other hand, does not make the difference significant, except for vowel lengthenings that were produced significantly more by female speakers than male speakers, and repetitions, which is the only type of dysfluency where the difference is significant and influenced by both age and gender, with older male speakers producing significantly more dysfluencies than any other...
Universal clothing and its use in museum education

In my thesis, built on the bachelor's thesis, I focus on the universal form of clothing. In the theoretical part I research the common features of garments in changing cultural and historical contexts. I focus on general aspects of clothing. I search for connections between functions and forms of clothing. I monitor the impact of historical styles on the work of contemporary clothing designers and I seek the universal features of today's fashion apparel. I observe the interest of children, teachers and also of publications in textile creation. In the art section I work on a series of proposals for universal variable apparel. By using patents it is created more models out of just one. I document and crtitically reflex the final results of possible model variations. In didactic part I suggest educational program for selected clothing exposition. The program includes worksheets and the posibility of using a universal garment as teaching tool. The program I verify in practice I also document and evaluate.
Structure- and sequence-based identification of functionally important amino acids in a protein family

A group of small GTPases consist of over twenty protein families in the super class P-loop. It has a very diverse cell functions. Small GTPases regulate the formation of vesicular follicles, cytoskeleton and nuclear transport. They participate also on cell proliferation and signaling. The aim of my work is to find important amino acids that define family and distinguish each other. I focus on families Arf, Rab, Ran, Ras and Rho. At the Rho family I am also devoted to classes Rho, Rac and Cdc42. Amino acids are identified using bioinformatic programs selected Consurf and Sca5. The objective is also to test P2RANK specialized tool developed at the Charles University in Prague that predict ligand binding sites from protein structure in different families. Founding amino acids can have a big role in the functional divergence of individual families and classes of small GTPases and can be the basis for future study example for the proliferation of cancerous cells. 1.1 Keywords Powered by TCPDF (www.tcpdf.org)
Comparison of difficulty and variability of fitness aerobic choreography in cadet and senior age categories - finalists of World Championship 2013

The topic of this thesis is the comparison of difficulty and variability of fitness aerobic choreography in cadet and senior age categories - finalists of World Championship 2013. The goal I set was to compare the difficulty and variability of these choreographies in regard to age, which influences the performance. Each choreography is analyzed in accordance with rules of FISAF fitness aerobic team and complemented by interview with an expert in the field. With the realized research we found out, that seniors choreographies are more difficult than cadets choreographies, the difference in the variability of choreographies of both categories is not significant. The results show the effort to improve choreographies, so differences between younger and older category are getting smaller in regard to possibilities of each age category. This work is part of a project focused on the study of the variability of human cytomegalovirus (HCMV) among clinical isolates with the aim to map the geographical distribution of HCMV genotypes, reveal the relationships between genotypes and the severity of HCMV-associated diseases, and identify regions in the HCMV genome with a potential for use as diagnostic and therapeutic targets. Attention was paid to the development of the methodology for the preparation of the material for next-generation sequencing (NGS) from HCMV clinical isolates and evaluation of the obtained sequencing data. Blood and urine samples collected from hematopoietic stem cell transplantat recipients and congenitally infected children were analyzed. Samples suitable for NGS were sequenced by the Illumina platform and sequences were created by de novo assembly followed by mapping assembly. Urine samples in comparison to blood samples had higher yield of material for NGS. Of the samples positive for HCMV DNA (7 of 50) after amplification in the cell cultures, only one sample had high purity of the viral DNA (98%) while six samples had purity of less than 7%.
