Current methods in protein-protein interactions research

Protein-protein interactions (PPI) have a crucial role in all processes in living cells. Understanding the interactions between proteins allows us to describe cell processes in more detail and their study opens new possibilities for drug design. The importance of the question of studying PPI is shown in the recent development of various methods for their identification and description. The aim of this thesis is to give an overview of new and improved experimental methods of identification and characterization of protein-protein interactions. Methods described in this thesis are divided in four chapters - proximity-dependent labelling methods (BioID, BioID2, APEX, TurboID, MiniTurbo, PUP-IT, AirID, SPPLAT, EMARS), cross-linking methods (XS-MS), fluorescence methods for identification and visualization (BiFC, FRET, BRET) and biophysical methods for description of kinetics and thermodynamics parameters of interaction (SPR, ITC, MT).
Interaction between hydrogenosomes and endoplasmic reticulum in Trichomonas vaginalis

Endoplasmic reticulum-mitochondria encounter structure (ERMES) is a protein complex tethering ER and mitochondria. ERMES consists of four core subunits - Mmm1, Mmm2 (Mdm34), Mdm10 and Mdm12. It was first discovered in Saccharomyces cerevisiae and most functional information is based on studies of this organism. ERMES affects mitochondrial distribution and morphology, participates in lipid trafficking and is important for homeostasis of the cell. In Trichomonas vaginalis, the human urogenital parasite, three genes for putative, highly divergent components of ERMES complex were predicted. However, the cell localization of these proteins and their function is unknown. This thesis is focused on investigation of ERMES components in T. vaginalis, their cellular localization, interactions between components and identification of their possible interacting partners.
The phenomenon of a stranger in male and female culture

Modern society does not support variety. Globalization proves this and it affects gender, as well. Society tends to negate variety and reacts inappropriately - by segregation, ridicule, suspicion, etc. Variety is still understood as something of little value. This work studies the issue of variety in gender, whose problems it compares to problems of a stranger. It analyzes both issues in the perspective of modern Western society and offers the analogy of gender issue and the issue of a stranger. This analogy is outlined in three major areas of social life: family x society, specific male and female occupations, and partner relationships. Our work especially stresses tolerance and willingness to find what is realistic in contrast to prejudices. Any kind of discrimination not only disturbs the discriminated but also the discriminating. The obscurity of gender roles in modern times and difficulties in understanding these roles lead to negative effects for both sexes, including a lack of freedom.
The specificity of male and female culture in the basic life areas (focused on contemporary occidental culture)

Male and female "culture" has many advantages and disadvantages in both the private and public sphere. Our society focuses attention on these advantages and disadvantages and thus invokes an atmosphere of two competing worlds. Competition is common in the western world today, but applied in the gender area, it does not help to create a more quality coexistence. The perfect perviousness of the rights and responsibilities throughout the male and female cultures, which is being theoretically promoted by current society, does not solve the problem of the question of gender. Moreover, the perfect perviousness itself is not even possible. This text suggests the way of respect and reverence to an individual and his or her originality and evaluation of one's work based on other criteria than one's salary and earnings. Hence I consider partnership as a productive area which offers a different perspective in perceiving the opposite sex. The possibility of a unique perception exhibited in a more tolerant attitude bordering with awe to differences of the opposite sex is visible not only in the initial state of being in love but also in the state of intimacy and mutual recognition of a partner's needs in later phases of the relationship. Men and women in men's culture: with a focus on education and professional life

Hned v úvodu bychom chtěli předeslat, že ač by se mohlo podle názvu práce očekávat, že bude zaměřena především na obhajobu žen a jejich nevýhodné pozice v mužské společnosti, nebudeme se věnovat pouze tomuto faktoru. Budeme se snažit poukázat především na rigiditu dnešní společnosti, která si od zástupců obou pohlaví nárokuje plnění stále stejných tradičních rolí bez ohledu na moderní posun většiny ostatních složek tvořících společnost. Nepůjde tedy výhradně o obhajobu žen, ale poukážeme i na nevýhody, které z pravidel "mužské" společnosti plynou mužům samotným. Muže a ženy budeme tedy prezentovat v určitém smyslu v rovnoprávném postavení - obě dvě pohlaví totiž podléhají kulturním normám a stereotypům. Chtěli bychom na tuto problematiku nahlédnout s určitým odstupem a neutralitou. Účelem této práce by měla být snaha o vyvarování se jakékoli zaujatosti. Zachování odstupu a racionality je pro nás v této práci cílem, protože svou stálou ožehavostí láká toto téma ke zpracování nové a nové představitele, především speciálně pro tuto oblast vyhraněných vědeckých a politických směrů, které se genderem a problematikou koexistence mužů a žen ve společnosti zabývají primárně (např. gender studies, různé druhy feminismu ... ). Představitelé takovýchto směrů mají však už výběrem svého zaměření často poněkud zúžený...
