[cze][eng] Protein-proteinové interakce (PPI) hrají nepostradatelnou roli ve všech procesech v živých buňkách. Pochopení interakcí mezi proteiny nám umožňuje bližší popis buněčných dějů a jejich pochopení otevírá nové možnosti i pro návrh léčiv. Důležitost otázky studia PPI se odráží v recentním rozvoji různorodých metod pro jejich identifikaci a popis. Cílem této práce je sumarizovat poznatky o nových a zdokonalených experimentálních metodách identifikace a charakterizace proteinových interakcí. Metody popsané v této práci jsou členěny do čtyř kapitol, metody blízkostí indukovaného značení proteinů (BioID, BioID2, APEX, TurboID, MiniTurbo, PUP-IT, AirID, SPPLAT, EMARS), využití síťování proteinů pro jejich identifikaci a strukturní charakterizaci (XL-MS), fluorescenční metody identifikace a vizualizace (BiFC, FRET, BRET) a biofyzikální metody pro stanovení kinetických či termodynamických parametrů interakce (SPR, ITC, MT).Protein-protein interactions (PPI) have a crucial role in all processes in living cells. Understanding the interactions between proteins allows us to describe cell processes in more detail and their study opens new possibilities for drug design. The importance of the question of studying PPI is shown in the recent development of various methods for their identification and description. The aim of this thesis is to give an overview of new and improved experimental methods of identification and characterization of protein-protein interactions. Methods described in this thesis are divided in four chapters - proximity-dependent labelling methods (BioID, BioID2, APEX, TurboID, MiniTurbo, PUP-IT, AirID, SPPLAT, EMARS), cross-linking methods (XS-MS), fluorescence methods for identification and visualization (BiFC, FRET, BRET) and biophysical methods for description of kinetics and thermodynamics parameters of interaction (SPR, ITC, MT).
