Changes in Lipid metabolism in acute diseases

Changes in lipid metabolism in acute diseases. Introduction. Abnormalities in lipid metabolism have been described in critically ill patients repeatedly. While triglycerides behave unpredictably, decrease of cholesterol levels is a typical finding. Further, low levels of cholesterol in critically ill patients have been found to be associated not only with poor prognosis but also increased mortality. Cholesterol is derived in two ways: via de novo synthesis and via dietary absorption. Both the modalities can be monitored by the determination of CH synthesis markers (lathosterol, squalene in blood plasma), and cholesterol absorption markers (sitosterol and campesterol in blood plasma) Material and method: In the first project sixty patients with acute internal disease were involved and their lipid profile was evaluated. Second and third projects have solved the same problem in the group of patients with active Cohn's disease (n=24) and acute upper gastrointestinal bleeding (n=24). Serum concentrations of total, LDL and HDL cholesterol and triglycerides were measured in an enzymatic automated system. Non-cholesterol sterols (lathosterol, squalene, sitosterol, campesterol) were analysed using gas chromatography - mass spektrometry. Influence of Diet on Blood Lipid Levels in Dyslipidemic Patients

The bachelor thesis is focused on the relationship of eating habits and dyslipidemia. Its aim is to find out whether diet influences blood lipid levels. The paper is divided into a theoretical and a research part. The theoretical part handles with lipids of the human organism in general, their metabolism and the risks of atherosclerosis. The research part describes work with patients who suffer from lipid metabolism disorders. The patients had a diet consultation and, two months after, they had their blood drawn to see the results of cholesterol and triglycerides levels. The patients also filled in a questionnaire concerning their eating habits. It turned out that nutritional intervention had resulted in an average decrease in triglycerides and total as well as LDL-cholesterol levels. However, HDL-cholesterol level decreased as well. Furthermore, the questionnaire enquiry showed that dyslipidemic patients do not, in general, disobey recommendations of the dyslipidemic diet. The results of the paper showed that dyslipidemic patients follow recommendations of a healthy diet. However, there is still space for improvement. The role of cholesterol-7alpha-hydroxylase in cholesterolemia regulation

Cholesterol 7α-hydroxylase (CYP7A1) is an enzyme catalyzing the first step of conversion of cholesterol to bile acids. The enzyme activity is regulated to supply enough bile salts necessary for absorption of fats in the intestine. In some species it contributes to cholesterol elimination from the body when dietary cholesterol intake is high and, in such a way, protects against the development of hypercholesterolemia. CYP7A1 activity can be therapeutically affected by administration of bile acid sequestrants that increase the enzyme activity and thus lower cholesterolemia, and also by administration of bile salts. The enzyme deficiency in humans results in hypercholesterolemia. Several single nucleotide polymorphisms were identified in gene encoding CYP7A1 in humans. They form three large haplotype blocks. The most attention has been paid to the -203A>C polymorphism that has an impact on cholesterol and lipoprotein concentrations, on the response of cholesterolemia to dietary intervention and on the response to hypolipidemic drugs. Awareness of overweight people and people without overweight on the risk of cardiovascular dise

This thesis deals with awareness of overweight and non-overweight people about of the risk of cardiovascular diseases. The goal is to determine the level of knowledge of both groups in the section of cardiovascular diseases and their interest in this issue. Next goal is to determine the degree of difference in awareness in this issue between the sexes. The theoretical part provides information about obesity (causes, symptoms, complications, treatment). It also deals with frequent cardiovascular diseases - hypertension, atherosclerosis, myocardial infarction, ischemic heart disease, cerebrovascular accident, and dyslipidemia. One of chapters is devoted to projects that address prevention of obesity and cardiovascular diseases. The practical part of work describes examined group that participated in the survey. Analysis of the results shows that people who are not overweight or obese are more informed about the risks of cardiovascular disease than overweight and obese people, because they are less interested in this issue. Women are better informed in the section of cardiovascular diseases than men. 14 individuals out of 89 members of research is interested in the issue of cardiovascular diseases. Pathogenesis of hypercholesterolemia in Prague hereditary hypercholesterolemic (PHHC) rat

Hypercholesterolemia represents a major risk factor of cardiovascular disease. A number of experimental models is used for study of hypercholesterolemia pathogenesis and therapy. This thesis concentrates on characterization of one of these models. Prague hereditary hypercholesterolemic (PHHC) rat is such a suitable model for study of hypercholesterolemia. Although the majority of plasma cholesterol is transported by high density lipoprotein in PHHC rat fed standard diet, PHHC rat fed cholesterol diet develops hypercholesterolemia comparable to that of humans. The advantage of this model is that hypercholesterolemia develops without the need for addition of bile acids or other hepatotoxic substances to the diet. The hypercholesterolemia of PHHC rat is caused by slowed down catabolism of cholesterol-rich very low density lipoprotein (VLDL). These cholesterol-rich particles are synthesized in the liver. We found out that PHHC rat fed 1% cholesterol diet accumulates cholesteryl esters (CE) in the liver and also in the VLDL. Acyl-CoA:cholesterol acyltransferase (ACAT) and microsomal triglyceride transfer protein (MTP) may participate in the increased incorporation CE into VLDL. Ecotoxicity of cholesterol-lowering-drugs

This bachelor thesis is focused on the ecotoxicological testing of blood cholesterol lowering drugs. At first it considers the issue from a theoretical point of view – describes the effect of residual drugs on the environment, the distribution of hypolipidemics, the risks of elevated cholesterol, then deals with ecotoxicology and introduces the principles of ecotoxicological testing. In the experimental part, these following drugs were tested: Corsim, Atoris, Lipanthyl Supra, Lipanthyl 267M. In addition, clofibric acid – the metabolite of fibrates was tested. Residues of these drugs are found mainly in the aquatic ecosystem, that is why tests of aquatic organisms have been used to evaluate ecotoxicity. Test organisms were Thamnocephalus platyurus, Lemna minor and Vibrio fischeri. LC50, EC50 and IC50 values were determined on the basis of test results. Using these values, the ecotoxicity of the above mentioned hypolipidemics was evaluated.
Mixed lipids and their interaction with biopolymers

This thesis focuses on the study of interactions between hyaluronan and lipid DOTAP and its mixtures with DPPC in water. First, the aggregation behavior of the lipid itself and its mixtures was measured, afterwards the influence of hyaluronan in different concentration was studied. Further the fluorescence anisotropy of fluorescence probe DPH in these mixtures and influence of cholesterol was measured. These properties were investigated by fluorescence spectoscopy using pyrene and DPH as fluorescence probes. The measurements revealed that the addition of hyaluronan has observable influence on the aggregation behavior of the lipid DOTAP and its mixtures with DPPC and that cholesterol influences liposomes fluidity.

Specifically, is concerned with the characteristics of fats, their technological processing and importance in human boarding. Detail is focused on finding specific knowledge about fats among selected adults. The aim of this work is to clarify the extent to which selected adult overview of fats and what have the knowledge, which in turn can use in your everyday life when choosing foods and preparing meals. At the same time also to provide input for knowledge of fats, which are subsequently used in further work, which will focus on a different target group namely students from secondary schools and grammar schools. For this purpose was performed a questionnaire survey among selected students of secondary education and its results are discussed in the conclusion of this theses.
