Title of diploma thesis: Study of expression of biotransformation enzymes and transport proteins in kidneys Xenobiotics are a part of the everyday life of living organisms. Their source is, among other things, diet, environment, administered medicines, food colorings and others. The living organisms have developed complex metabolic processes that eliminate these substances from the body in order to protect themselves against their negative influence. Within this work, we specifically focused on the effect of the murine Car agonist - TCPOBOP on influencing the expression of biotransformation and transport genes. The essence was the isolation of mRNA from the murine kidneys using the classic phenol-chloroform method, followed by the transcription of mRNA into cDNA. In the last step, the RT-PCR reaction was performed. The results are graphs showing how the administration of TCPOBOP affected the expression of the monitored genes. It is clear from these graphs that TCPOBOP has some effect on the expression of target genes. Lipid emulsions in lonf-term parenteral nutition

The combination of intestinal failure and parenteral nutrition is a condition associated with mild chronic inflammation and organ injury, the most significant of which is hepatic injury. This crossover-controlled trial investigated the effects of different intravenous lipid emulsions (IVLE) enriched with increased doses of omega-3 polyunsaturated fatty acids (n-3 PUFA) from fish oil (FO) in patients on home parenteral nutrition (HPNP) for chronic intestinal failure. Twelve patients were consecutively given three different IVLEs, with an FO emulsion added after each IVLE. The study also included twelve paired healthy controls for comparison. The aim of this study was to investigate how long-term administration of these IVLEs affected primarily the erythrocyte and plasma fatty acid (FA) spectrum, as well as the inflammatory response, antioxidant status, markers of liver function and bile acid spectrum in HPNP, and how these changes differed from healthy subjects. We demonstrated successful incorporation of n-3 PUFA into erythrocyte phospholipids and corresponding changes in the plasma FA spectrum, characterized in particular by significant increases in n-3 versus n-6 PUFA, with a decrease in the n-6/n-3 PUFA ratio. Interpretation of temporary structures in the context of activation of abandoned sites

Temporary use serves as a useful tool in a process of activation of unused buildings. The final thesis puts the phenomenon of short–term function in a centre of attention and it examines successful application of transient use in a strategy of restoration of abandoned buildings. Principles of temporary use define an opportunity to test new ideas in the process of designing long–term use. A newly established database on www.archipop.cz is based on an analysis of case studies of temporary use. The publicly accessible database provides a list of selected projects that successfully activated neglected spaces via temporary use. Many of these projects are managed by a temporary planning institute. General goals of an institute, which sets up principles of temporary use in close cooperation with municipalities, are summarized in an individual chapter. The acquired knowledge of the process of analysis of principles of temporary use was verified in a practical task. The assignment was to create a strategy for an activation of an unused complex, specifically a site of a former paper factory in the Spanish province of Pontevedra.
Specifics of a family with a handicapped child

The diploma thesis is focused on researching the impact of a disabled child on the family from birth to adulthood and follows the specifics of a family with a disabled child. It is about the effect on the family as a whole, as well as the effect on individual family members and the relationships between them. An important aspect is also the impact on the formation of relationships between siblings and relatives, as well as the influence on the social environment of the family. In the theoretical part, the thesis deals with basic concepts such as family and its functions and factors affecting it, disability, types and consequences of disability. In the practical part, the work focuses on researching the impact of a disabled child on the family from birth to adulthood. The goal of this work was the best possible understanding of the impact of the arrival of a disabled child in the family and the impact of the newly created situation on family members. The main topic is to focus on their states and reactions in the individual stages of life with a disabled child and to find out how and in what form it is possible to help eliminate the negative effects of a disabled child on individual family members and their own lives. Cooperation between a teacher and a teaching assistant at the first stage of primary school.

The diploma thesis deals with the issue of cooperation between a teacher and a teacher's assistant at the first level of elementary school. The theoretical part defines the role of the teaching assistant, the prerequisites for good cooperation between the teacher and the teaching assistant, and the personality requirements of the teaching assistant. The goal of the thesis is to clarify how the cooperation between these two teaching staff currently takes place, including various possibilities for mutual cooperation, which can subsequently positively affect the effectiveness of teaching. In the empirical part, a qualitative scientific method called a semi-structured interview was used. Data were collected from both teachers and teaching assistants at the first level of elementary school. Data saturation was supported by content analysis of school documents. Data analysis was conducted through open coding and other methods appropriate for qualitative scientific methods. International Commercial Mediation in the Context of the Singapore Convention Abstract The thesis deals with the topic of mediation in private international law, focusing on the enforceability of mediation agreements arising from international commercial disputes. The thesis is focused on the Singapore Convention, which was signed in August 2019, and marginally on the related UNCITRAL Model Law on Mediation. Although the success of the Singapore Convention will depend largely on the political actions and motivations of individual states, in order for it to be used by private actors, an analysis of the construction of the international norm in question is necessary. The aim of this work is to answer the question whether the regulation of mediation in international commercial law has its justifiable importance. In order to answer this question, the an analysis of the current legislative framework and the conditions for the performance of mediation agreements in international conventions and other international source sis performed, in relation to the EU and the Czech law. At the same time, unclear issues of the legislation in question are highlighted, also with regard to the transposition of EU regulations into Czech legislation, and possible ways of bridging them. Family of child with Kabuki synrome

The thesis is about a family of a child with Kabuki syndrome. The first chapter deals with the syndrome itself, diagnosis, clinical symptoms and cognitive functions. The second chapter describes the family with the rare condition, the family in the context of the history and what stages the family of the child with the rare condition goes through. The next chapter discusses support for the family with a rare disease, and also includes the concept of a non-profit organization and an association. The practical section explores the family of an individual with Kabuki syndrome, and includes an interview with a family member. The research section is supplemented with a case study.

It is a new family house with workroom which has its own entrance. The house is situated in the Pardubice region in the town called Česká Třebová, cadastral area Česká Třebová. The building consists of three floors. Two of them are aboveground and one is underground. In the basement there is a garage suitable for two cars. The family house with workroom is covered with wooden saddle shape trusses. The building is designed from the structural system Heluz and contact insulation ETICS.
The Body (Intimity)

With this set I am following my previous work in which I was occupying with woman body. I catch body in intimate moments – but I encode it, so it is almost not obvious. I use light irony and exaggeration in my pictures. In this set I work with woman body as a object of voyer, who observes the woman with a peephole, lense, field glass or camera. But I keep it all in abstract and stylized position. I work with code, thanks to which we at first perceive the picture as colourful and shaped painting forms that gradually show womans body. I use bigger ammount of abstract, when forms of the body balance at the edge between figural and abstract. I enjoy the moment when there is not everything visible at the first sight in the picture. I want the viewer to look for the body, observe it and explore it. I do not aim for catching the beauty of the body or is real visual shape. I work with light irony of voyeur looks by using some deformations of the body parts.
