Geometry and stability study of nucleic acids distinctive single-strand motifs

Subject of this thesis is the regulatory DNA segment, the so-called Serum Response Element (SRE), which possesses highly specific structural characteristics and its structural flexibility plays probably a key role in its physiological activity. Structural properties of 16-mer SRE segments capable to form hairpin structures were studied by means of accurate measurements of temperature dependent Raman spectra. Due to the close connection of oligonucleotide chain structural stability with the stacking interaction, temperature dependent Raman and UV absorption spectra were measured on dinucleotides serving as a simple model system for the stacking investigation. The measurements results processed by factor analysis provided characteristic spectral patterns and confirmed that stacking interaction depends on the base order. By fitting the thermodynamic equilibrium equations to the factor analysis outputs, the thermodynamic characteristics of the thermally induces structural changes in hairpinforming complexes were estimated and difference spectra characterizing opened and closed hairpins, which enable the further specification of the geometry of these complexes, were obtained.

The influence of the antiquity on the oldest French poetry

The dissertation focuses on comparison of medieval writings presenting the actions of Julius Caesar to French readers, specifically Li Fet des Romans, Roman de Jules César and Li Hystore de Iulius Caesar. Interpretation of accessible texts is based on analysis of the translation process: comparison of the antique original and its medieval variants in French language as presented in medieval manuscripts. The author opens the dissertation with introduction to factors influencing the medieval translator; among others the ability of complex understanding of the original text, education and awareness of other literary works, oral component of culture; and the way of thinking and of interpretation of the world.

Possible ratings for photographs

This thesis addresses the question of the perception of photographs. It is based on the work of R. Greene and A. Oliva (2008) and further expands it. These authors investigated which global properties people identify on photographs of natural scenes and how consistent their evaluation of these properties is. In this paper I used general photographs without any thematic limitations. In the first part of the research I examined what global properties people determine during a quick viewing of a general photograph. The obtained general properties (movement limitation and fun) were added to the properties found in R. Greene's and A. Oliva's work. In the second part of the research I used a computer program for hierarchical grouping to determine how consistent the rating of these global properties is when different people rate the photographs. The resulting consistency is expressed as the average of all interobserver correlations for a given property. The experiment revealed that the transience property has the highest consistency rate (r=0,516), followed by movement in the scene (r=0,457) and openness (r=0,450). Interpersonal dynamics in dubbing of TV series: dominance, verbal and non-verbal action of characters

So far the filmic dialogue and its translation have been researched mainly from the perspective of the translator of the script, this approach being justified by the claim that the translator cannot manipulate any other but the verbal code. The nature of the audiovisual text, whether in its original or dubbed version, is, nevertheless, explicitly multimodal, and its complex meaning is coded on multiple level via diverse means. An important factor which makes audiovisual research complicated is the fact that the audiovisual text is essentially a teamwork determined by collaboration of many different people in different stages of its production. This thesis attempts to approach the audiovisual text of as a whole, that is, in the form it is presented to the audience. The opening part identifies the specifities of audiovisual text and its dubbing, including its multidimensionality, role and function of its dialogue, process of dubbing production and constraints of the contemporary Czech dubbing market. The role of eIF3 in mRNA recruitment to the 43S pre-initiation complex

Translation initiation in eukaryotes plays an important role in gene expression and relies on interactions between many eukaryotic initiation factors, small and large ribosomal subunits, mRNA and initiator tRNA. The largest of these initiation factors, the eukaryotic initiation factor 3 (eIF3), participates in the most of translation initiation reactions. In yeast, eIF3 occurs together with eIF1, eIF5 and ternary complex (TC, eIF2-GTP-Met-tRNAi Met complex) in the multifactor complex (MFC) and promotes formation of the 43S preinitiation complexes. eIF3 also stimulates mRNA recruitment to the 43S preinitiation complexes, scanning the 5' untranslated region of mRNA for AUG recognition and has an important role in the gene-specific translational control mechanism called reinitiation involving short upstream open reading frames (uORFs).

The complex of wabet and court in temples of the Thirtieth Dynasty and the Ptolemaic and Roman period

The study at hand focuses on an architectural complex or ensemble that regularly occurs in temples of the Thirtieth Dynasty and the Ptolemaic and Roman period. This ensemble generally combines an open court with an elevated and covered chapel, and is referred to as 'court' and wabet, respectively. The present study aims to bring together the common characteristics and essential components of all ensembles of wabet and court in order to get a better understanding of the ensemble's role in the temple and its development over time. Following a brief introduction to the thirteen temples that appear to contain this ensemble in their core and to the complexes themselves (chapter 1), the following chapters focus on the ensemble's location, orientation, layout, and chronology (chapter 2), the designations in use to refer to this complex (chapter 3), a description and analysis of the texts and reliefs applied to its walls (chapter 4), and a study of the origin and development of this decorative scheme (chapter 5) in order to gain a better insight into the activities performed in the complex and its role within the temple. The text deals with creation of the new type of institution, so-called Family Centre. Its aim is to contribute to the integration of disabled people into the society. The Family Centre will be an open institution, where families with disabled children will have an opportunity to meet each other. The idea of the integration will be realized in the form of café with playroom which will be adapted to needs of disabled children. The special educational care for children with various kinds of disability will be ensured. Next, the paper deals with particular parts of programme which The Family Centre will offer to its clients.
