The development of Leishmania in the insect vector of the genus Phlebotomus
Kvapilová, Kateřina ; Volf, Petr (advisor) ; Bilej, Martin (referee)
The work was focused on the study of various aspects of development of Leishmania in vectors of genus Phlebotomus and can be divided into two main parts. In the first part, we studied the competition of Leishmania major and L. turanica during their development in the sand flies who are natural vectors of L. major using the experimental co-infections of fluorescently marked promastigots of these two species. While both leishmania species developed similar in the intestine of Phlebotomus papatasi, L. turanica prevailed in P. duboscqi in the late stages of infection. The fluorescent marking of Leishmania should allow us also to study possible genetic exchange between species at different stages of Leishmania infection (2nd, 9th and 14th day after the infective feeding). Using the flow cytometry (FACS) we have repeatedly identified dozens of objects emitting red and green signals in the intestinal homogenates of co-infected sand flies, however further analysis with confocal microscope disproved these objects as the hybrid promastigots of Leishmania. In the second part of this thesis we investigated the role of L. major genes HASP and SHERP, which lies on the locus LmcDNA16 and are expressed exclusively in metacyclics. We used mutant lines KO (lacking the locus LmcDNA16) and HASPB (KO line with gene...

Kvapil, Jan ; Stromšík, Jiří (advisor) ; Petrbok, Václav (referee) ; Kurzke, Hermann (referee)
The Dissertation considers the Catholic song propaganda of the early modern times. Its research is based on Bohemian literate and musical sources. The basic methods of the song-propaganda were actually invented by Protestants. The Catholic reformers were able to react to its influence through their own song-propaganda (for instance Leisentrit and Jesuits) and modernized and developed their own new methods with just a minute delay. The song-propaganda was an outstanding device for indoctrinating common people anyway because of their analphabetism. The main attention was focused on persuasive methods used in the songpropaganda by Catholic missionaries. These methods can be classified as follows: The repetitive method was the simplest one based on a simple reproduction of fundamental Christian texts (Lord's Prayer, Credo, Ave), religious dogmas (purgatory, confession, communion) and ideological doctrines (singularity of the Catholic Church) in a musical form. It was mainly applied in the catechesis. The songs inspired by this method were mainly addressed to children and youth. The exemplary method gives either positive or negative example people should or should not follow. These songs narrated mostly a life of a exemplary saint. They were very popular among children and therefore applied in catechesis. A more...

The purpose of the paper "Valentín García Yebra and his Contribution to Translation Studies" is to provide an overall analysis of García Yebra's works and their reception in Spanish press and magazines specialized on translation. Part of the paper is dedicated to the summary of García Yebra's predecessors on this field, to his biography and his career as a professional translator. But the major emphasis rests upon the analysis of his thoughts on theory, critique and history of translation. As a part of the analysis, we provide a summary of all the books in question. The purpose of the analysis is to determine the most important areas of García Yebra's thoughts and to synthesize them into thematic units. The most significant areas contain the definition of translation and some general ideas on the role and possibility of translation. Next, we deal with the types of translation, the two-phase process of translation and the factors affecting it. We also study the function of linguistics in García Yebra's approach to translation. The following sections deal with the investigator's approach to dynamic equivalence, with the role of translation in enriching a language and with the education of translators, which was one of his life's missions. Next sections of the analysis are dedicated to the problems of...

Kopecký, Tomáš ; Skalický, Milan (advisor) ; Jan, Jan (referee)
The educational trail Dobrošov fort is situated in the village of the same name. It is an advance specified marked route which leads in the footsteps of the border fortification from 1935 - 1938. The length of the route is approximately 4 km and visitors are mainly informed about historically interesting places in the surrounding and about the events before World War II. Historical affairs are presented in the form of information panels at individual stops. The panels tell visitors not only about historical military facts of 20th century but also about the building of the fortress fortification Dobrošov. The educational trail has eight official stops. On the route, visitors can see additional panels with the description of the light fortification and they learn interesting things about the surrounding. The content of this thesis is a brief characteristic of events in Czechoslovakia before World War II. There is a description of the fortification system of a fort at that time in Europe and Czechoslovakia. Furthermore, this thesis deals with educational trails in general from the viewpoint of their functionality and equipment. This study points out legislation and illustrates new trends of this form of tourism. The aim of this thesis was to document the educational trail Dobrošov fort, with the focus on historical events of fortification building in Czechoslovakia before World War II. The analysis of the trail and information panels was conducted. A questionnaire survey found out the needs of visitors and highlights the usefulness of this trail. The questionnaire survey had another important role for findings of stated hypotheses. Four scientific hypotheses were formulated. The first one is if the access to more objects increases, its attractiveness and attendance will also increase. The second hypothesis is whether the visitors are satisfied with the infrastructure of the educational trail. Furthermore, the awareness of the educational trail helps to its high attendance, and both citizens and tourists will be happy about a fortification extension which also includes the preservation of decaying objects in the surrounding. The used methods for achieving these aims were the observation and questionnaire survey in the location of the educational trail. The conclusion shows that the access to more objects will increase the attractiveness and attendance of the trail. However, the visitors are not satisfied with the infrastructure of the trail. The awareness of the educational trail contributes to the high attendance. The hypothesis that local citizens and tourists will be happy about the trail extension which includes the preservation of decaying objects in the surrounding was disproved.
