Mucin hydrogels - artificial models of native mucus systems

The scope of this masters thesis is the preparation of a model mucin system and its utilization as an artificial model of the native mucus system. The creation of this model system, according to several designed methods was a part of experimental part of the thesis. The preparation of mucin system comprised of physical and chemical methods of hydrogel formation, screening and characterisation of the various physical conditions of the mucin properties on its molecular level, and the preparation of sorbent with sorption surface containing mucin. Methods of light scattering, namely dynamic light scattering (DLS), used for mucin particles size change monitoring, and electroforetic light scattering (ELS), used for Zeta potential change monitoring, were used for the screening of the impact of physical factors on the properties of mucin.For the characterisation of impact of the temperature on changes in mucin sctructure was, apart from monitoring of light scattering, used also a diferential scanning calorimetry (DSC), which registered temperature value, at which mucin thermal denaturation occurs. In the next part of the thesis we subdued the created sorption surfaces to various physical-chemical analyses, which task is the characterisation and projection of surface and confirmation of mucin presence.Substancial part in monitoring and characterisation of changes in surface sctructure of sorption surface was accomplished by Fourier transform infrared spectroscopy (FTIR). Scanning electron microscophy (SEM) was used for the final, more detailed, projection of the mucin enriched, sorbent surface structure. Suggested methods of mucin hydrogel, didnt prove sufficient results for the possibility of application of hydrogel as a artificial model of real mucus system, but the sorbent application was indicated as a suitable alternative and an instrument for the further mucin behaviour research and possibly subsequent bacterial adhesion, which represents the first step in the formation of the bacterial biofilm.
The role of CRL4 ubiquitin ligase complex in homeostasis and regeneration of gastro intestinal tract

Ubiquitination represents a central regulatory mechanism for protein function utilization. Dysfunction of ubiquitin ligases is associated with the development of various pathological conditions. However, the full complexity of the ubiquitinating network in tissue homeostasis and carcinogenesis remains to be elucidated, limiting its potential as a therapeutic target. In my PhD project, I aimed to reveal the role of E3 ubiquitin ligases, particularly CUL4A, in the alteration of regulatory pathways that result in gastrointestinal (GIT) homeostasis disorders and tumor expansion. By using a novel semantic biclustering technique, we performed expression profiling of the E3 ubiquitin ligases and uncovered the potentially redundant features of GIT-specific ubiquitin ligases based on their tissue-specific expression and ontological relationships. Testing the identified compensatory network on the mouse model showed that genes from the same ontology group simultaneously altered their expression pattern after induced epithelial damage, exposing their complementary role during tissue regeneration. Atresia of gastrointestinal tract and possibilities of physiotherapeutic intervention

This bachelor thesis deals with atresia of the gastrointestinal tract and possibilities of physiotherapeutic intervention. The thesis is presented in the form of a review with a case study. The review part contains information about the development and anatomy of the parts of the gastrointestinal tract that are most often affected by atresia. The classification, diagnosis and surgical treatment options of individual atresia are also mentioned. The options of physiotherapy intervention include scar treatment, respiratory physiotherapy, physical therapy, basal stimulation and orofacial stimulation. The practical part of the bachelor thesis is presented in the form of a case study of a patient after gastrointestinal atresia surgery. The content includes the examination before and after the therapy in the physiotherapist's ambulance and the follow-up after 2 months of regular therapy. The positive effects of physiotherapy are documented by photodocumentation and by using the Alberta Infant Motor Scale to evaluate motor development. The practical part also includes an educational handout for parents about the implementation of respiratory physiotherapy after discharge from hospital, which will be subsequently used in the Department of Paediatric Surgery at FN Motol.
Circadian regulation of digestive system function and its disturbances due to external factors

Circadian clocks regulate the behavior of cells in living organisms in 24hour periods. They react to environmental influences and, by synchronizing with them, ensure optimal cell function and the overall health of the organism. Biological clocks in the gastrointestinal tract and their functions can be influenced by several factors, including the timing of food intake or the influence of the microbiome. The correct timing of food intake can be essential in maintaining the functional physiology of the organism as well as the appropriate composition of the gut microbiome. Disruption of the synchronization between these factors and the biological clock in the gastrointestinal tract leads to disharmony of the organism, its improper function and the development of disease. The timing of food intake and the cultivation of an appropriate microbiome is critical to maintaining optimal health and should not be neglected by the public.

The effect of gut microbiota composition on condition in birds

The aim of this thesis is to summarize present knowledge concerning the effects of gut microbiota composition on condition in birds. The effect of gut microbiota on various aspects of animal physiology is currently intensively studied and the understanding of its biomedical importance is a topical theme. However, the majority of the current research is focused on humans and other mammals, and in birds on agriculturally significant species. Therefore, this thesis focuses also on the applicability of results of agriculturally orientated research on free-living birds' study. The results of agricultural research show a high interspecies and intraspecies variability in the effects of certain bacteria on the condition of the host. That should be taken into account when designing experiments in wild-living species. This thesis summarizes current knowledge concerning the effects of bacteria on condition of birds that was gained in correlational and manipulative studies. Manipulative studies include those in which probiotics or antibiotics were given to birds. The main modes of action of probiotic bacteria are also summarized here. Expression of ubiquitin ligases in gastrointestinal tract

Ubiquitin (Ub) ligases are important regulatory and signalling molecules, which are involved in majority of cellular processes such as differentiation, DNA repair, and regulation of energetic metabolism or immune response. E3 Ubiquitin ligases are also responsible for pathophysiological changes in the organism and their activity is associated with many human diseases including cancers. This makes E3 Ubiquitin ligases to be new diagnostic markers and interesting pharmaceutical targets. Based on previous studies, these enzymes evince very specific expression in the level of tissues or cell populations. Determination of this specific expression is important for a better understanding of their biological function. In this diploma thesis we systematically screened presence of 370 genes of E3-Ub ligases in gastrointestinal tract under physiological conditions and during acute inflammatory damage of distal colon. Obtained data allowed us to select genes, which can play important role in homeostasis as well as pathophysiology and regeneration of gastrointestinal tract. The role of the gastrointestinal hormones in the control of energy homeostasis

Zachování tělesné homeostázy vyžaduje precizní komunikaci mezi všemi buňkami organismu. Velkou měrou k tomu přispívají gastrointestinální hormony, které jsou významnými signálními molekulami a podílejí se na distribuci i zpracování přijatých živin. Každý h typem buněk a jeho sekrece je regulována v závislosti na přítomnosti konkrétních nutrientů. Vzhledem rozdílné anatomii jednotlivých částí gastrointestinálního traktu, je lokalizace buněk produkujících šná. Účinky gastrointestinálních hormonů jsou rozmanité a hrají důležitou roli energetické kontrole organismu. Některé hormony (gastrin) působí jen lokálně, naproti tomu jiné (inkretiny, ghrelin) jsou zapojeny do signalizace přes centrální i periferní ne do signalizace nervové soustavy umožňuje těmto hormonům podílet se na regulaci pocitu hladu a mnoha onemocněními. V důsledku nevyváženého příjmu živin (například při nadměrném příjmu lipidů nebo sacharidů) dochází narušení některé ze složek hormonálního regulačního systému a následně vzniku onemocnění. Nejznámějšími onemocněními, vzniklými v důsledku nutriční a následně hormonální dysbalance, jsou například inzulínová rezi hormony je naprosto klíčová pro zachování lipidového metabolismu i glukózové homeostázy. Investigative methods of gastrointestinal tract

The bachelor thesis deals with investigative methods of examination of the gastrointestinal tract and their frequency in a given period carried out in the Hospital of Milosrdní bratří Brno, Znojmo Hospital and the Hospital of Uherské Hradiště a. s. It is graphically processed amount of medical services carried out during 20092014. Data were obtained from the database od radiodiagnostic department. It also to evaluates the work of radiological assistant during these medical examinations. I tried to point out that the basis of good results of individual tests is not only professionalism, both theoretical and practical knowledge of radiology assistant, but also sensitive and helpful and respectfull psychological support to the patient, which may fundamentally affect the test results.
