Structure and properties of bacterial biofilms

This thesis deals with the study of basic properties of cells of bacterial biofilm produced by PHA producing bacteria, specifically Burkholderia cepacia and Burkholderia sacchari. Two cultivation approaches were used to compare different types of cells: at first, the cells were cultivated in bioreactor providing both planktonic and biofilm attached cells, secondly, cells grown on Petri dishes were also included in the study. In experimental part the affinity of each type of cells to hydrophobic phase was studied using BATH method. It was found that sessile cells, either biofilm associated cells as well as cells taken from Petri dishes, prefer hydrophobic phases more than planktonic cells. Also size and Zeta potential were measured using Dynamic Light Scattering (DLS). It was found for studied samples that, cells in these samples tend to aggregate and the difference of size between both types of cells occurs only for biofilm of the strain Burkholderia cepacia. Finally the impact of presence various bacterial cells types on surface tension of water as well as amount of PHA cumulated in different types of cells were observed.
The effect of magnetic field and other selected stressors on physiology of bacterial cells

This thesis deals with the effect of magnetic field and organic substances, namely benzene and p-nitrophenol, on cell of PHA producing bacteria Cupriavidus necator H16 and mutant strain Cupriavidus necator PHB4, which does not produce polyhydroxyalkanoates. Static magnetic field was generated by both permanent magnet and electromagnet. The effect of magnetic field on the growth of bacterial cells was studied using growth curves. It was found that cultivation in magnetic field and mineral medium mostly inhibits bacterial growth. Also the amount of polyhydroxyalkanoates was observed using FT-IR, flow cytometry and microscopy with fluorescent dye. Growth curves and flow cytometry were also used to study the influence of organic substances on bacterial cells. It was found that while benzene does not affect either C. necator H16 or C. necator PHB4, p-nitrophenol acts as the inhibitor of bacterial growth for both cultures. Finally the impact of p-nitrophenol on the accumulation of PHA was studied using gas chromatography.
Mělník as the residence of Czech queens

The topic of this bachelor thesis is Mělník, a famous royal dowry town that used to be an essential residence of numerous queens of Kingdom of Bohemia. The focus is on the importance of Mělník throughout history, from its creation in the 9th century to the 15th century. The first two rulers to reside in Mělník were Przemysl princesses Ludmila and Emma. The last queen to ever stay in Mělník was Joanna of Rosenthal in the 15th century. This thesis is dedicated mainly to queens Elizabeth of Bohemia, Barbara of Cilli and Joanna of Rosenthal. The aim is to analyze available historical documents and secondary literature sources and use them to explain the connections of the queens to the town, portray Mělník as their residency and explain its economic and political effects. The use of art therapy practice for individuals with inadequate self-concept

This bachelor thesis deals with the use of art therapy in working with adults with inadequate self-concept. The thesis is divided into theoretical and practical part. The theoretical part contains two main chapters that deal with self-concept and art therapy. Self-concept deals with a person's relationship to himself, witch fundamentally influences the possibilities of how an individual can relate to the world and suceed in it. Art therapy in a narrower sense uses fine art for therapeutic purposes. The practical part reports on art therapy work with adults with inadequate self-concept. Research outputs are presented in the form of case studies.

Analysis of the performance management in the organisation

The Bachelor Thesis deals with the analysis of the performance management in the company Dum zahranicni spoluprace p.o. The main goal of the Bachelor Thesis is to analyze and evaluate the performance management in the company and to formulate potential suggestions for optimization. The main sources for the system analysis are interviews with employees, internal documents from the company and quantitative questioning of employees using questionnaires. Based on the results, strengths and weaknesses of the system were identified. The performance management in the company can be described as correctly set according to the theoretical findings in comparison with the data from the employee survey.
Fetal Alcohol Syndrome - The Effect of Alcohol Abuse during Pregnancy on Early Psychomotor Development of the Child

The thesis devotes to the negative effects of alcohol abuse during pregnancy on early psychical development of the child. It surveys the problematic fields of development of the children who were prenatally exposed to alcohol. The theoretical survey is accompanied by qualitative empirical part, which contains four case studies of children in the age of 6 - 72 weeks whose mothers were drinking alcohol during pregnancy. There are several problematic areas in the development of these children, for example growth deficit, retardation of the psychomotor development (especially in speech, gross motor skills and social behaviour), attachment disorders and eating disorders. The Way to Literature using the Resources of Drama Education Methods

Z různých teorií o úloze dramatické výchovy jako metody při vyučování nejen na uměleckých, ale na základních školách vůbec, vyplývá, že by mohla být úspěšně použita při výuce. Úkolem této práce bylo její metody vyzkoušet v praxi a zjistit, zda je opravdu možné cíle, jež si klade primární škola v rámci výuky a výchovy k literatuře, skutečně naplnit, navíc formou, která je pro děti zábavná, atraktivní. Nebylo jednoduché používat prostředky dramatické výchovy jako cesty k literatuře stejným způsobem, jako například na základních uměleckých školách, kde vzhledem ke specifickému zaměření dramatického oboru má učitel snadnější možnosti. Na základních školách přece jen učitel musí lépe zvažovat, s jakými konkrétními metodami a prostředky může zacházet vzhledem k náročnosti metod tohoto oboru (například na čas, který práce s prostředky dramatické výchovy vyžadují, na potřebu většího prostoru než jaký bývá obvykle ve třídách i vzhledem k vyšším počtům žáků).
