Strong Epistasis in Genetics of Leishmaniasis - Identification of Genes and Mechanisms

Leishmaniasis, a disease caused by Leishmania parasites, ranks as the leading neglected tropical disease in terms of morbidity and mortality. Genotype of the infected organism is an important factor that influences susceptibility to and manifestations of this disease. To study human disease using mouse models, several strains are required, which could collectively exhibit different human pathophysiology. In the current thesis, we investigated the genetic influence on resting levels of immune cells in mice, because these resting levels could influence susceptibility to many of clinical disorders, including infectious diseases; and we performed a systematic review of the role of host genetics and cytokines in Leishmania infections. Moreover, we employed systems genetics to map genes causing susceptibility to leishmaniasis, and to identify additional mechanisms controlling response to Leishmania parasites. Also, we fine mapped the locus Leishmania major response 15 (Lmr15) in order to functionally characterize its role and to identify novel potential candidate genes regulating the response to L. major. Finally, we show a novel role of guanylate binding proteins 2b and 5 in the resistance to L. major. Pilgrimage, countryside and pilgrim

The subject of the diploma thesis is the relationship between the artist and the landscape. The thesis focuses mainly on the tradition of pilgrimage and outlines the aspects of the landscape that the artist uses as a pilgrim in his work. The pilgrimage phenomenon is shown here through the works of Czech as well as foreign authors, from the illustrations from the travels to the performances in the landscape. In the didactic part, the tasks focused on the activities in the landscape are outlined, which reflect the perception of the landscape of the secondary school pupils. Information education at secondary schools: research at Business Academy in Tábor and implementation of the results into practice

Cílem rigorózní práce je zmapování současného stavu informační gramotnosti na středních školách na základě konkrétního průzkumu na Obchodní akademii v Táboře. Úkolem je analyzovat proces efektivní práce s informacemi v souladu s osnovami obchodní akademie a podat výčet informačních dovedností žáků střední školy. Úvodní kapitola by měla podat přehled o základních pojmech týkajících se informační výchovy a vzdělávání. V dalších kapitolách jsou ilustrovány informační dovednosti žáků střední školy na základě dotazníkového šetření, možnosti zapojení informační výchovy do jednotlivých předmětů, spolupráce školy a knihovny, přínos informační výchovy pro žáky střední školy atd. Práce vychází z praktické zkušenosti s výukou informační výchovy na Obchodní akademii v Táboře. Věnuje se také teoretickým aspektům informační výchovy se zřetelem na prostředí středních škol. Rigorózní práce je určena zájemcům o problematiku informační výchovy a zároveň nepatrným pokusem o návrh úpravy vzdělávacího systému v České republice.

Pathway to inclusion: attitudes towards inclusive education of intellectually disabled children in the Czech Republic

A small-scale research involving a total of 39 special education teachers and parents was conducted in the city of Sumperk and Rakovnik, in the Czech Republic. Since the implementation of the Equal Education Act in 2004, the numbers of children in segregated special schools remained high and the largest number of children attending these segregated special schools are intellectually disabled. History and the present situation of the municipal library of Slaný

Cílem bakalářské práce je deskripce a posouzení historického vývoje a současnosti Městské knihovny ve Slaném. V práci je nejdříve stručně popsána historie města Slaný, podrobněji potom kulturní historie tohoto města a historie Městské knihovny ve Slaném, a to i se zřetelem na historii čítárny, která po značnou část kulturní historie Slaného působila mimo rámec Městské knihovny. Velká pozornost je v práci věnována významným kulturním osobnostem, které působily ve Slaném a ovlivnily jeho kulturní život a v části věnované historii knihovny i osobnostem, jež se zásadním způsobem podílely na založení a dalším rozvoji knihovny. Těžiště práce však spočívá v popisu a rozboru současnosti knihovny. V práci jsou popsány základní dokumenty důležité pro zřízení Knihovny V.Štecha a její provoz a dále potom charakteristika a základní parametry v její činnosti, a to především se zřetelem k dlouhodobějším i krátkodobým tendencím, které se v práci knihovny projevují. Podrobně je rozebráno především poskytování služeb, a to jak v oddělení pro dospělé, studovně i v oddělení pro děti. V této souvislosti je věnována pozornost i doprovodným propagačním, vzdělávacím a kulturním akcím knihovny. Pozornost je věnována rovněž organizaci knihovny, jejímu personálnímu zabezpečení, úloze poboček, technickému a ekonomickému zabezpečení...

charitable projects were born in church environment and church members were among the first social workers ever. Do Christian churches have such a remarkable influence on social work evolution? Is Christian altruism, help without the expectation of subsequent satisfaction, involved in social services development? Is the social mission of humanity truly part of the accents of church doctrine? This paper tried to evaluate whether the church highlights, supports and develops social and charitable pursuit, if it invites its members to gratuitous support for other members of the community and if it was at the eve of social activities in contemporary society.

National libraries of Italy

Práce se zabývá v evropském, ale i světovém měřítku ojedinělým systémem národních knihoven, který má své místo v Itálii. Úvodní část práce, složená z kapitol 1 a 2, je věnována jednak úvodu do italského knihovnictví a nastínění možných dělení italských knihoven obecně (kapitola 1 ), jednak podrobnějšímu seznámení s postavením národních a ústředních národních knihoven v italském systému, charakteristice jejich rolí a funkcí, seznámení s italskou knihovnickou legislativou a Italskou knihovnickou asociací (kapitola 2). Druhá část práce, skládající se z kapitol 3, 4 a 5, obsahuje rozbory jednotlivých druhů národních knihoven spolu s charakteristikami jejich historie, fondu, sbírek, katalogů a přehledem projektů, jichž se aktivně účastní. Kapitola 5 je věnována dvěma nejvýznamnějším projektům na národní úrovni, na nichž knihovny kooperují, a to Italské národní bibliografii a Národní knihovnické službě. Závěrečná část práce tvořená kapitolou 6 znovu shrnuje základní poznatky týkající se národních knihoven v Itálii a jejich postavení v systému knihoven v komparaci s všeobecně uznávanými principy fungování národních knihoven v rámci jedné země.

Analysis of adenosine triphosphate and adenosine diphosphate by HPLC-MS/MS

In this bachelor thesis, ADP and ATP samples were analysed and detected with HPLC- MS/MS method. Approximate limit of detection (LOD) for these particular substances were found and their values were compared with the LOD values published in the literature obtained via the same methods and under very similar experimental conditions. Our limits of detection for nucleotides were comparable with the limits described in the literature. Mass spectrometry analysis was performed in the positive and the negative mode of multiple reaction monitoring analysis and electrospray was used for the analyte ionization. The optimal conditions for high performance liquid chromatography of ATP and ADP analysis were acquired on a ZIC - HILIC column with the mobile phase of 75:25 (v/v) acetonitrile / 10 mM ammonium acetate. Ammonium acetate buffer was adjusted to pH of 7.15 and the separation was done under the isocratic elution.
