Two methods of measurement velocity and concentration in flow of a two gases mixture

The small aerodynamic facility with variable density, concentration of the air-helium mixture and flow velocity is described. Then two methods of measurement velocity and concentration of a known two gases mixture flow are presented. The first one make use of twoheated sensors and the second one aply a single hot-wire probe.

The methodology of hydrogeologic tracer tests in fractured environment using fluorescent tracers

The aim of the methodology is defining of optimal tools and effective work flow to perform and well evaluate tracer tests using fluorescent, non toxic tracers. The methodology focuses on in-situ tests in fractures rock environment, applying artificial hydraulic gradient and monitoring of changes in tracer concentration during test period. Data record is continuous and provide inputs into the numerical models.

SPARK PLASMA SINTERING OF BALL MILLED AND ATOMIZED POWDER BASED ON Fe-Al

High-quality compacts were prepared using the spark plasma sintering (SPS) method from powders of similar composition Fe-Al-Zr-B. The properties of the sintered compacts are strongly dependent on the morphology and properties of the feedstock powder. The first powder was obtained by ball milling of the as cast alloy and the second was prepared by atomization under argon atmosphere. The morphology and structure of the two powders are compared and mechanical properties and microstructure of compacts prepared under the same conditions of the SPS procedure are discussed. The milled powder has an irregular morphology and shape of the polycrystalline particles, which have a completely disordered BCC structure with considerable internal stresses, high concentration of structural defects and the presence of aluminum oxide. The atomized powder particles are nearly spherical, polycrystalline with ordered B2 structure, with no significant signs of internal stresses and oxidation. Microhardness of particles of both powders was measured and compared with the microhardness of compact materials, the results of compression tests of compacts at room temperature were compared and discussed.

Determination of biological activity of adiponectin, a recombinant protein using cell culture

This thesis deals with the determination of biological activity of adiponectin, a recombinant protein using cell culture. First it was important to acquire the working skills for the cell culture of cell line 3T3-L1. An optimal concentration of inactivated fetal bovine serum in cell culture media was determined. A stimulation of the cell proliferation by HB-EGF, PDGF-BB and bFGF growth factors was observed at various concentration levels. Afterwards the biological activity of adiponectin was determined as an inhibition of growth stimulation with 5 ng/ml PDGF-BB. This biological activity assay for adiponectin was also conducted with lyophilized adiponectin and a growth factor bFGF (0.1 ng/ml). The lyophilization did not affect the biological activity of adiponectin.

Acquisition, Modeling and Signal Processing in Ultrasound Perfusion Imaging

This work deals with possibilities of ultrasound perfusion analysis for the absolute quantification of perfusion parameters. In the theoretical part of this work are discussed possibilities of using of the ultrasound contrast agents and approaches for the perfusion analysis. New methods for the perfusion analysis are suggested and tested in the practical part of this work. The methods are based on convolutional model in which the concentration of the contrast agent is modeled as aconvolution of the arterial input function and the tissue residual function. The feasibility of these methods for the absolute quantification of perfusion parameters is shown on data from phantom studies, simulations and also preclinical and clinical studies. The software for the whole process of the perfusion analysis was developed for using in hospitals.

The fate of the Roma in Czechoslovakia between 1918 - 1945 from the point of view of the settlement Bohusoudov destruction.

The thesis generally deals with the situation of the Roma on the territory of newly formed Czechoslovakia between 1918 - 1945 on the backgroundof the particular example of the Roma settlement in Bohusoudov. The introductory part concentrates on coming of the Roma ethnicities on our territory and on circumstances of formation of the Roma settlement in Bohusoudov. The following one tries to look closer at the state of Roma population in the Czechoslovakia Republic from the point of view of legal framework, subsistence, criminality, schooling and religion. This view is complemented by circumstances in Bohusoudov. The next chapter concentrates on Roma genocide which took place in forced concentration camps in Hodonín u Kunštátu and Auschwitz II-Birkenau. The second part of the thesis contains a survey among contemporary inhabitants of neighbouring villages which tries to find out knowledge of history and life of the perished Roma community in Bohusoudov. The enclosure of the thesis offers a set of preserved texts from Bohusoudov and a nominal list of victims from the settlement.

Mass Spectrometry Imaging of Metformin in Tissue Sections

Mass spectrometry imaging is a powerful technique suitable for visualization of the distribution of a wide variety of compounds within tissue sections. The main aim of the study was the development and optimization of a sample preparation procedure allowing determination of the distribution of orally dosed metformin in mice kidney sections. Metformin is the first-line medication for the treatment of type 2 diabetes. The optimization of the sample preparation step before imaging experiments included the selection of a suitable matrix and the optimization of various parameters of MALDI analysis. 2,5-dihydroxybenzoic acid was identified as the best matrix providing highest sensitivity. A sublimation method was successfully used for the matrix deposition. The highest relative concentration of metformin was found in the inner zone of kidney 30 minutes after the drug administration.

8 Abstract Counterurbanisation research, which is a central topic of this thesis, represents an appropriate field where transformations of contemporary societies and their socio- spatial organization can be examined. The dynamics of social and societal changes creates a demand for developing new methods and tools which helps us to understand contemporary change. Counterurbanisation is a multi-faceted concept which has been controversial since its initial use. An introduction of the concept of counterurbanisation in the field of population geography provoked a general debate on new and emerging trends in development of settlement system. Different logics of concentration and deconcentration of human activities has been challenged. Counterurbanisation operationalized as a migration poses a challenge for many standard conceptualizations of migration. The logic of non-economically motivated migration from the centre to the periphery requires a search for alternative explanatory mechanisms. Impact assessment and the impact of counterurbanisation in rural communities are analogous to the classical social science studies evaluating the integration of immigrants from rural areas in the cities.
