Lipidomic analysis of vernix caseosa

Methods of analytical chemistry are widely used in lipidomics. Separation techniques coupled to mass spectrometry or nuclear magnetic resonance are used very often. They make it possible to identify lipids present in the matrix in very small quantities. This work summarizes the application of modern analytical methods and instrumentation for identifying and characterizing lipids in vernix caseosa. It is shown how I contributed during the Ph.D. studies to the elucidation of the structure and characterization of unknown lipid classes followed by more detailed description of those lipid classes already identified in vernix caseosa. An integral part of my work was the application of the method enabling the localization of double bonds developed by our laboratory in triacylglycerols and 1,2-diol diesters in vernix caseosa. This analytical method is based on the formation of an acetonitrile adduct in an ionization source of a mass spectrometer enabling atmospheric pressure ionization. The complexity of the triacylglycerol class did not allow a complete characterization of the double bonds. However, the fragmentation mostly showed that double bonds up to n-12 position are present, but small peaks in some spectra also indicated double bonds at more distant positions from the chain termini. Nursing Problems of Patients with Skin Cancer

The character of oncological diseases can be benign or malignant. Both variants have fundamental consequences for the patient. If the changes in the skin are malignant, treatment is necessary. In case the medical finding is benign, it depends on the patient whether the tumour would have it removed or not. In any case, the patient must be informed about all risks and especially the dispensarisation which is the most significant factor for early diagnosis and effective treatment. Oncological disease of skin entails many significant changes in the patient's life. The patient's order of values, lifestyle, as well as perception of life changes. In this difficult situation, the support of the patients from the side of family and loved ones, as well as of the medical personnel, is crucial. The patient's need of security and safety must be satisfied, as it is a significant factor in coping with the disease development from diagnostics to treatment and eventual dying. The concept of the bachelor thesis is theoretical and based on the study and analysis of information sources related to the issues of oncological disease of skin. To introduce the largest and very important organ the skin its anatomy and physiology is briefly described at the beginning. Subsequently, oncological diseases of skin, divided into benign tumours, their variants and characteristics, are outlined. The subchapter of malign tumours lists the most frequent malignant skin tumours, i.e. basal cell carcinoma (BCC), squamous cell carcinoma (SCC), and melanoma. Again, their characteristic and seriousness is described. Diagnostics and therapy of oncological diseases of skin is worth mentioning. The prognosis of the entire disease depends on early diagnostics. If it is timely, the treatment is less demanding and the patient handles it much better. In the area of therapy, the most frequent methods of treating this disease are illustrated. In connection with this disease, it is important to mention dispensarisation and prevention, oncological and palliative care as well as the impact of the disease on the patient's psychic condition. The bachelor thesis is complemented with supplements in the form of tables. One of them refers to incidence, mortality and prevalence of malignant melanoma from 1994 to 2014. Other tables serve for easier understanding of classification of skin tumours. The objective of the thesis was to map the nursing care of oncological diseases of skin. A nurse is an important element of prevention. She can be helpful and educate the population about self-monitoring of skin. During the treatment, the nurse applies medicaments and treats skin after surgical procedures or radiotherapy. The nurse plays an essential role within oncological and palliative care. That is why her relevance is described in several chapters. For many patients, the support of the nursing staff is a very important factor also in the area of psychic condition, on which the approach to the disease, as well as the development of the treatment, are based. It was necessary to acquire many findings about oncological diseases of skin to write this thesis. Many experts are engaged in this area nowadays, especially in the area of treatment. Information was obtained from both Czech and foreign expert bibliographic publications and articles. The studied information is incorporated into individual chapters that can be beneficial for anyone who is engaged in this specific area of oncology.
Liveness Detection on Finger Using Various Light Sources

The purpose of this work was to increase quality, security and reliability in biometric systems. This work presents electric and optical characteristics of a human finger. In the first part of this work you can find a theoretical basis about biometric systems, a skin and light. The next part is devoted to electric characteristics of a human finger. The third part of this work describes design, implementation and results of testing optical characteristics of a finger. In the end you can find information about some of the improvements for the future.
Cell growth on biomaterials for skin replacements and wound dressings

Tissue engineering is an emerging interdisciplinary field developing new ways of treatment of patient's tissue defects using artificial substitutes. Skin tissue engineering is developing skin substitutes and wound dressings that would replace current treatment using autologous, allogeneic or xenogenic substitutes. There are high demands on materials which should serve as a scaffolds for dermal fibroblasts and keratinocytes. They must be non-cytotoxic and biodegradable with a rate proportional to formation of a new tissue. The materials should support adhesion and proliferation of the cells and even they could release growth factors and antimicrobial substance to enhance healing and new tissue formation. In this master thesis, the cell adhesion and proliferation were evaluated on sodium carboxymethyl cellulose (Hcel® NaT), poly-ε-caprolactone (PCL), poly-L-lactide-co-ε-caprolactone (PLA/PCL) and cellulose acetate (AC) nanofiber membranes. Primary human dermal fibroblasts and HaCaT cell line keratinocytes were selected for evaluation. The cell adhesion was observed by fluorescent microscopy, the proliferation was determined by metabolic assay (WST-1) and the material cytotoxicity was evaluated in xCELLigence® system. Eva Kmentová and the Body Imprint in the Sixties and Seventies Art

The bachelor thesis deals with the physical imprint in the sixties and seventies Art. The time limitation is mainly based on the selected works of sculptor Eva Kmentová (1928-1980), in which the phenomenon of body imprint is present. The issue of body imprint is gripped in a broader theoretical context and is based on two decisive foreign works. Primarily the book of George Didi - Huberman Le Ressemblance par contace, and secondly the theoretical thesis of Rosalind Krauss and Yve-Alain Bois in their catalogue Formless : A User's Guide to the exhibition L'informe. The bachelor thesis shows a possible view on the body imprint as a dialectic phenomenon generated by meeting of formlessness - shapeless mass and of the form - represented by imprinted object. For deeper understanding of Eva Kmentová's background, the chapter summarizing the changes in Czechoslovakian informel art in the late fifties and early sixties is included. Within the context of imprint's indexical value the bachelor thesis turns to Roland Barthes and his theory of photography. ABSTRAKT: The main subject of this thesis is focused on preparation of taxidermal mounts with use of mammal bones and skins and their use as teaching aids. The target group are school children both of elementary and secondary schools. The thesis describes methods of bone preservation and cleaning, skin tanning and preparation of taxidermal mounts. I deal in it also with preparation of large vertebrates' skulls and preparation of taxidermal mounts. The procedures are described in details and the following chapters include photos of dissections made by me. Preparation of 4 bird and 6 mammal species is described. Problems of the Patient with Psoriasis and his Integration to the Society

The bachelor work is dealing with the problems of patients, afflicted with psoriasis.In the first half of the teoretical part I am fixing on the description of the anatomy and physiology of the skin.The second half of the theoretical part makes clear, what actually psoriasis yields, what is its history, how manifests, what is the posssibility of diagnostics, what types of psoriasis we can meet in the practice, which are the possibilities of the treatment and why is important education of the diseased person,the family and the society.In several chapters I describe,which is the fall-out on the psychical and physical disposition of the patient. Last but not least I am joining on the chapter about the integrating of persons with psoriasis to the society.The empirical part is fixed on the life and stories of the people who did not choose this disease wilfully, however it guides them for the big part of their life. There was used the probe of the qualitative investigation with the technique of the half structured dialogue.The dialogues were recorded and subsequently analyzed by the help of the method of stationary theory.The principal aim is to find out,in what social spheres people with psoriasis have most often problems.
