Abiotic stress resposes in apple (Malus domestica Borkh.) tissue cultures
Krsek, Daniel ; Pospíšilová, Jana (referee) ; Lipavská, Helena (advisor)
Abiotic stress factors, primarily those which disturb plant water balance, cause extensive crop losses. With regard to sessile lifestyle, many mechanisms how to cope with these unfavourable conditions were developed during evolution of plants. As common impact of many of these stresses like drought, salinity or low temperatures is of osmotic nature, the osmotic adjustment represents important part of plant stress response. Mostly, this component of stress reaction is provided by organic compounds, which are referred to as compatible solutes, including polyhydroxyl compounds (sucrose, sugar alcohols, cyclitols, and oligosacharides) and nitrogen-containing compounds (mainly proline). The effect of these solutes is not only osmotic, but also osmoprotective as they are able to preserve integrity of membranes and macromolecules by mimicking their water envelope. The compatible solutes also contribute to quenching of reactive oxygen species overproduced under these stress conditions. This study is focused on apple (Malus domestica) producing, beside sucrose, sugar alcohol sorbitol as primary photosynthetic product and transporting these carbohydrates along with raffinose family oligosaccharides (RFO) for a long distance. In vitro tissue cultures derived from leaves of two apple cultivars differing in...

Agaricales, russulales and their application in school practise
Košťáková, Veronika ; Ziegler, Václav (referee) ; Dostál, Petr (advisor)
Agaricales and Russulales mushrooms have recently been evaluated from the locality of Čeřovka hill in the vicinity of Jičín town. The locality is also briefly characterized climatologicaly, geologicaly and biologicaly; short history of mycology, origin of mushrooms, systematics and taxonomy are also discussed . Methodology is based on both - i. e. macro- and microphotography (complete specimens and sporas). Every-week sampling enabled a study on fungi abundance, seasonality and diversity. Basic characteristics of each taxa are enclosed like a distribution maps. Two working hypothesis are introduced: The first occurrences of Agaricales and Russulales mushrooms in southern slopes of the locality and the differenet diversity in northern and southern slopes. Didactic part is divided into four parts - suggestion of natural trail, a key for mushroom species distinguishing, creating of personal collection and medical help in cases of poisoning. The school programm is established for secondary schools.

Distributed Sytem for Verification of Properties of Natural Numbers
Tomisová, Martina ; Peterek, Nino (referee) ; Mírovský, Jiří (advisor)
The result of my work is a system for distributed verification of properties of natural numbers. It has two parts - server and client. These communicate via HTTP protocol. The clients perform the computation, the server distributes the work (numbers) and gather results (properties of the given numbers). The input of one computation should be one natural number, as well as the result (output). The distribution can be used for verification of a given property for several natural numbers. Particular jobs can be added to the client as plugins. Two examples of plugins are a part of the work. The first one is very simple and shows how to create plugins. The second example searches for prime numbers (and has it's own arithmetics library for long numbers) - the server can distribute (possibly big) numbers, the client will verify whether a given number is a prime number.

Forest industries in experimental archaeology
Pauknerová, Karolína ; Popelka, Miroslav (referee) ; Matoušek, Václav (advisor)
The diploma work informed by the theoretical platform of postprocessualism deals with forest industries in the context of experimental archaeology and particularly with questions of how to research and present them. Forest industries are defined by the author as branches of handicraft closely connected to forest both spatially and regarding raw material. Tar, charcoal and potash productions are presented in detail (their technological aspects, chemical substance, terminological problems, and use of the products are discussed and a brief overview of archaeological research of production places is offered). In the chapter on experimental archaeology, the author elucidates the sense of experiment and presents her own division of experiments in archaeology to science-nature-experiments and experiments-simulacra. Further on, the author describes particular experiments made by experimental archaeology and compares them with how forest industries are presented in museum. In the practical part of the paper, experiments with tar production in pits carried out by the author between 2007 and 2009 are described and discussed.

The Development of Mimetic Desire towards Latent Conflict in the Work of Katherine Mansfield
Nováčková, Zuzana ; Wallace, Clare (advisor) ; Nováková, Soňa (referee)
Thesis abstract Using Aristotelian notion of imitative behaviour and René Girard's theory of mimetic desire, several stories by Katherine Mansfield are analysed in order to demonstrate the development of mimetic desire together with its implications. The analysis follows the negative aspects of mimesis: the problems it causes in relationships, as well as the positive aspects including the self-knowledge. Since Mansfield's stories do not correspond fully to Girard's theory, the analysis explores a specific way of dealing with mimetic desire: keeping the conflict latent. At first, the stories about childhood offer an insight into Aristotelian concept of mimesis - imitative behaviour being a natural and pleasing human activity that is best observed in children's plays. The stories show how children choose their models, how they comprehend the world that surrounds them, especially the interpersonal and social codes, and how important is imagination in their mimetic activities. The analysis proceeds from natural imitation to the origins of mimetic desire, focusing on two modes of mediation and on the process of realization of one's own self-authenticity. The search for self-authenticity is possible due to external or internal mediation of desire. The transition from one type to the other is explained by the...

The Relapse of Drug Additions in the Civic Association Sananim.
Faltinková, Lucie ; Dočkal, Jan (advisor) ; Radimecký, Josef (referee)
This thesis presents the problem of relapse of drug addictions. The relapse is presented as a possible constituent of the treatment of a drug addiction. It is the moment when client uses a drug once or repeatendly during a short time after the particular time of total abstinence. The thesis examines a process of aftercare which engages in relapse prevention. The most important topic is also a cooperation with client's family because of relapse prevention and as a kind of therapy for them in case of relapse their addicted relative. My thesis presents the relapse as a something natural. It doesn't have to be definitive and a client can make a instruction of it and continue in his abstinence successfully. The Picture as a Phenomenon
Universalová, Jana ; Kornatovský, Jiří (advisor) ; Cikánová, Karla (referee)
My thesis consists of three parts: the practical, the theoretical and the didactic part. In the practical part I represent my own work, which examines the phenomenon of the pictures using pictures of one persone. By other words I represent my personal conception. I unrestand picture as something that stands at the begining of the work and can be expresed by depiction. The practical part became the starting point for the following parts. In the theoretical part I look for connections between my Picture and Pictures of the Czech artists. By other words I look for some resemblance between them. The third part deals with my pedagogical activities. An important intention of my thesis is not only the view of my work, or my own self-reflection and a way of searching for relations to the art. But especially the afford to use my inspiration in the pedagogical activitiy. I verify the possibility of using my own inspiration at school with the students of secondary schools. The key words: picture, visualization, art, nature, movement, line, drawing, transcript, creative proces, communication, education

Footbridge across the Labe
Krč, Rostislav ; Romportl, Tomáš (referee) ; Stráský, Jiří (advisor)
The thesis focuses on design of prestressed concrete footbridge which passes pedestrian and bicycle path over the Elbe river near city of Celakovice in the Czech Republic. Three different bridge options were analyzed and for further development a cable-stayed footbridge was chosen. Its bridge deck is formed of a box girder supported by cables in its vertical axis and all cable stays are anchored into two concrete A-shaped pylons. This structure was analyzed in SCIA Engineer and assessment of serviceability limit states and ultimate limit states according to recent European standards (Eurocodes) was made. Both the global structural behavior and the local structural integrity of box cross-section were assessed as well as construction stages and cross-sections of pylons and cable stays. Assessments were performed in IDEA StatiCa combined with hand calculations. Eventually a dynamic response of structure was analyzed. Natural modes and frequencies were found and forced oscillation response was evaluated. The thesis includes technical drawings, construction process and visualization.

Cost calculation on the farm - cattle breeding, milk
Šilhavý, Luboš ; Řezbová, Helena (advisor) ; Dagmar, Dagmar (referee)
The topic of this dissertation is the cost calculation of cattle breeding and cost of auxiliary activities focused on the milk production of the Mecholupska zemedelska company in 2015. The thesis is devided into two parts, the theoretical part and the practical one. The theoretical part outlines the cattle breeding and the breeds both with milk production and meat breeds. Further it describes the economic terms of milk production, costs of cattle breeding, costs itself, calculation methods and calculation formulas. The practical part of the thesis describes the nature of the company and its production activities. The part focused on the arable farming describes sowing areas of plants and revenues and cost calculation of corn silage. The main target of the thesis is the an milk production, which is based on cost calculation of milk, described in details in the focused on stock farming. The cost indicators are stated in the table and then the particular items are defined. Several variants with modified internal costs of inputs are calculated and compared with each other afterwards. Based on this analysis, the conclusions of the cost calculation of milk are summed up and some suggestions to stabilise and develope the sector are outlined.
