Lužová, Veronika ; Polášek, Miroslav (advisor) ; Pospíšilová, Marie (referee)
A novel capillary electrophoretic method for the separation of pancuronium (PM) and vecuronium (VM) ions utilizing capacitively coupled contactless conductivity detection (C4 D) was devised and validated. The separation was carried out in bare fused-silica capillaries (50 µm i.d., 75/45 cm) at 25 řC. Optimal background electrolyte was 50 mM borate buffer of pH 9.5 containing 12.5 mg/ml of hydoxypropyl-γ-cyclodextrine; (phenyltrimethylammonium iodide was used as internal standard). The samples were injected hydrodynamically at 1000 mbar for 3 s. The separation was performed at +30 kV. Under such conditions the PM and VM were base-line resolved and the separation took < 4 min. Calibration curves were linear for both PM bromide and VM bromide in the range 25-250 μg/ml with r > 0.9968. The limits of detection were 7 and 6 μg/ml for PM bromide and VM bromide, respectively. The accuracy tested by recovery experiment at three concentration levels of added PM bromide and VM bromide was satisfactory (95.7-102.7 %, n=3, with RSD < 2.61 %). The method was successfully applied to the assay of PM bromide and VM bromide in commercial injection solutions.

Kociánová, Sabina ; Vávrová, Milada (referee) ; Zlámalová Gargošová, Helena (advisor)
Brownfields are one of the unfavorable parts of industrial heritage that developed from 18th to 20th century. Brownfields are very different and in most cases they have negative impact on the environment. Crucial for this thesis were brownfields with certain level of contaminations. The site of a former refinery of mineral oils OSTRAMO, which represents ecological burden, is a brownfield that can threaten the environment in all its compartments. This was the reason why OSTRAMO was selected for this thesis. The goal of this thesis was to evaluation of ecotoxicological effects of contaminated samples taken from the location via ecotoxicity testing in aquatic and contact arrangement. The result of each test confirmed expected negative impact of the samples from the location of former rafinery on testing organisms.

The bachelor's work deals with quality of life of patients with disseminated sclerosis. It is trying to find the influence of chronic illness over the quality of life fřom the view of the patients themselves. It adverts to the impact that the illness has on the mental and physical part of their lives. It nears to public (laity or medics) the disorders and limitations which this illness can make the lives of patients worse. I chose and translated the questionnare MSIS - 29 for my research. The sample was made from the people who they diagnosed the illness disseminated sclerosis, 75 informants, 49 women nad 26 men, aged from 20 to 68. Interviews were filled in Domov sv. Josefa in Žireč near Dvůr Králové, in Rehamedica in Žacléř, in Pražské kongresové centrum during " The International Day of People witd disseminated sclerosis", in the Trutnov's hospital and by people in home care.

Rapid detection microogranism in pharmaceutical materials.
Novosád, Aleš ; Jílek, Petr (advisor) ; Voxová, Barbora (referee)
1 Summary Many microbiology laboratory, no matter what already in pharmaceutical firms or to hygienic stations ground and firms conversant food production used to analysis big quantity microbiology samples conventional techniques based on cultivation appropriate culture. These techniques take hours to days to yield a result, are tedious and are not suitable for non- culturable microorganisms. Further, culture-based techniques do not provide real-time information on the physiological status of the organism in situ which is important in the industrial manufacture of many microbial products (1) and often at high of the number of micro - organisms, when isn't possibility number micro - organisms determine, all time cultivation about next dilution sample elongate and about time growth this micro - organism. Flow cytometry makes it possible to obtaining real microbial determination single micro - organisms, without dependencies on type microbial culture. Nevertheless, flow cytometry has not been extensively used as a tool for routine microbial analysis. Reason is mainly high cost and complexity of instrumentation, the need for trained flow cytometrists and the lack of assay kits with appropriate biological reagents for specific applications. Much modern instruments are now relatively simply serviceable, thanks...

Oslejová, Kateřina ; Kubíček, Vladimír (advisor) ; Lázníčková, Alice (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of biophysics and physical chemistry Candidate: Kateřina Oslejová Supervisor: ing. Vladimír Kubíček, CSc. Title of diploma thesis: HPLC Analysis of Benzimidazoles Using Core-shell Columns I The aim of this work was to transfer the HPLC method of determination of flubendazole and its metabolite, i.e. reduced flubendazole, from a classical C18 column (250×3,0 mm; 5 µm) to core-shell column Ascentis® Express C18 (10×3,0; 2,7 µm) and to find a suitable internal standard. Experiments were performed using liquid chromatograph Agilent 1200 Series® with both DAD and fluorescence detection. Flubendazole was detected with the DAD detector. Low concentrations of reduced flubendazole were detected with the fluorimetric detector. Several mobile phase compositions were tested. The mobile phase consisting of phosphate buffer (pH = 3,0; 0,025 mol/l) and acetonitrile (70/30, v/v) appeared to be a good choice for the separation under study. Two internal standards were exploited for the quantification, depending on the detection principle. Mebendazole served as the internal standard in the case of DAD detection, while oxibendazole was utilized for fluorimetric detection. The proposed method was successfully applied for analyses of real samples.

Kasková, Barbora ; Matějček, Tomáš (advisor) ; Kučerová, Silvie Rita (referee)
11 ABSTRACT The study reported in this paper was undertaken during autumn 2007 to assess the status of environmental education and awareness of eight-formers living in chosen localities of a different population range. The main objectives of the study were to determine the depth of pupils' knowledge and awareness of environmental issues and problems and their attitude to the environment. Is there a certain homogeneity of environmental awareness and attitudes among most schoolchildren at village, town and the capital? To test the veracity or otherwise of this widely held assumption. The study comprised a detailed questionnaire of a sample of 324 schoolchildren. The study was designed to assess the general characteristics, environmental knowledge, and behavioural pattern of the sample. The research finding in the study indicated that positons of pupils result mainly from all-society views and less reverberate thein own experiences. Informants cann`t context aglomerated enviromental problems with the others global ones yet and they perceive them separately. Only few informats are able to answer what a global problem means. Also a few of them can concretely enumerace just one of the global problems. Lesser of them have awareness of a sustainable growth. Absolute majority of them identify thein selfs with...

Lukš, Tomáš ; Dittrich, Milan (advisor) ; Kubíček, Vladimír (referee)
The methods of nanoparticles size and their zeta potential measurement based on dynamic laser light scattering a microelectrophoresis were reviewed. The procedures of the size and the shape factors of microparticles measurement with use of image analysis were presented. Also technique of microparticulate systems preparation by spray drying was described. The most important properties of biodegradable polymers including their degradatiom mechanisms and specific interactions with hydrophilic media were presented. In the experimental section were studied interactions of oligoester matrices with buffer solutions. Matrices contained not only oligoester carriers, but also mixtures of these carriers with mannitol microparticles and mannitol microparticles with microencapsulated silver nanoparticles. As oligoester carriers were chosen samples with branched constitution of molecules. As branching agent were used two polyhydric alcohols with six hydroxy groups - mannitol and dipentaerythritol. As hydrophilic testing medium were used water and range of buffers differing by ionic strength. Correlations between buffer concentration and swelling time course and erosion kinetics of matrices were demonstrated. Swelling degree characteristics of matrices were influenced by ingredients originated from the silver...
