Internet Communication
Hubka, Michal ; Pokorný, Pavel (advisor) ; Přibil, Jiří (referee)
Internet communication is becoming increasingly frequent communication tool, compared to the normal communication (like phone calls, sending letters, etc.). Work described the development of Internet communication, focusing also on its forms (formal and informal). Based on these forms of work it also demonstrated how to create a username or avatar, that applies to the traffic. The thesis focused on the social networking site Facebook, which is becoming increasingly popular. Pointed to the history, usage and also describes the basic functions. Facebook is very good commercial tool, so the work is focused on displaying the possibility of commercialization of this social network. The thesis pointed to other social networks, which may not be as popular, but worth a demonstration. Generally it's not just a social network that could be used for Internet communication. On the contrary, this form is younger than communicators, which are also described and introduced. It focused on the best-known communicators such as ICQ, Skype, etc. The work is a survey and analysis, which focused on the global Internet communications. The thesis is based on research. Firstly, there is a focus on the security account on Facebook, as well as the use of online communications companies and finally to use Internet communications of the faculty.

Fish husbandry in the mountains conditions at Borová Lada fish - hatchery in Šumava
PLIČKA, Vítězslav
Central parts of the Šumava national park has specific nature conditions which we must respect when fish farming is carry out . In 1998 fish {--} hatchery near Borová Lada was established. The fish hatchery exists more than 10 years and we still have no professional report about exploitation this object. (using assorted technologies of rearing, rearing under controlled conditions). The aim of this graduation thesis is evaluation results of trout culture and make proposition new improved technologic process in fish farming on this site. The thesis also includes experiments with initial feeding of fry of brown trout (Salmo trutta morpha fario) carryed out during the spring 2008. The main observed values were survivance, specific growth rate (SGR) and feed conversion ratio (FCR). After 17 weeks of initial feeding fry of brown trout the observed values were achieved: survival of fish 38,2 % , average length SL = 69,0 mm, average weight 4,9 g. Experiments showed that initial feeding of fry of brown trout (Salmo trutta morpha fario) with using feeding mixtures in the Borová Lada fish {--} hatchery is possible and effective.

Potrava doprovodných druhů ryb ve vodárenské nádrži
Zapletal, Tomáš
Limnological and hydrobiological parameters were compared with food composition for three fish species (bream, roach, perch) in a meso-eutrophic reservoir (Czech Republic). Fish were sampled using a 100 m beach seine, a fry seine and gillnets at both the lower and upper ends of the reservoir in 2011 and 2012. Food composition was evaluated using gravimetric methods. 0+ bream (36-48 mm standard length) diet consisted of zooplankton only; while in 3-4+ bream (146-186 mm), detritus and aquatic vegetation dominated. In summer, diet of 6-9+ (210-315 mm) bream from beach seines consisted mainly of aquatic vegetation and mainly of detritus when caught in gillnets. Benthic macroinvertebrates and zooplankton were minor dietary components throughout the season. For all roach age groups studied, zooplankton were a negligible food item. Detritus was dominant in 0+ (36-52 mm) and 1+ (72-110 mm) fish. Detritus, periphyton and macrophyte fragments were dominant in 3-4+ (73-163 mm) and 6-8+ (150-266 mm) fish, with benthic macroinvertebrates taken rarely. Zooplankton was dominant in 0+ (16-86 mm) and 1+ (52-81 mm) perch diet. In summer and autumn, cyprinid and perch fry dominated in adult 5-7+ (112-300 mm) fish, while detritus dominated in spring. Benthic macroinvertebrates were recedent. The food intake of bream and roach could be explained the shallowness of the reservoir banks, the rich littoral zone being used for spawning, hiding and for food periphyton. Adult perch play an important role in fish stock management, consuming fish fry that otherwise consumes zooplankton, which subsequently affects phytoplankton levels and water quality.

The use of differential scanning fluorimetry in characterization of selected carbonyl reductase
Tomanová, Radana ; Zemanová, Lucie (advisor) ; Novotná, Eva (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Radana Tomanová Supervisor: RNDr. Lucie Škarydová, Ph.D. Title of diploma thesis: The use of differential scanning fluorimetry in characterization of selected carbonyl reductase Differential scanning fluorimetry (DSF) is simple, rapid method that enables to determine optimal conditions for stabilization of proteins and discover their ligands. DSF monitors thermal unfolding of a protein in the presence of fluorescent dye (e.g. SYPRO Orange, 2,6-ANS). The dye is highly fluorescent in non-polar environment such as hydrophobic sites of unfolded protein that appear on the surface in gradual increase of temperature. Melting temperature (Tm) of a protein expresses its stability. Ligand screening relies upon the fact that protein stability is enhanced upon ligand binding (ΔTm  0). The aim of this study was to introduce the DSF method on our department, use it for characterization of carbonyl reductase 1 (CBR1) and evaluate the obtained results with an independent methods and literature. First, the functionality of method was verified with citrate synthase and the conditions were optimized for CBR1. The group of several buffers pH 3-10 without additives or with NaCl or glycerol were tested...

Treated Waste Water Irrigation
Kohut, Rafael ; Hyánková, Eva (referee) ; Kriška-Dunajský, Michal (advisor)
The thesis deals with the issue of simulation of treated wastewater irrigation by the HYDRUS 2D software. The first part of the thesis is designed as a theoretical research of academic information and knowledge. It aims at introducing the issue of treated wastewater utilization for the purpose of the irrigation. Furthermore it provides basic theoretical information on numeric simulation of the dissolved materials motion and transport in the soil. The last chapter of the theoretical part describes the calibration of the simulation by the HYDRUS 2D software. The second part of the thesis is devoted to the formation of filtration columns placed in the chosen site of the root zone wastewater plant. Within the filtration columns the automatic irrigation was formed and set off. Measuring of humidity in filtration columns was taking place for 81 days as well as the water sampling after the flow through the soil horizons. On the grounds of the measuring results the calibration of numeric simulation was carried out by the HYDRUS 2D software. The last part of the thesis presents the applications through the extending and alternation of the modelled setting. The main aim of the thesis is to calibrate the model and propose a general solution to advancement at realization of irrigation details when using the treated wastewater for the purpose of irrigation.

Study of Factors Influencing the Development of "Water Bloom" in the Water Reservoir Seč
Forczek, Sándor ; Holík, Josef ; Rederer, L. ; Koza, V.
In this work, we monitored several parameters that influence the development of algal bloom on the\nwater reservoir Sec. Primarily climatic conditions belong to the monitored parameters that influence\nthe population dynamics of algae (air temperature and rainfall, incident light), temperature\nstratification of water, concentration of oxygen, concentrations of total phosphorus, phosphate,\nvarious forms of nitrogen and organic substances, turbidity and also diversity and frequency of\nphytoplankton and zooplankton species. Between 2012 and 2014 have not occurred massive algal\nblooms in the summer months, however the measured physical, chemical and biological data, can be\nused to assess the development of algal population. The population dynamic factors influencing algal\ngrowth must be understand to be able to predict formation and to attempt suppression of algal\nblooms.
