Jindrich Fairaizl

Jindřich Fairaizl is a pioneer of Czech television documentary. He was born on June 14, 1934 in Prague. Beginning the age of 18, he worked in various media ? he started out in Československý rozhlas (Czechoslovak radio broadcast) and then he went over to Československá televize (Czechoslovak television), where he was active as a creator of many programs. His main contribution lies in television journalism which he co-founded and defined through his works. The main part of his work is from late 1960s, especially his series Inzerát (The Advertisement) and Starci (Old People). After 1970, he was not allowed to work in his field and had to accept routine jobs. For example, he was making instructional films about security of work. He became the first Managing Director of Česká televize after the Velvet revolution in 1989. Facing difficult conditions, due to psychological stress and a hospitalization, he had to step down from his position after a week. He died on October 10, 1993 in Měchenice after the house which contained his archive burned down. The conditions of the accident remain unclear.

The analysis of manager training and development

The bachelor thesis focuses on education and development of managers at AV Media. The aim of the work is to analyze the manager training system and development in the company by emphasizing the strengths and weaknesses of the system and proposing optimization methods. The theoretical part defines the basic concepts, such as training and development and the role of the manager in the company. Further the various stages of the training cycle, including identification of training needs, planning, implementation and evaluation are discussed. The practical part includes possible adjustments of AV MEDIA with an analysis of the manager education system, which is set in the company. The analysis is based on an interview with the HR manager of the company, internal documents and a questionnaire survey among the managers of the company. The last part includes a summary of the analysis and an evaluation system together with proposals for improvements following the latest trends and knowledge in the field.

Václav Voska's theatre, film and television acting in Czech artistic and social context in the second half of twentieth century

SUMMARY: This thesis focuses on theatre, film and television acting of one of the important representatives of Czech dramatic art in Czech artistic and social context in the second half of the twentieth century. The aim of this work is to depict means of expression used by Vaclav Voska in his roles. It provides summary of actor's theatrical work and in the part dedicated to his TV acting tries to comprehensively describe compering television series "We invite you to the theatre". The following part is dedicated to film acting, especially to his cooperation with the Film Academy of Performing Arts in Prague. The work neither neglects his teaching at the Faculty of Performing Arts Theatre nor his political career. The essential part of this thesis is focuses on his role of Emile Magis in Félicien Marceau's game The Egg. The work purposely omits on Vaclav Voska's production of spoken word (radio, Poetic Cafe Viola, Lyra Pragensis) and it refers only to his major achievements only if they are relevant for analyzed topics. Preparation of monoclonal antibody against the major structural protein, VP1, of BK virus and construction of plasmids for protein interaction analysis by BiFC method.

The aim of this study was to create a monoclonal antibody against VP1 protein BK virus for study of individual steps of its replication cycle. BK virus is human polyomavirus which causes serious disease - polyomavirus associated nephropathy in immunocompromised patients after renal transplantation or hemorrhagic cystitis after bone-marrow transplantation. Virions of BK virus have capsids with icosahedral symmetry consisting of 360 molecules of the major capsid protein VP1 and two minor structure proteins VP2 and VP3, which are not exposed on the capsid surface. VP1 is highly immunogenic what gives us possibility to produce antibodies capable of BK virus monitoring by capsid protein detection. Empty BK virus particles produced in insect cells from a recombinant baculovirus were used as antigen to immunize mice for preparing hybridomas. Cloning of hybridomas created by fusion of myeloma cells with B-lymphocytes from immunized mouse gave us series of primary hybridoma clones. One of them, 5/G10/A5/B2/8, was cloned to homogenity and monoclonal antibody against VP1 BK virus was purified from hybridomas medium by affinity chromatography. Metabolism of Saccharides - production of Didactic Materials for High School Education

My rigorous thesis focuses on creating educational materials for High School Education, namely materials for teaching about metabolism of saccharides. This thesis is based on a research of international multimedia textbooks and Internet links. Results of this thesis are two educational materials - an educational program and a worksheet. Both of these materials were created to make teaching of biochemistry more effective, respectively teaching of the chapter about metabolism of saccharides in High School Education. The Worksheet composes of 21 tasks, which are mainly used for practicing of the discussed curriculum. The Educational Program is of a multimedia character and is created in Adobe Flash 8. This Educational Program is composed of a total of 12 animations (Starting Animation, Digestion, Glycolysis, Gluconeogenesis, Lactate and Alanine Cycle, Degradation of Pyruvate, Citric Acid Cycle, Interesting Facts, Test 1, Test 2, Didactic Game "Pexeso" ("Memory Game"), and About the Program). To make the use of this material easier for teachers a manual was created. The Educational Program is a part of the rigorous thesis and it is placed on a separate CD-R media.

Magazines for women today: a shop-window of consumerism?

The graduation thesis Magazines for women today: a shop-window of consumerism? focuses on analysis of editorials in periodicals Blesk pro ženy and ELLE. I analyzed the year 2009 by using qualitative content analysis. In this thesis I am trying to find out how much these editorials are engaged in consumerism and how much they are appealing to the consumer life style. I was also interested in values connected with consumerism and in the ways the consumer life style is being tied in the editorials. By using semiotic theories, sociology and critical media studies literature I am decoding that the pleasurable images and editorials are primarily rooted in the economic system of consumer goods distribution. This thesis is devoted to anyone who is interested in women's magazines and their economic background.

The use of comet assay for the estimation of DNA damage during the anticancer chemotherapy.

We have monitored the DNA damage induced by tumor chemotherapy. The combination of platin derivate (cis-platine or carboplatin) and another chemotherapy agent (gemcitabin, paclitaxel or vinorelbin) was applied to patients with lung carcinoma. The whole chemotherapy includes 4 cycles. We have monitored the DNA damage in peripheral blood lymphocytes through the first and the third cycle of chemotherapy and 3 weeks after the last cycle of chemotherapy. For detection we have used comet assay which seems to be accurate and reliable method. Lymphocytes were isolated from whole blood through the medium LSM lymfocyte and then were processed by the alkaline version of the single cell gel electrophoresis. We have analysed single strand breaks (SSB) and interstrand cross links (ICL). For the measurement of ICL isolated lymphocytes were incubated in the presence of 200 µM of styrene oxide (SO) to degrade the DNA by inducing strand breaks. Incubation of peripheral blood lymphocytes with 200 µM SO leads to the formation of comets. If ICLs are present in the DNA, the alkaline unwinding of DNA is blocked and the % of DNA in tail is reduced. Comets were stained with ethidium bromide and then analyzed by the software LUCIA Comet Assay. The damage of DNA caused by SSB before the chemotherapy was relatively low and...

Práce se snaží vysvětlit monetární příčiny hospodářských cyklů a zkoumá media a mechanismy jejich šíření v ekonomice. Vychází při tom z rakouské teorie kapitálu. Metodou zkoumání jsou postupy logické dedukce. Výstupem je deskriptivní analýza obecných příčin hospodářských cyklů z hlediska obecné ekonomické teorie rakouské školy. V praktické části je proveden rozbor těch nejdůležitějších hospodářsko-politických opatření před a během Velké deprese. Na té byla teorie monetárních příčin hospodářského cyklu prakticky testována. Z této aplikace vyplývá, že Velká deprese měla tak dlouhé trvání právě kvůli nevhodným hospodářským politikám americké vlády a byla způsobena nevhodnou měnovou politikou Federálního rezervního systému.

Analysis of the Yeast Colony Structure: Peroxisomes and Autophagy

Analysis of yeast colony structure: peroxisomes and autophagy Abstract This study is a part of the project "Molecular biology and physiology of yeast communities". Yeast colonies grown on solid complex medium communicate by periodical release of volatile ammonia (PALKOVÁ et al. 1997), which is accompanied by periodical alkalization of colonies and growth substrate. Transition of colonies to the ammonia producing period of their life is accompanied by global changes of gene expression, as was shown using RNA microarrays (PALKOVÁ et al. 2002). Changes in many metabolic genes were observed, indicating activation of peroxisomal fatty acids β-oxidation, activation of glyoxylate cycle enzymes and inhibition of mitochondrial oxidative phosphorylation. These data suggest a significant switch of cellular metabolism during early alkali phase, based on peroxisomal induction and mitochondrial decline. This switch could be facilitated by autophagy, the process which can selectively or non-selectively remove unnecessary cellular components, providing the cell with energy and removing parts interfering with the new metabolism.
